Abstract
【解決手段】 分析対象物の存在を測定するための組成物であって、ジアゾニウム塩、及び、白血球、エステラーゼ又はプロテアーゼの存在下で加水分解されうるエステル、及びアルカリ土類金属の塩を含むことを特徴とする組成物、それを用いる試験具及び試験方法。

Description
Claims (20)
- 試料中の、白血球、エステラーゼ及びプロテアーゼよりなる群から選択される分析対象物の存在を測定するための組成物であって、ジアゾニウム塩、及び、白血球、エステラーゼ又はプロテアーゼの存在下で加水分解されうるエステル、及びアルカリ土類金属の塩を含むことを特徴とする組成物。
- アルカリ土類金属塩が、マグネシウム、カルシウム、又はバリウムの塩である、請求項1記載の組成物。
- アルカリ土類金属塩が、マグネシウムの塩である、請求項2記載の組成物。
- 塩が、MgSO4である、請求項3記載の組成物。
- 組成物の最適pHが、白血球、エステラーゼ又はプロテアーゼの存在下でジアゾニウムカップリングを促進し、その際バックグラウンドのジアゾニウムカップリング及び色の変化が、最小に抑えられている、請求項1記載の組成物。
- 最適pHが、約8.8〜9.0である、請求項5記載の組成物。
- 最適pHが、約8.95である、請求項6記載の組成物。
- 化合物B−OHのBが、ピロール、チオフェン、フラン、アゾレソルシノール、フェノール、ロイコインドフェノール、アゾ染料、5−(4−ヒドロキシ−3,5−ジメトキシフェニルメチレン)−2−チオキソチアゾリン−3−酢酸、2−置換−6−ヒドロキシ−ベンゾチアゾール誘導体、ω−ニトロスチリルエステル、レソルフィン、アクリジノン、メロシアニン、8−ヒドロキシ−2H−ジベンズ−(b,f)アゼピン−2−オン、ジベンゾアゼピノン、ジベンゾチアゼピノン、クマリンエステル、及びこれらの混合物よりなる群から選択される、請求項8記載のエステル。
- ジアゾニウム塩が、1−ジアゾ−2−ナフトール−4−スルホナート、1−ジアゾフェニル−3−カルボナート、4−ジアゾ−3−ヒドロキシ−1−ナフチルスルホナート、4−ジアゾ−3−ヒドロキシ−7−ニトロ−1−ナフチルスルホナート、4−ジアゾ−3−ヒドロキシ−1,7−ナフチルジスルホナート、2−メトキシ−4−(N−モルホリニル)ベンゼンジアゾニウムクロリド、4−ジアゾ−3−ヒドロキシ−7−ブロモ−1−ナフチルスルホナート、4−ジアゾ−3−ヒドロキシ−7−シアノ−1−ナフチルスルホナート、4−ジアゾ−3−ヒドロキシ−7−〔1−オキソプロピル〕−1−ナフチルスルホナート、及びこれらの混合物である、請求項1記載の組成物。
- 該組成物が、さらに促進剤を含む、請求項1記載の組成物。
- 該促進剤が、8〜15個の炭素原子を有するアルコールである、請求項13記載の組成物。
- 試料中の白血球、エステラーゼ又はプロテアーゼの存在又は濃度を測定するための試験装置であって、試薬組成物(該試薬組成物は、ジアゾニウム塩、及び、白血球、エステラーゼ又はプロテアーゼの存在下で加水分解されうるエステル、及びアルカリ土類金属の塩を含む)を均一に組み込んだ担体マトリックスを含むことを特徴とする試験装置。
- 該アルカリ土類金属塩が、バックグラウンドのジアゾニウムカップリング及び色の変化を低下させる、請求項15記載の試験装置。
- 該組成物が、さらに促進剤を含む、請求項15記載の試験装置。
- 該促進剤が、約8〜15個の炭素原子を有するアルコールである、請求項17記載の試験装置。
- 試料中の、白血球、エステラーゼ又はプロテアーゼよりなる群から選択される分析対象物の存在を測定するための方法であって、試料を、試薬組成物、又は、試薬組成物を均一に組み込んだ試験装置(ここで、該試薬組成物は、ジアゾニウム塩、及び、白血球、エステラーゼ又はプロテアーゼの存在下で加水分解されうるエステル、及びアルカリ土類金属の塩を含む)と接触させて、次いで試薬組成物の検出可能な応答を検査することを特徴とする方法。
- 該検出可能な応答が、色原体、蛍光、反射率変化、化学発光、比色、又は分光測光によるものである、請求項19記載の方法。
