Abstract
【解決手段】 本発明は、好ましくは骨粗鬆症の予防又は治療のための、日向夏みかんの処理物を含有する骨代謝改善剤、及び、この骨代謝改善剤を含有する、骨代謝改善のための食品に関する。

Description
(1)日向夏みかんの処理物を含有する骨代謝改善剤。
(2)骨粗鬆症の予防又は治療のための上記(1)に記載の骨代謝改善剤。
(3)上記(1)又は(2)に記載の骨代謝改善剤を含有する、骨代謝改善のための食品。
確実に骨粗鬆症を生じるモデル動物として卵巣摘出(OVX)ラットを用い、日向夏みかん摂取が大腿骨骨密度及び血液組成に及ぼす影響を評価した。
採取した大腿骨を1枚のX線フィルムに乗せ、軟X線写真を撮影した。X線写真を目視で確認する限り、日向夏みかん投与群の骨量は一見して明らかに対照群より多かった(図1)。図1中の数値はそれぞれ、撮影したX線フィルムを最大解像度でスキャナー処理し、画像解析ソフト(Photoshop ver.4.0)を用いて長方形で囲んだ部分(大腿骨遠位端と大体骨頭頸部)のdensityを測定した1例を示す。すなわち、測定範囲のdensityとそのばらつき(SD)を示す。densityの数値は骨密度を反映している。全症例の結果を表1に示す。表1中の数値はPhotoshop ver.4.0を用いて測定されたdensityの値(無単位)である。当然ながら、表1に示す全てのサンプルの解析は同一条件で行った。大腿骨の同一部分の骨密度を比較したところ、表1に示される通り、日向夏みかん投与群(OVX+日向夏)は対照群(OVX)と比較して有意に骨密度が高かった。
骨代謝に影響を与える因子の血中濃度を測定した。具体的には、エストラジオール、ラットオステオカルシン(以上、骨形成系の因子)、ラット酒石酸抵抗性アルカリフォスファターゼ(TRAP)、Type I コラーゲン(以上、骨吸収系の因子)及びヘパラン硫酸の血液中における濃度をそれぞれ測定した。
結果を表2に示す。
ヒト破骨細胞とヒト骨芽細胞の培養系に日向夏みかん試料を添加して各細胞の増殖性を検討した。
宮崎市内で市販されている日向夏みかんの果実から、表面の皮を除いた部分、すなわち、通常生食する皮の一部とさのう部分のホモジネートを調製し、このホモジネート(20g)に酢酸エチル(20ml)を添加して脂溶性成分を抽出した。水溶性部分(水層)と酢酸エチル部分(有機層)をそれぞれ、ロータリーエバポレーターを用いて乾固した。水層から得られた成分をリン酸緩衝液に1重量%となるように溶解させた(得られた溶液を「水溶性分画」と称する)。また有機層から得られた成分を最終濃度で0.1%になるようにDMSOに溶解させた後にリン酸緩衝液で溶解させた。こうして得られた試料を以下の実験に用いた。
ヒト破骨前駆細胞はCambrex社より購入し、同社のプロトコールにより培養した。すなわち凍結細胞を1ウェルあたり10000個になるように96ウェル培養プレートに撒き、各種添加剤(牛胎児血清、L-グルタミン、M-CSF、水溶性RANKリガンド、抗生剤)を添加したOPBM培養液(破骨前駆細胞基礎培地)で95%O2, 5% CO2、37℃で培養した。倒立顕微鏡でコンフルエント状態になり、多核細胞化(破骨前駆細胞が破骨細胞に分化した状態)したことが確認された時点で、日向夏みかんの水溶性分画を、相対的に1:10:50:100の割合で添加されるように各ウェルに添加した。すなわち、水溶性分画を培養液で10倍、100倍、1000倍希釈し、各々10倍希釈液10μl(100)または5μl(50)、100倍希釈液10μl(10)および1000倍希釈液10μl(1)各ウェルに添加した。添加48時間後に倒立顕微鏡で細胞数に変化のないことを確認し、TetraColor ONE(生化学工業株式会社製)を添加し1時間反応させ、450nmと650nmの吸光度の差から増殖性を確認した。また、細胞を撒かないウェルを1列作り、培養液を他のウェルと同様に処理し、同様にTetraColor Oneを添加したものを測定時の対照とした。本方法は、ウェルにほぼ同数の生細胞が増殖していれば、細胞自体の増殖性の指標となることが知られている。
結果を図2に示す。日向夏みかん試料は破骨細胞の増殖能を用量依存性に抑制した。
Cambrex社からヒト骨芽細胞を購入し、添付のプロトコールに従い処理した。すなわち凍結細胞を1ウェルあたり10000個になるように96ウェル培養プレートに撒き、添加剤(牛胎児血清、アスコルビン酸と抗生剤)を添加したOGM培養液(骨芽細胞用増殖培地)で95%O2, 5% CO2、37℃で培養した。倒立顕微鏡でコンフルエント状態になったことが確認された時点で、日向夏みかんの水溶性分画を、相対的に1:10:50:100の割合で添加されるように各ウェルに添加した。すなわち、水溶性分画を培養液で10倍、100倍、1000倍希釈し、各々10倍希釈液10μl(100)または5μl(50)、100倍希釈液10μl(10)および1000倍希釈液10μl(1)各ウェルに添加した。24時間後又は48時間後に倒立顕微鏡で細胞数に変化のないことを確認し、破骨細胞と同様な手法でTetraColor ONE(生化学工業株式会社製)を添加し1時間反応させ、増殖性を確認した。プレートの一列(合計8ウェル)に水溶性分画が同一量添加されており、ウェルごとに吸光度を測定し、8ウェル分の平均値と標準誤差を求めた。
骨芽細胞増殖能の実験で得られたヒト骨芽細胞の培養液(水溶性分画添加後24時間培養×2シリーズ、48時間培養×2シリーズ)を本実験に使用した。各培養シリーズにおいて水溶性分画添加量が同一のものがそれぞれ8ウェル(100μL/ウェル)存在していたため、その8ウェルを合わせ、得られた各懸濁液中のインターロイキン6濃度を測定した。インターロイキン6は骨芽細胞が分化・骨化する際に産生し、単核球を破骨細胞に分化させる役割を持つサイトカインである。
