Claims (13)
- 生体系の代謝適応能または好気的要求度を改変することができる薬剤をin vitroにおいて同定する方法であって、
a) 該生体系から得られた第1のミトコンドリア分子の合成速度または分解速度を測定する工程であって、該生体系は、第1のアイソトープ標識前駆体分子を、該第1のアイソトープ標識前駆体分子の標識が該第1のミトコンドリア分子に取り込まれるのに十分な時間にわたり投与されたものである、工程;
b) 該第1のミトコンドリア分子の合成速度または分解速度に基づいて、該生体系の第1レベルの代謝適応能または第1レベルの好気的要求度を評価する工程;
c) 該生体系から得られた第2のミトコンドリア分子の合成速度または分解速度を測定する工程であって、該生体系は、該第1のミトコンドリア分子を得た後、該薬剤と第2のアイソトープ標識前駆体分子を、該第2のアイソトープ標識前駆体分子の標識が該第2のミトコンドリア分子に取り込まれるのに十分な時間にわたり投与されたものである、工程;
d) 該第2のミトコンドリア分子の合成速度または分解速度に基づいて、該生体系の第2レベルの代謝適応能または第2レベルの好気的要求度を評価する工程;
e) 該第1レベルの代謝適応能または該第1レベルの好気的要求度と該第2レベルの代謝適応能または該第2レベルの好気的要求度との間のデルタ値を測定する工程;および
f) 該デルタ値に基づいて該生体系における代謝適応能または好気的要求度を改変することができる該薬剤の効力を同定する工程、
を含む、上記方法。 - 前記第1のミトコンドリア分子の合成速度または分解速度が、該第1のミトコンドリア分子のアイソトープ含有量、アイソトープパターン、アイソトープ含有量の変化速度またはアイソトープパターンの変化速度に基づき;そして
前記第2のミトコンドリア分子の合成速度または分解速度が、該第2のミトコンドリア分子のアイソトープ含有量、アイソトープパターン、アイソトープ含有量の変化速度またはアイソトープパターンの変化速度に基づく、請求項1記載の方法。 - 前記生体系が哺乳動物または哺乳動物の組織である、請求項1記載の方法。
- 前記生体系がヒトまたはヒトの組織である、請求項3記載の方法。
- 前記生体系が非ヒトまたは非ヒトの組織である、請求項3記載の方法。
- 前記生体系が細胞である、請求項1記載の方法。
- 前記細胞がハイスループットスクリーニング検定系における培養細胞である、請求項6記載の方法。
- 前記第1のミトコンドリア分子が、運動または他のトレーニング計画にさらに付された哺乳動物または哺乳動物の組織から得られ;そして
前記第2のミトコンドリア分子が、運動または他のトレーニング計画にさらに付された哺乳動物または哺乳動物の組織から得られる、請求項3記載の方法。 - 前記第1のミトコンドリア分子が、前記運動または他のトレーニング計画の後にデコンディショニング期間にさらに付された哺乳動物または哺乳動物の組織から得られ;そして
前記第2のミトコンドリア分子が、前記運動または他のトレーニング計画の後にデコンディショニング期間にさらに付された哺乳動物または哺乳動物の組織から得られる、請求項3記載の方法。 - 前記第1のアイソトープ標識前駆体分子および前記第2のアイソトープ標識前駆体分子がそれぞれ安定したアイソトープで標識されている、請求項1記載の方法。
- 前記第1のアイソトープ標識前駆体分子および前記第2のアイソトープ標識前駆体分子が、 2 H標識グルコース、 13 C標識グルコース、 2 H標識アミノ酸、 15 N標識アミノ酸、 13 C標識アミノ酸、 2 H標識酢酸、 13 C標識酢酸、 2 H標識リボヌクレオシド、 13 C標識リボヌクレオシド、 15 N標識リボヌクレオシド、 2 H標識デオキシリボヌクレオシド、 13 C標識デオキシリボヌクレオシド、 15 N標識デオキシリボヌクレオシド、 2 H標識脂肪酸、および 13 C標識脂肪酸からなる群から独立して選択される、請求項1記載の方法。
- 前記第1のアイソトープ標識前駆体分子および前記第2のアイソトープ標識前駆体分子が、それぞれ 2 H 2 O分子である、請求項1記載の方法。
- 前記第1のミトコンドリア分子および第2のミトコンドリア分子が、デオキシリボ核酸(DNA)、リボ核酸(RNA)、タンパク質、または脂質からなる群から独立して選択される、請求項1記載の方法。
