Abstract
Description
本明細書に記載の方法及び装置は、成分混合物の典型的なサンプルから、一以上の成分を分離するのに適する。サンプルは、何百又は何千の成分を含む複合混合物であってよい。成分は性質が均一であってよいが、むしろ均一でないのが好ましい。好ましい側面では、二以上の成分を一以上の性質、例えば荷電、質量、等によって識別してよい。
本発明者らの方法及び装置を使用して、任意のサンプル、特にタンパク質含有サンプルを分析してよい。好ましい実施形態では、本明細書に記載の方法及び装置は、タンパク質含有サンプルを分離するのに適する。等電点にしたがって集束及び/又は分析された分子にはタンパク質が含有されるのが好ましい。
好ましい実施形態では、検出又は分析されるタンパク質又はタンパク質群には疾患関連タンパク質が含まれる。この用語は、そのタンパク質が個体の細胞、組織又は器官中に存在することにより、その細胞、組織又は器官が疾患状態にあることを示すものを意味する。好ましい側面として、このタンパク質は病理学的症状のフラッグ又はマーカーである。このタンパク質は、疾患状態の原因物質であっても、あるいはいかなる原因作用も有さなくてもよい。このタンパク質は疾患の「下流の」指標であってもよい。疾患関連タンパク質により、個体に疾患又は疾患に対する感受性が存在することが示されてもよい。
このモジュールでは、通路、好ましくは狭い通路に沿った等電点電気泳動を利用する。
等電点電気泳動モジュールには、通路を有する(一般に平面形状の)基板が含まれる。本明細書に記載の等電点電気泳動モジュールはまた、「カートリッジ」と称されることもあり、等電点電気泳動の技術及びモジュールは「CIEF」(毛細管等電点電気泳動)と称されることもある。
通路は、一般に細長く、好ましくは直線状である。通路の構成は管状であってよいが、その全長の少なくとも一部に沿って開放されているのが好ましい。好ましくは、通路はその長さの実質的に全部に沿って開放され、その結果基板上で断面が溝状又は上が開いた通路の形状をしている。
等電点電気泳動モジュールには、通路をその長さの少なくとも一部に沿って露出するための手段が含まれる。このように通路を露出すると、通路内のサンプル又は成分(群)は露出され、好ましくはMALDI分析のためにアクセス可能になる。非常に好ましい実施形態では、通路は「開放」通路であり、この語は、通路の長さの少なくとも一部、好ましくは相当な部分が、閉鎖又は遮蔽されていないことを意味する。換言すれば、このような好ましい実施形態では、通路の形状は長い上側が開放している。この開放部分の幅は、すくなくとも通路の内容物、例えばサンプル、及び好ましくは分離され集束したサンプルの成分、例えばタンパク質にアクセスするのに必要な程度であるのが望ましい。 好ましくは、開放部分の長さは、集束した成分又はタンパク質の全部又は実質的な全部に及ぶ。
基板は、等電点電気泳動に適した任意の材料、例えば、プラスチック、ポリマー、セラミック、ガラス又は複合材料で形成させてよく、当分野では既知である。一般に、基板としての使用に適している非伝導性材料はどれでもよい。
担体両性電解質は、種々の酸性と塩基性の両方の緩衝基を組み込んだ合成ポリマーの不均質混合物である。両性電解質分子の正味の電荷は、環境のpH及び特定分子上に具体的に混在している酸性基と塩基性基の数及びpKに依存する。等電点電気泳動(IEF)では、担体両性電解質を通路内に導入する。電場を架けていなければ、担体両性電解質はランダムに分配されているので、ゲルマトリクスの至る所で均一なpH、すなわちpH3-10の勾配を形成する場合には約pH 7が確立している。

マトリクス支援レーザ脱離/イオン化-飛行時間(MALDI-TOF)質量分析は、高分子分析用、特にタンパク質用のツールである。MALDI-TOFは、タンパク質及び核酸の配列、構造、純度、不均一性、切断、翻訳後修飾、及び他の分子の特性であって、時折他の手段では解明が困難な特性を識別可能である。MALDIは、Chapman, J.R., Mass Spectrometry of Proteins and Peptides, 2001, Humana Press, Dass, C., Principles and practice of biological mass spectrometry, 2001, John Wiley & Sons, James, P., Proteome research: mass spectrometry, 2001, Springer, Kellner, R., F. 以後、添付の図面を参照して本発明の好ましい実施形態を説明するが、ここでは終始同じ数字は同じ要素を表す。本明細書に提示の説明中で使用する用語は、その合理的に最も広義に解釈されるものとする。この解釈は本発明のある具体的な好ましい実施形態の詳細な説明と組み合わせて用いる場合も同じである。このことは本明細書で使用するいくつかの特別用語に関して以下にさらに強調される。読者が何か限定的に解釈したいとする用語は、本明細書でそのように明確にかつ具体的に規定する。
ポリ(メチルメタクリレート)(PMMA)は地元の供給元(Swees Engineering Co. (PTE) Ltd.)から購入する。ミオグロビン、グリセロール、ファーマライト(pharmalyte)、メチルセルロースはSigma Chemicalsから注文する。すべての他の化学物質はAldrich Chemicalsから入手する。すべての溶液はNanopure水システムによって精製された水を用いて調製する。白金線はFine Metal Cropによって供給される。
PMMAプレート片、横90 mm、縦30 mm及び厚さ3 mmを原料のプラスチックプレートからカットする。直径0.005インチ及び0. 007インチの白金線を用いて、プラスチック基板に通路を刻む。末端をワイヤの張力弓にクランプすることにより長さ約150 mmの白金線を伸ばしてぴんと張る。PMMA及びガラスプレートでこの白金線をサンドイッチし、これを二つのアルミニウムブロックの間に共にクランプする。クランプをきつく締めてアルミニウムブロックに圧力を架けながら、白金線に電流を通してこれを赤熱する。組立物を完全に冷却したら、クランプを解放し、白金線をプラスチックから引き出すと通路が現れる。
ミオグロビンを本方法の開発用モデルタンパク質として用いるが、これはミオグロビンが褐色系の色を有し、ヒトの眼で検出可能であるからである。0.02μg/μlミオグロビン、1% Pharmalyte 3-10の濃度でサンプルを調製する。マイクロピペットにより約5μlのサンプルを開放微小通路に投入する。サンプルは毛細管作用により微小通路内に一様に拡がる。
MALDI-MS実験は、線形モードで作動するBruker Daltonics Autoflex MALDI飛行時間質量分析計(TOF-MS)で行う。
以下のプロトコルを用いてブタ肝臓からタンパク質を抽出する。ブタ肝臓を2倍容量の脱イオン水と混合し、ブレンダーを用いてホモジナイズする。超音波処理器を用いて細胞を溶解する。この混合物を遠心分離し、上清を脱イオン水で二回希釈し、さらに精製せずに開放通路CIEF用に用いる。開放通路CIEFは基本的には実施例3に記載のように実行し、MALDI分離は基本的に実施例4に記載のように実行してよい。
図4Aは本発明の一実施形態の上面図を示す。多数の開放微小毛細管通路(2)は基板(1)上にエッチング又は構築される。二つの電解質液槽(3及び4)はサンプル基板上にエッチング又は構築される。一方の液槽は微小毛細管通路の一端に接し、他方は微小毛細管通路の他端に接する。開放毛細管通路の二つの端はそれぞれ二つの液槽に、それらの境界にある狭い通路、半透膜又はゲルを介して連結されているが、サンプルと陽極液又は陰極液の混合は防止されている。液槽は陽極及び陰極の溶液用である。基板は任意の非伝導性材料、例えばポリマー、セラミック、ガラス、又は複合材料が可能である。図4Bは複数の開放微小毛細管通路(2)の横断面を示す。通路の横断面は長方形に限定されず、任意の形状が可能である。種々のタンパク質サンプルのそれぞれが固有の通路内に載置される。電位を陽極液と陰極液の間にかけると、タンパク質はこの電場の下で自身のpI点へ移動する。分離されたタンパク質は、開放通路の上でMALDIレーザを用いて直接イオン化できるので、このタンパク質を移動することはない。分離されたタンパク質を移動する代わりに、CIEFカートリッジ全体を並進台に載せ、この並進台の移動によりタンパク質をMALDIレーザスポットに持ってくる。結果として、CIEFを用いて達成可能な高い分解能は、検出プロセスの中にそのまま持ち越される。さらに、多数の開放毛細管通路はCIEFカートリッジ上で構築可能であり、その通路数は一定ではなく、カートリッジが保持可能な限り多数である。したがって高処理化に適用するために、多数のサンプルを平行に分離することができる。
Claims (44)
- 等電点電気泳動(IEF)モジュールであって:
(a)サンプルを載置し、サンプル中の成分又は成分群を等電点にしたがって集束させる通路を有する第一の平面部材と;
(b)通路をその長さの少なくとも一部に沿って露出し、これにより通路内のサンプル又は成分(群)を露出する手段と、
を備えるモジュール。 - サンプル又は成分(群)を、露出した通路に沿って実質的にそれらが集束した位置でアクセス可能である、請求項1に記載のモジュール。
- 通路が、その長さの少なくとも一部に沿って露出している開放通路を備える、請求項1又は2に記載のモジュール。
- 通路が、第一の平面部材の表面上に形成された直線状の溝を備える、請求項1、2又は3に記載のモジュール。
- 通路が微小通路又は毛細管通路を備える、請求項1〜4のいずれか1項に記載のモジュール。
- 微小通路又は毛細管通路が第一の平面部材上に微細加工されている、請求項5に記載のモジュール。
- 通路が、1〜500マイクロメートルの範囲、より好ましくは50〜350マイクロメートルの範囲、より好ましくは50〜350マイクロメートルの範囲、最も好ましくは約150マイクロメートル又は約175マイクロメートルの幅を有する、請求項1〜6のいずれか1項に記載のモジュール。
- 第一の平面部材が、プラスチック、ポリマー、セラミック、ガラス又は複合材からなる群から選択される材料から形成されている、請求項1〜7のいずれか1項に記載のモジュール。
- 通路の少なくとも一の壁がポリ(メチルメタクリレート)(PMMA)又はポリカーボネートを備える、請求項1〜8のいずれか1項に記載のモジュール。
- 第一の平面部材を被覆又は誘導体化して表面荷電を減少し、これにより電気浸透流(EOF)を最小化する、請求項1〜8のいずれか1項に記載のモジュール。
- 電解質用の液槽を備え、この液槽が通路と電気的に連結している、請求項1〜10のいずれか1項に記載のモジュール。
- 液槽が、第一の平面部材上に通路の各端に隣接して形成される、請求項11に記載のモジュール。
- 第二の平面部材である蓋をさらに備え、これにより通路内のサンプルの蒸発が減少するようになっている、請求項1〜12のいずれか1項に記載のモジュール。
- 蓋がその内側表面に細長い凹部を備え、この凹部は、蓋を第一の平面部材と合わせたときに通路に含まれるサンプルを実質的に漏出させない位置に配置されている、請求項13に記載のモジュール。
- 凹部の長さ及び幅が、第一の平面部材の通路と少なくとも同じ大きさである、請求項14に記載のモジュール。
- 液槽が蓋の上に配置されている、請求項11又は請求項13〜15のいずれか1項に記載のモジュール。
- 通路と液槽との間に電気的連結のための手段を備え、それによってサンプルと電解質の実質的な混合が防止されるようになっている、請求項11〜16のいずれか1項に記載のモジュール。
- 前記手段が半透膜、アガロース、又はゲル栓を備え、請求項17に記載のモジュール。
- 実質的に平行に配向している複数の通路を備える、請求項1〜18のいずれか1項に記載のモジュール。
- 通路又は通路群が閉鎖通路(閉鎖通路群)を備え、この閉鎖通路(閉鎖通路群)を露出する手段として、この閉鎖通路(閉鎖通路群)の長軸方向の平面に沿って開裂可能な脆いラインが含まれる、請求項1〜19のいずれかに記載のモジュール。
- 並進台をさらに備え、その上に第一の平面部材が取り付けられる、請求項1〜20のいずれか1項に記載のモジュール。
- サンプル中の一以上の成分を分離する装置であって、請求項1〜21のいずれか1項により特許請求される等電点電気泳動(IEF)モジュール並びに等電点に基づいて集束した成分をそれらの各質量に応じて分離することができるモジュールとともに備える装置。
- 質量分離モジュールが質量分析用のモジュールを備える、請求項22に記載の装置。
- 質量分離モジュールがマトリクス支援レーザ脱離/イオン化質量分析(MALDI-MS)モジュール、好ましくはマトリクス支援レーザ脱離/イオン化-飛行時間(MALDI-TOF)モジュールを備える、請求項22又は23に記載の装置。
- サンプル中の一以上の成分を分離する方法であって:
(a)通路を有する第一の平面部材を含む等電点電気泳動(IEF)モジュールを準備するステップと;
(b)通路にサンプルを載置するステップと;
(c)サンプルの成分又は成分群を、通路に沿って、等電点にしたがって集束するステップと;
(d)通路の長さの少なくとも一部を露出し、これにより通路内のサンプル又は成分(成分群)を露出するステップと;
(e)任意に、等電点電気泳動(IEF)モジュールを電気伝導性の薄膜又は層で被覆するステップと;
(f)任意に、一以上の分離された成分を質量分析、好ましくはMALDI、より好ましくはMALDI-TOFによって分析するステップと;
を含む方法。 - 請求項2〜21のいずれか1項に規定する一以上の特徴をさらに含む、請求項25に記載の方法。
- (e)開放通路内の一以上の成分にアクセスしこれを分析する操作をさらに含む、請求項25又は26に記載の方法。
- 該又は各成分を、質量分析、好ましくはMALDI-MS、より好ましくはMALDI-TOF質量分析によって分析する、請求項27に記載の方法。
- サンプルがMALDIマトリクスを含む、請求項28に記載の方法。
- MALDIマトリクスを等電点電気泳動後のサンプルに添加する、請求項28に記載の方法。
- MALDIマトリクスが、シアノ-4-ヒドロキシ桂皮酸(CHCA)、2,5-ジヒドロキシ安息香酸(DHB)、アルファCCA、シナピン酸(SA)、3-ヒドロキシピコリン酸(HPA)、IAA(Na+)、2-(4-ヒドロキシフェニルアゾ)安息香酸HABA(Na+)、ジスラノール(Na+)、レチノイン酸(Na+)、コハク酸、2,6-ジヒドロキシアセトフェノン、フェルラ酸、コーヒー酸、グリセロール及び4-ニトロアニリンからなる群から選択される、請求項29又は30に記載の方法。
- 分子を分析する方法であって:
(a)細長い開放通路を準備するステップと;
(b)複数の分子を細長い開放通路内に導入するステップと;
(c)細長い開放通路に沿って等電点にしたがって分子を分離するステップと;
(d)細長い開放通路内の分子にアクセスしそれを分析するステップと;
を含む方法。 - 分子を等電点電気泳動(IEF)する装置であって、細長い通路を備え、この細長い通路はその少なくとも一部に沿って開放されており、それにより分離した分子にアクセスすることができる装置。
- CIEF-MALDI装置。
- 請求項1〜24、33又は34のいずれか1項で特許請求されているモジュール又は装置を、分析対象のタンパク質を含むサンプルとともに備えるキット。
- サンプル中のタンパク質を検出する方法であって:
(a)細長い開放通路を準備するステップと;
(b)サンプルを細長い開放通路内に導入するステップと;
(c)細長い開放通路に沿って、その等電点にしたがってタンパク質を分離するステップと;
(d)細長い開放通路中でタンパク質にアクセスするステップと;
(e)質量分析によってタンパク質を検出するステップと;
を含む方法。 - タンパク質分析のための、質量分析計、好ましくはMALDI-MSユニット、より好ましくはMALDI-TOFユニットと組み合わせた、請求項1〜24のいずれか1項に記載の等電点電気泳動モジュールの使用。
- プロテオーム分析のための、請求項37に記載の使用。
- サンプル中の疾患関連タンパク質を検出するための、請求項1〜24のいずれか1項に記載の等電点電気泳動モジュールの使用。
- 疾患関連タンパク質を、質量分析計、好ましくはMALDI-MSユニット、より好ましくはMALDI-TOFユニットと組み合わせたモジュールによって検出する、請求項39に記載の使用。
- 個体における疾患の診断方法であって:(a)個体由来の細胞、組織又は器官のサンプルを準備し、それのタンパク質含有抽出物を作成するステップと;(b)請求項1〜24のいずれか1項に記載のモジュール、請求項22、23又は24に記載の装置、又は請求項25〜32のいずれか1項に記載の方法を用いて、そのサンプル中の疾患関連タンパク質を検出するステップと;を含む方法。
- 毛細管等電点電気泳動(CIEF)装置とMALDI-MS装置、好ましくはMALDI-TOF装置とを連結する手段であって、等電点電気泳動を実行する通路、並びに該通路をその長さの少なくとも一部に沿って露出して通路内のサンプル又は成分(群)を露出する手段、を含む連結手段。
- 開放通路を含む、請求項42に記載の連結手段。
- 添付図面の図1〜図12を参照して前述しかつ図示したものと実質的に同じモジュール、装置、方法、キット、使用又は連結手段。
