Abstract
Description
本発明は、ヒトキナーゼ挿入ドメイン含有受容体(KDR)の抗原決定基に対する特異性を有する抗体分子に関する。この抗体分子は、ヒト血管内皮増殖因子(VEGF)より大きな親和力でKDRに結合し、VEGFとKDRとの相互作用を妨害する。本発明はまた、抗体分子の治療的使用と抗体分子の産生方法に関する。
軽鎖可変ドメインのCDRは、カバト(Kabat)の番号付けによると、残基24〜34(CDRL1)、残基50〜56(CDRL2)および残基89〜97(CDRL3)に位置する。

mRNAの選択的スプライシングから生じる、VEGFの5つの異なるモノマーアイソフォームが存在する。このアイソフォームには、2つの膜結合型(VEGF206 とVEGF189)および3つの可溶性型(VEGF165、VEGF121、およびVEGF145)がある。ヒト胎盤を除くすべての組織で、VEGF165 が最も豊富に存在するアイソフォームである。
(1つのVEGFR型のみを発現する細胞株で)KDR単独のVEGF媒介活性化が、細胞増殖と移動を引き起こすのに充分であることが証明された(ワルテンバーガー(Waltenburger)ら、J.Biol.Chem.,269,26988,1994)。逆にFLT−1単独を活性化すると、細胞増殖は見られず、相反して細胞移動が観察された。
KDRとの相互作用をブロックするがFLT−1との相互作用はブロックしない抗VEGF Mabは、VEGF誘導性の血管透過性を阻害できたが、非ブロック性抗VEGF抗体は効果がなかった(ブレッケン(Brekken)ら、Cancer Res.,60,5117,2000)。
現在、KDRはVEGFの作用を媒介する最も重要な受容体であると考えられており、血管形成の促進と新血管の生存におけるその役割が一般的に認識されているようである。
本発明の第1の態様の抗体分子は、重鎖可変ドメインにH1、H2およびH3(配列番号1〜配列番号3)から選択される少なくとも1つのCDRを含有する。好ましくはこの抗体分子は、重鎖可変ドメインに少なくとも2つの、さらに好ましくは3つすべてのCDRを含有する。
本発明の第2の態様の抗体分子は、軽鎖可変ドメインにL1、L2およびL3(配列番号4〜配列番号6)から選択される少なくとも1つのCDRを含有する。好ましくはこの抗体分子は、軽鎖可変ドメインに少なくとも2つの、さらに好ましくは3つすべてのCDRを含有する。
好ましくは本発明の第1の態様と第2の態様の抗体分子は、可変ドメインが、CDRH1として図1のH1に示す配列(配列番号1)、CDRH2として図1のH2に示す配列(配列番号2)、またはCDRH3として図1のH3に示す配列(配列番号3)を有するCDR(カバト(Kabat)ら(前述)により規定されたもの)を含有する重鎖と、可変ドメインが、CDRL1として図1のL1に示す配列(配列番号4)、CDRL2として図1のL2に示す配列(配列番号5)、またはCDRL3として図1のL3に示す配列(配列番号6)を有するCDR(カバト(Kabat)ら(前述)により規定されたもの)を含有する軽鎖とを、含む。
そのようなCDR移植抗体は、好ましくは上記のヒトアクセプターフレームワーク領域ならびに1つ以上のドナーCDRを有する可変ドメインを含有する。
本発明のCDR移植抗体において、アクセプター抗体として、ドナー抗体鎖に相同的な鎖を有するものを使用することが好ましい。アクセプター重鎖および軽鎖は、必ずしも同じ抗体から得られる必要は無く、所望であれば、異なる鎖から得られるフレームワークを有する複合鎖を含むものでもよい。
本発明のCDR移植抗体分子において、アクセプター軽鎖がヒト第1群フレームワーク領域(図3に示す)(配列番号10)を有する場合には、好ましくは軽鎖のアクセプターフレームワーク領域は、36、44、60、66、69、70および71位(カバト(Kabat)ら(前述)に従う)にドナー残基を有する。
ドナー残基は、ドナー抗体、すなわち元々CDRを得た抗体に由来する残基である。
ポリマー分子は一般に、合成のまたは天然に存在するポリマー、例えば任意に置換された直鎖または分岐鎖ポリアルキレン、ポリアルケニレンもしくはポリオキシアルキレンポリマー、または分岐もしくは非分岐ポリサッカライド、例えばホモもしくはヘテロポリサッカライドである。
特に好適なポリマーには、ポリアルキレンポリマー、例えばポリ(エチレングリコール)、または特にメトキシポリ(エチレングリコール)またはこれらの誘導体、特に約25000Da〜約40000Daの範囲の分子量を有するものである。
必要に応じて、抗体断片は、それに1つ以上の他のエフェクターまたはレポーター分子が結合しているものでもよい。エフェクターまたはレポーター分子は、断片中に存在する任意の利用可能なアミノ酸側鎖または末端アミノ酸官能基(例えば、任意の遊離のアミノ、イミノ、ヒドロキシル、またはカルボキシル基)を介して、抗体断片に結合することができる。
本発明のDNA配列は、合成DNA(例えば化学工程により産生される)、cDNA、ゲノムDNA、またはこれらの任意の組合せを含んでよい。
本発明はまた、本発明の1つ以上のDNA配列を含むクローニングベクターまたは発現ベクターに関する。好ましくはクローニングベクターまたは発現ベクターは、それぞれ本発明の抗体分子の軽鎖と重鎖とをコードする2つのDNA配列を含む。
好適な実施態様において本発明は、本発明のDNA配列を含む大腸菌(E.coli)発現ベクターを提供する。好ましくはこの発現ベクターは、図8に概略的に図示したpTTOD(CDP791)である。
アクセプターフレームワーク配列をコードするDNAは、当業者が広く利用することができ、その公知のアミノ酸配列に基づき容易に合成することができる。
本発明はまた、本発明の抗体分子をコードするDNAからタンパク質を発現させるのに適した条件下で、本発明のベクターを含む宿主細胞を培養し、抗体分子を単離することを含む、本発明の抗体分子の産生法を提供する。
本発明はまた、本発明の抗体分子を、薬剤学的に許容される賦形剤、希釈剤または担体とともに含む、治療用または診断用組成物の調製法を提供する。
抗体分子は、治療用または診断用組成物の唯一の活性成分であってもよく、他の抗体成分(例えば抗T細胞、抗IFNγ、または抗LPS抗体)、または非抗体成分(例えば、キサンチン)を含む他の活性成分と共に加えられるものでもよい。
本発明の抗体分子が投与される用量は、治療すべき症状の性質、中和すべきVEGFのレベル、または所望のレベルより高くなる事が予測される程度に、および抗体分子が予防的に使用されるのか、または既存の症状を治療するために使用されるのかに依存する。用量はまた、患者の年齢と症状に従って選択される。
抗体分子の半減期が短い(例えば、2〜10時間)時、1日に1回以上投与することが必要かも知れない。あるいは抗体分子の半減期が長い(例えば2〜15日)なら、1日、1週間、または1もしくは2ヶ月に1回の投与でいいかも知れない。
薬剤学的に許容される塩、例えば塩酸塩、臭化水素酸塩、リン酸塩、および硫酸塩のような鉱酸の塩、または酢酸塩、プロピオン酸塩、マロン酸塩および安息香酸塩のような有機酸の塩が使用できる。
いったん調製されると、本発明の組成物は直接被験体に投与される。治療される被験体は動物でもよい。しかし、組成物はヒト被験体への投与に適合することが好ましい。
組成物の直接送達は一般に、注入、皮下、腹腔内、静脈内または筋肉内で、または組織の間質スペースに投与されるであろう。組成物はまた病変中に投与することができる。投与処理は単回投与または複数回投与スケジュールでもよい。
薬剤学的に許容される担体の詳細な考察は、レミントンの薬剤科学(Remington’s Pharmaceutical Sciences)(マックパブリッシング社(Mack Publishing Company)、ニュージャージー州、1991)に記載されている。
本発明はさらに、VEGFおよび/またはKDRが関与している治療のための薬剤を製造するための、本発明の抗体分子の使用を提供する。
本発明の抗体分子は、ヒトまたは動物の体に存在する生物学的に活性なKDRのレベルを低下させることが望まれる治療法で使用される。VEGFは、体内を循環していたり体の特定の部位で望ましくない高いレベルで存在することがある。
本発明はまた、本発明の抗体分子の有効量を被験体に投与することを含む、VEGFおよび/またはKDRが関与している疾患にかかっているかまたはそのリスクのあるヒトまたは動物被験体の治療法を提供する。
本発明の抗体分子はまた、診断、例えば高レベルのKDRを伴なう病状のインビボ診断およびイメージングのために使用することができる。
本発明はさらに、添付の図面を参照する以下の実施例で例示のためにのみ記載される。
インハウス免疫プログラムを開始して、そのリガンドVEGFとの相互作用をブロックするのに有効なヒトKDRに対する抗体を選択した。完全長ヒトKDR、精製ヒトKDR−ヒトFc融合タンパク質、およびこれらの融合タンパク質をコードするDNAでトランスフェクトしたCHO細胞を含む種々の免疫原を用いて、マウスを免疫した。細胞/タンパク質免疫原で免疫した動物からの全部で19の融合体と、DNAで免疫した動物からの4つの融合体から、約23、000個のウェルを、ヒト7−ドメインKDR−Fcへの結合について1次ELISAフォーマットでスクリーニングした。次に約800の抗体を、ヒト7−ドメインKDR−Fcへの125−I VEGF結合のブロッキングを測定する放射免疫定量法の2次スクリーニングにかけた。3次スクリーニングにより、ヒト臍帯静脈内皮細胞(HUVEC)からのVEGF刺激組織因子放出のブロッキングを測定した。このスクリーニングカスケードから、抗体VR165が選択された(データは示していない)。
VR165を発現するハイブリドーマ細胞からRNAを調製し、DNAに逆転写した。次にこれを一連のPCR反応の鋳型として使用して、V領域配列を増幅した。保存された重鎖および軽鎖シグナル配列内でアニーリングするように設計された縮重プライマープールをフォーワードプライマーとして使用し、フレームワーク4/C領域ジャンクションをコードするプライマーをリバースプライマーとした。こうして、重鎖と軽鎖の両方のV領域遺伝子を増幅し、次にクローン化し配列決定した。DNA配列を翻訳してVR165 V領域アミノ酸配列を得て、これは、N末端配列決定により決定されたタンパク質配列と比較して確認した。次にマウスV領域遺伝子を、発現ベクターpMR10.1とpGamma−4中にサブクローン化した。これらは、ヒトカッパ軽鎖とgamma−4重鎖の定常領域遺伝子をコードするゲノムDNAを含有するそれぞれ軽鎖と重鎖の発現のための別々のベクターである。CHO細胞への同時トランスフェクションにより、キメラVR165抗体が得られる。
免疫原性を低下させ大腸菌(E.coli)発現を促進するために、VR165をヒトフレームワークにCDR移植した。サブグループVHIIIとVLIから、ヒト生殖細胞系アクセプターフレームワークを選択した。重鎖アクセプターフレームワークは、ヒト生殖細胞系配列VH3−7であり、ヒトJH領域生殖細胞系JH4のこの部分から来るフレームワーク4を有する。軽鎖アクセプターフレームワークは、ヒト生殖細胞系配列A30であり、ヒトJK領域生殖細胞系JK1のこの部分から来るフレームワーク4を有する。整列すると、ドナーとアクセプター重鎖の間に15のフレームワークの差があることを示す。これらの位置のそれぞれで、抗原結合に寄与する残基の可能性について分析した;重要であると考えられたら、マウスドナー残基を保持した。軽鎖整列は、ドナーとアクセプター重鎖の間に24のフレームワークの差があることを示す。マウス残基が抗原結合に寄与する可能性を、再度分析した。こうして、VH移植片を設計し、3つのVL移植片を得た(図9、配列番号17〜22)。各場合に、移植片1は、マウスフレームワーク残基の無い移植片である。移植片gH3はまた、CDR−H2のC末端に追加のヒト残基を含有する。CDRのこの部分は、抗原結合表面ではない。大腸菌(E.coli)で頻繁に使用されるコドンを使用し、「まれ」な大腸菌(E.coli)コドンを避けて、移植配列をコードするように遺伝子を設計した(ワダ(Wada)ら、Nucleic Acids Res.19、1981−86、1991)。さらなる遺伝子操作を促進するために、間隔を置いてDNA配列中に制限部位を導入した。図10は、gH1とgL1の遺伝子の設計を示す(配列番号23と配列番号24)。遺伝子を構築するのに使用したオリゴヌクレオチドを図11に示す(配列番号25〜40)。
PCR組み立て法を使用して、CDR移植gH1とgL1 V領域遺伝子を構築した。10mMトリス塩酸(pH8.3)、1.5mM MgCl2、50mM KCl、0.001%ゼラチン、0.25mMの各デオキシリボヌクレオシド三リン酸、1pmolの各「内部」プライマー(F2、F3、F4、R2、R3、R4)、10pmolの各「外部」プライマー(F1、R1)、および1単位のTaqポリメラーゼ(アンプリタク(AmpliTaq)、アプライド・バイオシステムズ(Applied BioSystems)、カタログ番号N808−0171)を含有する100μlの反応容量を準備した。PCRサイクルパラメータは、94℃で1分、55℃で1分、および72℃で1分を30サイクルであった。次に反応生成物を1.5%のアガロースゲルで流し、切り出し、キアゲン(Quiagen)スピンカラム(キアクイック(QIAquick)ゲル抽出キット、カタログ番号28706)を使用して回収した。30μlの容量でDNAを溶出した。次にアリコート(1μl)のgH1とgL1 DNAを、インビトロゲン(InVitrogen)トポタ(TOPO TA)クローニングベクターpCR2.1 TOPO(カタログ番号K4500−01)中に製造業者の説明書に従ってクローン化した。この非発現ベクターは、多量のクローンの配列決定を促進するためのクローニング中間体とした。ベクター特異的プライマーを使用するDNA配列決定を使用して、gH1とgL1を含有する正しいクローンを同定して、プラスミドpCR2.1(gH1)、およびpCR2.1(gL1)を作成した(図12を参照)。
結合を、KDR結合ELISA、KDRへの標識VEGF結合の阻害の放射免疫定量法、およびKDR結合のビアコア(BIAcore)アッセイにより測定した。移植したすべての型は、ELISAと放射免疫定量法で良好に機能し、キメラに似た活性を示した。ビアコア(BIAcore)分析から、移植片gL3gH3を最適として選択(データは示していない)し、これを以後g165と呼ぶ。
プラスミドpTTO−1を以下のように構築した。
プラスミドpTTQ9をアマシャム(Amersham)から得た。アリコート(2μg)を制限酵素SalIとEcoRIで消化し、消化物を1%アガロースゲルに流し、大きいDNA断片(4520bp)を精製した。2つのオリゴヌクレオチドを合成し、これらはアニーリングすると、OmpAポリリンカー領域をコードする。この配列は、pTTQ9の制限切断により作成されたSalIとEcoRI末端に適合する粘着末端を有する。このオリゴヌクレオチド「カセット」をpTTQ9ベクター中にクローニングすると、SalI部位は再生されないが、EcoRI部位は維持される。このカセットは、大腸菌(E.coli)外膜タンパク質Omp−Aのシグナル配列の最初の13アミノ酸をコードし、これは、OmpA遺伝子のシャインダルガルノ(Shine Dalgarno)リボゾーム結合部位が先行する。さらに、酵素XbaI、MunI、StyIおよびSplIの制限部位が存在する。MunIとStyI部位は、OmpAシグナル配列のコード領域内に有り、遺伝子の挿入のための5’クローニング部位となる。このカセットを構成する2つのオリゴヌクレオチドを、5pmol/μlの濃度で混合し、水浴中で95℃で3分加熱して、次に室温までゆっくり冷却することにより、アニーリングした。次にアニーリングした配列を、SalI/EcoRI切断したpTTQ9に連結した。得られたプラスミド中間体(pTQOmpと呼ぶ)をDNA配列決定により証明した。
プラスミドpACYC184からの1つのDNA断片を、pTQOmpから作成した2つの断片に連結して、プラスミドpTTO−1を構築した。プラスミドpACYC184はニューイングランドバイオラボズ(New England Biolabs)から得られた。アリコート(2μg)を制限酵素StyIで完全に消化し、次に大豆ヌクレアーゼ(Mung Bean Nuclease)を用いて処理した;この処理により、5’塩基オーバーハングを切断して、平滑末端を作成する。フェノール抽出とエタノール沈殿後、DNAを酵素PvuIIで切断し、2348、1081、412および403bpの断片を生成した。アガロースゲル電気泳動後に2348bp断片を精製した。この断片は、テトラサイクリン耐性マーカーとp15A複製開始点をコードする。次に断片を子ウシ小腸アルカリホスファターゼで処理して、5’末端リン酸を除去し、こうしてこの分子の自己連結を防止した。
次に、ほぼ等モル量の各断片を使用して3つの断片を連結して、プラスミドpTTO−1を作成した。すべてのクローニングジャンクションをDNA配列決定により証明した。
pTTO−1から、PvuII(3部位)、EcoRV(2部位)およびApaI(1部位)の骨格制限部位を除去して、プラスミドpTTODを得た。Fab’コード法を単純化するために、これらの変化を加えた。これらの変化を加える時に、lacIq遺伝子とテトラサイクリン耐性遺伝子のコードタンパク質配列は変化させなかったが、DNAレベルの「サイレント」変化を行った。PCR法を行い、ここでこれらの制限部位を除去した「サイレント」変化を有するプライマーを設計し、これを使用して親プラスミド(pTTO−1)のセクションを増幅した。次にフランキング制限部位(無変化)を使用して、親プラスミド中の配列をこれらの修飾配列で置換した。この多工程プロセスにより、プラスミドpTTODが作成された。遺伝子にフランクするユニークなPstIとEcoRI部位を使用して、ベクターpTTO内の既存のFab’遺伝子のpTTODへの移行を行い、pTTOD(Fab’)を作成した。
g165配列の挿入のための開始点は、無関係のFab’の発現のための3つのベクター(pTTOD(Fab’IGS−1)、pTTOD(Fab’IGS−2)およびpTTOD(Fab’IGS−3))である(例えば、図16を参照)。これらは、軽鎖遺伝子を重鎖遺伝子から分離する、いわゆるIGSすなわち遺伝子内配列でのみ異なる。これらのIGS領域を図17に示す(配列番号53〜55)。これらのベクターへのg165配列のクローニングは、2工程プロセスとして行った。まず、pCR2.1(gL3)から軽鎖をEcoRV−BsiWI断片(395bp)として制限切断し、pTTOD(Fab’IGS−1)、pTTOD(Fab’IGS−2)およびpTTOD(Fab’IGS−3)のEcoRV−BsiWI消化からの大きいベクター断片中に挿入した。これで、クローニング中間体pTTOD(g165L IGS−1)、pTTOD(g165L IGS−2)およびpTTOD(g165L IGS−3)が作成された。次にこれらのクローニング中間体をPvuIIとApaIで切断し、大きいベクター断片を精製し、pCR2.1(gH3)からの435bpのPvuII−ApaI断片を挿入した。これで、3つのFab’発現プラスミドpTTOD(g165 IGS−1)、pTTOD(g165 IGS−2)およびpTTOD(g165 IGS−3)が作成された。
プラスミドpTTOD(g165 IGS−2)をpTTOD(CDP791)と再度名付けた。この構築体のプラスミド地図を図8に示す。図19は、このベクター中のFab’のコード領域の完全なDNAとタンパク質配列、および5’と3’フランキング配列の一部を示す(配列番号56)。
精製し修飾Fabは、mPEGの分岐分子に部位特異的に結合した。これは、修飾Fabの端を切り取ったヒンジ領域中の単一のシステイン残基の活性化と、次に、既に記載されているように(チャップマン(A.P.Chapman)ら、Nature Biotechnology 17,780−783,1999)(mPEG)リジルマレイミドと反応させて行った。PEG化分子を図6に示す。あるいは、EP−A−1090037号に記載されているように、活性化Fabを(mPEG)リジルビスマレイミドと反応させると、PEG化ジ(修飾Fab)が得られる(以後DFMと呼ぶ)。
ヒトFcに融合した7つのIgドメインヒトKDRを、抗Fcで被覆したチップ上に捕捉し、CDR移植抗体g165の種々の断片とマウス親抗体VR165とを、親和性測定のために流した。以下の表は、得られた結果を要約する。このアッセイフォーマットで、2価の種の低いオフレート(Kd)で示される2価の利点がある。移植したDFMの親和性は、マウスIgGと非常によく似ており、DMF−PEGは親和性のわずかな低下を示す。g165 DFM-PEG分子のKDは、このアッセイでは約4×10-11Mである。
KDRへのVEGF結合をブロックする断片の能力を、放射免疫定量法で測定した。ポリクローナル抗Fcを使用して、マイクロタイタープレート中のヒトFcに融合した7つのIgドメインKDRを補足し、抗体または断片を加え、次に125−I標識VEGFを加えた。このアッセイの結果を図20に示す。このアッセイの構成においてFab’に対するDFMの優れたブロッキング性能により証明される2価の利点がる。ビアコア(BIAcore)試験で見られたように、DFM-PEG構築体は裸のDFMと比較して活性のわずかな低下を示す。
分子g165 DFM PEGはまた、細胞ベースのアッセイで活性を示した。KDRのVEGF刺激をブロックする能力は、ヒト臍帯静脈内皮細胞(クラウス(Clauss)ら、J.Biol.Chem.,271,17629−17634,1996)による組織因子放出を介して証明された。活性はまた、ヒト微小血管内皮細胞(カニンガム(Cunningham)ら、Am.J.Physiol,276,C176−181,1999)におけるVEGF介在Ca2+動員の阻害により証明した。

- 重鎖のC末端の1つのシステイン残基に結合した、リジル−マレイミドまたはリジルビス−マレイミド基を有する、請求項12の抗体分子を含む化合物であって、
リジル残基の各アミノ基は、分子量約20,000Daのメトキシポリ(エチレングリコール)残基をそれに共有結合している、上記化合物。 - 請求項1〜21のいずれか1項に記載の抗体分子の重鎖および/または軽鎖をコードするDNA配列。
- 請求項27のDNA配列を含有するクローニングベクターまたは発現ベクター。
- 請求項27のDNA配列を含む大腸菌(E.coli)発現ベクター。
- pTTOD(CDP791)である請求項30の大腸菌(E.coli)発現ベクター。
- 請求項29〜31のいずれか1項に記載のベクターを用いて形質転換された宿主細胞。
- 請求項32の宿主細胞を培養し、抗体分子を単離することを含む、請求項1〜21のいずれか1項に記載の抗体分子の産生方法。
- 請求項29〜31のいずれか1項に記載のDNA配列を含む大腸菌(E.coli)発現ベクターを含む大腸菌(E.coli)を培養し、抗体分子を単離することを含む、請求項1〜21のいずれか1項に記載の抗体分子の産生方法。
- 抗体分子はペリプラスムに標的化される、請求項34の方法。
- 請求項1〜21のいずれか1項に記載の抗体分子または請求項23〜27のいずれか1項に記載の化合物を含む、治療用または診断用組成物。
