Abstract
Description
GGBPは、E.coliのMg1B−1遺伝子によってコードされている。Mg1B−1遺伝子の部位特異的変異誘発によりアミノ酸であるシステインを様々な位置に導入することによって、このタンパク質を変化させた。次いで、E.coli中でこれらのタンパク質を発現させ、そして精製した。
部位特異的変異誘発またはカセット式変異誘発によってGGBP変異を設計した。部位特異的変異誘発(QuikChange, Stratagene, La Jolla, CA)を実行し、1つのアミノ酸を別のアミノ酸(特に選択した他のアミノ酸)で置換することによって、pQE70ベクタの個々のアミノ酸を変更した。カセット式変異誘発法(非特許文献17)を実行して、GGBP遺伝子の指定された領域のアミノ酸をランダム化した。次いでこれらの変異したカセットをpQE70発現ベクタの中でサブクローン化した。
Claims (32)
- a)少なくとも1つの変異結合タンパク質およびこれに結合した少なくとも1つのチオール基と、
b)前記チオール基を介して前記変異結合タンパク質がカップリングした少なくとも1つのセンサー表面と
を含み、前記センサー表面は、前記変異結合タンパク質が分析物に結合するときの屈折率の変化に起因する検出可能な信号を生成することを特徴とするバイオセンサー。 - 前記変異結合タンパク質は、グルコース/ガラクトース結合タンパク質のなかから選択されることを特徴とする請求項2に記載のバイオセンサー。
- 前記分析物はグルコースまたはガラクトースであることを特徴とする請求項1に記載のバイオセンサー。
- 前記変異グルコース/ガラクトース結合タンパク質は少なくとも1つのアミノ酸置換を含むことを特徴とする請求項2に記載のバイオセンサー。
- 前記変異グルコース/ガラクトース結合タンパク質は少なくとも2つのアミノ酸置換を含むことを特徴とする請求項2に記載のバイオセンサー。
- 前記変異グルコース結合タンパク質は、位置1のシステイン、位置1のセリン、位置11のシステイン、位置14のシステイン、位置19のシステイン、位置43のシステイン、位置74のシステイン、位置107のシステイン、位置110のシステイン、位置112のシステイン、位置113のシステイン、位置149のシステイン、位置213のシステイン、位置216のシステイン、位置238のシステイン、位置287のシステイン、位置292のシステイン、位置152のシステイン、位置182のシステイン、位置236のシステインおよび位置296のシステインからなる群から選択される少なくとも1つのアミノ酸置換を含むことを特徴とする請求項4に記載のバイオセンサー。
- 前記変異結合タンパク質は少なくとも1つのヒスチジンタグを有することを特徴とする請求項4に記載のバイオセンサー。
- 前記変異グルコース/ガラクトース結合タンパク質は位置213に存在するシステインを含むことを特徴とする請求項2に記載のバイオセンサー。
- 前記変異グルコース結合タンパク質はヒスチジンタグをさらに含むことを特徴とする請求項8に記載のバイオセンサー。
- 前記変異グルコース結合タンパク質は、位置149に位置する前記センサー表面にカップリングしたシステインを含むことを特徴とする請求項2に記載のバイオセンサー。
- 前記変異グルコース結合タンパク質はさらにヒスチジンタグを含むことを特徴とする請求項10に記載のバイオセンサー。

- 前記変異グルコース結合タンパク質はヒスチジンタグをさらに含むことを特徴とする請求項12に記載のバイオセンサー。
- 前記変異グルコース結合タンパク質はヒスチジンタグをさらに含むことを特徴とする請求項13に記載のバイオセンサー。
- a)少なくとも1つの変異結合タンパク質およびこれに結合した少なくとも1つのチオール基を提供する工程と、
b)前記チオール基を介して前記変異結合タンパク質が結合した少なくとも1つのセンサー表面と、
c)前記変異結合タンパク質を様々なグルコース濃度を含む生物学的溶液に曝す工程と、
d)屈折率の変化に起因する検出可能で可逆的な信号を検出する工程と
を含み、前記検出可能で可逆的な信号は、前記様々な分析物濃度に対応する結合後の屈折率の変化に起因するものであることを特徴とする分析物検出方法。 - 前記検出は、連続的であるか、プログラムされているか、散発的であるか、またはこれらの組合せであることを特徴とする請求項16に記載の方法。
- 前記少なくとも1つの変異結合タンパク質はグルコース/ガラクトース結合タンパク質であることを特徴とする請求項16に記載の方法。
- 様々な分析物濃度の検出可能で可逆的な信号の前記検出は、in vivoでの検出であることを特徴とする請求項16に記載の方法。
- 前記分析物はグルコースまたはガラクトースであることを特徴とする請求項19に記載の方法。
- 前記変異グルコース/ガラクトース結合タンパク質は、細菌ペリプラズムの結合タンパク質から選択されることを特徴とする請求項19に記載の方法。
