Abstract
Description
−VPAは抗てんかん薬である。
−VPAは催奇性である。抗てんかん薬として妊娠中に用いられた場合、VPAは生まれた子供の2〜3%に先天性欠損(神経管閉鎖障害及びその他の奇形)を起す可能性がある。マウスにおいて、VPAは、適切に投与すると大多数のマウス胎児について催奇性である。
−VPAは核内ホルモン受容体(PPARδ)を活性化する。他のいくつかの転写因子もまた抑制解除されるが、それほど抑制解除されない因子(グルココルチコイド受容体、PPARα)もある。
−VPAは時に肝毒性を引き起こす。これは代謝され難い補酵素Aとのエステルによる可能性がある。
この目的のため、驚くべきことに、VPAが、それぞれ全く異なった作用機序に基づく極めて多様な他の抗腫瘍療法との併用で使用された場合に、おそらく多くの異なった型のヒト癌の治療では予期し得なかった有益な効果を持つことが今や見出された。したがって、VPAを多くの抗腫瘍剤の組合せにおける一成分として治療に使える可能性は、特定の分子機構を有する薬剤との組合せに限られるものではないと思われる。このことは、実際VPAを現存する抗腫瘍アプローチの大半と組合せられる薬剤とするであろう。ここでVPAの正確な作用機序は完全には解明されていないが、その分化誘導能がそのような広範な抗腫瘍薬剤の活性に対して腫瘍細胞を感受性にする基礎となる可能性がある。このVPAの驚くほど幅広い潜在能力は、VPAの、HDAC活性を有する酵素の特定の組合せに対する阻害剤としての活性に基づいているものと考えられる。
このような薬剤は、通常標準的な外科的手法に加え、活発に増殖及び分裂しているいかなる細胞にも損傷を与えることを目的として使用される。大抵の場合、正常細胞よりも癌細胞の方に増殖及び分裂過程にある細胞が多いので、化学療法及び細胞毒性は正常細胞より癌細胞により重大な影響を与える。化学療法/細胞毒性薬剤は明確に分類することができ、
− アルキル化剤
− 細胞毒性抗生物質
− 代謝拮抗剤
− ビンカアルカロイド及びエトポシド
− その他
などが挙げられる。
細胞毒性抗生物質は、直接的にDNA又はRNA合成を阻害することにより作用し、全細胞周期を通して有効である。
代謝拮抗剤は、細胞分裂過程に関連する細胞の酵素あるいは天然の代謝産物と相互作用し、かくして細胞分裂を混乱させる。
“その他”に分類された化合物群には、主としてタキサン類(例えば、パクリタキセル、タキソール、ドセタキセル、タキソテールなど)及び金属錯体(例えば、シスプラチナム)が含まれる。
ある種の癌、例えば乳癌あるいは前立腺癌等の進行は、からだの中のホルモンの過剰あるいは不在に依存している。これらの場合、これらの器官での腫瘍増殖を抑制すべく体内のホルモンレベルを上昇又は下降させるため、ホルモン療法が用いられる。
ホルモン療法の区分には5種類の主要な製品がある。すなわち、
− プロゲストーゲン類
− 抗アンドロゲン類
− 抗エストロゲン類
− 黄体形成ホルモン放出ホルモン(LHRH)類似体
− アロマターゼ阻害剤
である。プロゲストーゲン類は子宮内膜癌の治療において使用される。なぜなら、これらの癌はプロゲストーゲンに対抗されていない高レベルのエストロゲンに晒された女性に発生するからである。
1990年代までは、細胞毒及びホルモン療法が癌の薬剤処置の基礎であった。しかしながら、最近の進歩により革新的療法の区分に

かくして、本発明のもう一つの特徴は、VPA及びその誘導体の、腫瘍転移の抑制及び微小残存病変の消滅のための使用である。
VPAのこの治療上の成功の根拠として最も可能性があるのは、VPAの、HDAC活性を有する酵素の新規な阻害剤としての活性である。しかしながら、例えばTSAはVPAが示すほどの相乗活性を示さないので(例えば図21及び25参照)、VPAによって成し遂げられた微調整された機構的ターゲッティングは、他のHDAC阻害剤に勝ると思われる。
加えて、VPAは腫瘍転移形成の抑制に用いられてもよく、したがって微小残存病変の処置に用いられてもよい。このことは、腎臓癌及び乳癌のインビボモデルを用いて成功裏に調べられた。
VPA又は化学療法剤/細胞毒性薬剤の5−フルオロウラシル単独及びVPAと5−フルオロウラシル(5−FU)の組合せによる処置による、HCT−15結腸癌細胞の総細胞量の相乗的な減少(図1)。
細胞量の減少はSRB試験により測定した。この試験用に、細胞を96ウェル培養皿にウェル当たり細胞数3,000から8,000の密度で播種した。24時間の回復の後、それらをVPA非存在あるいは示された濃度のVPAの存在下で48時間培養した。培養物を冷TCAで最終TCA濃度が10%になるようにして固定し、4℃で1時間培養後、細胞を5回水洗しついで風乾した。固定した細胞を、1%酢酸に溶かした0.4%(wt/vol)スルホローダミンB(SRB)で30分間染色し、1%酢酸で4回洗浄して結合していない染料を除いた。風乾後、結合した染料を10mMの緩衝化していないトリス塩基(pH10.5)で5分間可溶化し、光学密度をTitertek Multiskan Plus分光測定用プレートリーダー上550nmで読んだ。各用量応答につき6個のテスト用ウェルを、細胞株ごとの12個の増殖対照ウェルと平行に設置した。時間0(T0;薬剤が添加された時点)における細胞集団密度の測定も、テストプレートに薬剤を加える直前に、12個の追加の参照ウェルの細胞をTCAで固定して行った。上記と同様に固定、染色した5%FBSを含む完全培地の背景ODも12個の別のウェルにおいて測定した。それぞれのマイクロタイター・プレートからの未処理ODデータから、背景OD測定値(すなわち、完全培地プラス染色分のOD及びT0における細胞のOD)を差し引き、その細胞の細胞生物量の減少を求めた。
HCT−15細胞をVPA非存在下又は表示濃度のVPA単独の存在下(図1A)、あるいは5−FUの非存在下又は表示濃度の5−FU単独の存在下、あるいは0.75mM VPAとの併用の下で(図1B)48時間培養した。細胞量の相乗的な減少が、VPAと5−FU両者の組合せ処置において、VPAあるいは5−FU単独での処置と比較して観察された。これは低濃度の5−FUを用いた場合に特に顕著であった。例えば、5−FUを単独で0.5μM未満の用量で用いた場合には、細胞量の観測値の増大さえもたらしたが、一方で同量の5−FUを0.75mMのVPAと併用した場合には、0.75mMのVPA単独使用よりもさらに大きな細胞量の減少をもたらした。したがって、これら二つの薬剤の組合せ活性は、この処置が相乗的な活性をもつとして説明すべきである。
〔実施例2〕
特に低濃度のシスプラチナムが用いられた場合に、相乗的な細胞量観測値の減少が見られた。なぜなら、シスプラチナムを単独で1μM未満の用量で用いた場合には、細胞量観測値になんらの減少ももたらさなかったからである。これと対照的に、同じ投与量のシスプラチナムと1mM VPAとの併用では、1mM VPA単独使用と比べて細胞量が減少した(図2B)。このように、これら二つの薬剤の組合せ活性は、この処置が相乗的な活性をもつとして説明すべきである。
ここでは、特に、10μM未満のシスプラチナム濃度を用いた場合、相乗的な細胞量観測値の減少があった。なぜなら、シスプラチナムを単独でこれらの投与量で用いても細胞量の減少はもたらされなかったからである。これと対照的に、同じ投与量のシスプラチナムと0.75mMのラセミ体4−yn VPAとの組合せでは、0.75mMの4−yn VPA単独使用と比べて細胞量の減少がもたらされた(図2D)。このように、これら二つの薬剤の組合せ活性は、この処置が相乗的な活性をもつとして説明すべきである。

VPAあるいはゲムシタビン単独処置と比べ、VPAとゲムシタビンの同時組合せ処置において細胞量の明瞭な相加的減少が観察された。
〔実施例5〕
両組合せ処置において明瞭な細胞量の減少が見られ、これはカンプトテシンとの併用で、VPAのみならずその誘導体である4−yn VPAも、他の抗癌剤と併用すると、ここでカンプトテシンについて示されたように、同様の少なくとも相加的な治療効果をもたらす付加的かつふさわしい作用を有することを示すものである。
〔実施例6〕
ヒトMDA−MB468、MDA−MB453及びSKBR3乳癌細胞及びA431扁平上皮癌を、10%の熱不活化ウシ胎児血清(FBS)、2mM L−グルタミン、100単位/mlペニシリン及び100μg/mlストレプトマイシンを補充したダルベッコ修飾イーグル培地(DMEM,BioWhittaker, Verviers, Belgium)中で維持した。
腫瘍細胞を通常の増殖培地中96ウェルプレートに1×104細胞/ウェルの密度で播種し、70時間の間、最終濃度3mMのバルプロ酸、2μg/mlの治療用アンチErbB2抗体ヘルセプチンTM、2μg/mlの治療用アンチEGF受容体抗体 c225(Fan & Mendelsohn, Curr. Opin. Oncol.,10: 67-73, 1998)、又はバルプロ酸と上記と同じ濃度のヘルセプチンTMあるいはc225抗体との組合せで処理した。対照細胞はバルプロ酸又は抗体の非存在下で増殖させた。各ウェルに10mg/mlの3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド(MTT)(Sgma,Deisenhofen, Germany)PBS溶液を10μl加え、さらに3時間細胞を培養した。細胞を90μlの溶解用緩衝液(20%SDSの50%ジメチルホルムアミド溶液、pH4.7)を加えて溶解した。ホルマザン生成物を可溶化させた後、590nmにおける吸収をマイクロプレート・リーダー(Dynatech, Denkendorf, Germany)中で測定し、生存細胞の相対量をバルプロ酸あるいは抗体を加えずに培養した細胞と比較して計算で求めた。
図6に示された結果は、バルプロ酸及び治療用抗体ヘルセプチンTM及びc225は、それぞれ単独薬剤として乳癌細胞及び扁平上皮細胞癌の生存率を抑制することを示している。しかしながら、バルプロ酸と治療用抗体ヘルセプチンTMの併用処置は、SKRB3細胞において顕著な相加的治療効果をもたらしている。しかし、さらに興味深いことには、VPAとの組合せ処置において、細胞生存率の相乗的な減少が試験した上記以外の3つの細胞株において観察されたことである。すなわち、MDA−MB453細胞におけるヘルセプチンTMとの併用、及びMDA−MB468とA431細胞におけるc225との併用である。これらの結果は、バルプロ酸が治療用抗体と併用されると、強く高められた治療効果を示すこと、そして上皮起源の固形癌に由来する多種多様な腫瘍細胞の生存率を強力に抑制することを実証するものである。さらにこの結果は、バルプロ酸及びその誘導体が治療用抗体との併用で、それらの腫瘍の療法として相乗的な治療的成功を収めつつ使用できるであろうことを示すものである。
〔実施例7〕
ヒトSKBR3乳癌細胞、A431扁平上皮癌細胞及びSKOV3卵巣癌細胞を10%の熱不活化したウシ胎児血清(FBS)、2mM L−グルタミン、100単位/mlペニシリン及び100μg/mlストレプトマイシンを補充したダルベッコ修飾イーグル培地(DMEM,BioWhittaker, Verviers, Belgium)中で維持した。
E.coliβ−ガラクトシダーゼをコードするpZeoSV2/lacZプラスミドを安定に遺伝子導入された腎臓細胞癌(Renca)細胞で、さらにcerbB2及びネオマイシン抵抗性をコードするプラスミドpSV2ErbB2N及びpSV2neoを導入されたRenca細胞(Renca−lacZ/ErbB2)(Maurer-Gebhard et al., Cancer Res. 58:2661-2666, 1998)、あるいは上皮性増殖因子(EGF)受容体、腫瘍原性に活性化されたEGF受容体変異株EGFRvIII及びネオマイシン抵抗性をコードしているプラスミドpLTR−EGFR又はpLTR−EGFRvIII及びpSV2neoを導入されたRenca細胞(Renca−lacZ/EGFR及びRenca−lacZ/EGFRvIII)(Schmidt et al., Oncogene 18: 1711-1721, 1999)を8%FBS、2mM L−グルタミン、100U/mlペニシリン、100μg/mlストレプトマイシン、0.25mg/ml Zeocin及び0.48mg/ml G418を補充したRPMI−1640培地中で増殖させた。
腫瘍細胞を通常の増殖培地中96ウェルプレートに1×104細胞/ウェルの密度で播種した。SKBR3、A431,及びSKOV3細胞は40時間の間、最終濃度3mMのバルプロ酸、又は10ng/mlの組換えアンチErbB2単鎖抗体毒素scFv(FRP5)−ETA(Wels et al., Cancer Res. 52: 6310-6317,1992)、又は10ng/mlの組換えアンチEGF受容体単鎖抗体毒素scFv(14E1)−ETA(Schmidt et al., Brit. J. Cancer 75: 1575-1584, 1997)、あるいはバルプロ酸と上記と同濃度のscFv(FRP5)−ETA又はscFv(14E1)−ETAのいずれかとの組合せにより処理した。Renca−lacZ/ErbB2、Renca−lacZ/EGFR及びRenca−lacZ/EGFRvIII細胞は40時間の間、最終濃度1mMのバルプロ酸、又は10ng/mlの組換えアンチErbB2単鎖抗体毒素scFv(FRP5)−ETA、又は1ng/mlの組換えアンチEGF受容体単鎖抗体毒素scFv(14E1)−ETA、あるいはバルプロ酸と上記と同濃度のscFv(FRP5)−ETA又はscFv(14E1)−ETAいずれかとの組合せにより処理した。対照細胞はバルプロ酸あるいは抗体毒素の非存在下で増殖させた。各ウェルに10mg/mlの3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド(MTT)(Sigma, Deisenhofen, Germany)PBS溶液を10μl加え、さらに3時間細胞を培養した。90μlの溶解用緩衝液(20%SDSの50%ジメチルホルムアミド溶液、pH4.7)を加えて細胞を溶解した。ホルマザン生成物を可溶化させた後、590nmにおける吸収をマイクロプレート・リーダー(Dynatech, Denkendorf, Germany)中で測定し、生存細胞の相対量をバルプロ酸あるいは抗体毒素を加えずに培養した細胞と比較して計算で求めた。
提示した結果(図7及び8)は、バルプロ酸及びアンチErbB2又はアンチEGF受容体免疫毒素は、単独の薬剤として乳癌、卵巣癌、腎臓癌及び扁平上皮癌の細胞の生存率を抑制することを示している。しかしながら、バルプロ酸とアンチErbB2又はアンチEGF受容体免疫毒素との併用処置は、顕著な付加的治療効果をもたらす。これらの結果は、バルプロ酸が免疫毒素等の抗体融合タンパク質との併用で強く高められた治療効果を示すこと、そして上皮起源の固形癌に由来する多種多様な腫瘍細胞の生存率を強力に抑制することを実証するものである。さらにこの結果は、バルプロ酸及びその誘導体が、免疫毒素類との併用でそれら腫瘍の治療法として使用できるであろうことを示す。
〔実施例8〕
Renca細胞及び遺伝子導入された細胞クローンRenca−lacZ(β−ガラクトシダーゼをコード)を10%のウシ胎仔血清(FCS)を補充したRPMI−1640中で培養した。宿主マウスに転移を形成するには、4〜6週齢のメスBalb/cマウスに側面尾静脈中に100μlのPBS中の105個のRenca−lacZ細胞を注射した。1群につき5匹の動物を、腫瘍細胞注入後4週まで1週間隔で屠殺し肺を切除した(Maurer-Gebhard et al., 1998, Cancer Research 58, 2661-2666)。
VPAでの処置は次の通りである。すなわち、2×400mg/kg/日 腹腔内(Na−VPA, 水中155mM)。対照動物はグルコース溶液で処置した(Maurer-Gebhard et al., 1998, Cancer Research 58, 2661-2666)(図9)。
肺転移の可視化のためのX−gal染色:
切除した肺を終夜、4℃で、2%ホルムアルデヒド及び0.2%グルタルアルデヒドを含むPBS中で固定した。固定液を除去し、肺をPBSで洗浄した。X−Gal溶液での染色は、37℃、暗所で10から12時間行った(Maurer-Gebhard et al., 1998, Cancer
Research 58, 2661-2666)。転移性表面小結節を解剖顕微鏡のもとで分析し、代表的な写真を撮った。それらを図9に示す。
〔実施例9〕
MT450細胞を、DMEM/10%FCS培質中で培養し、注射直前にマイコプラズマが存在しないことを検査した。細胞をPBS中で2回洗い、PBS1ml当たり5×106細胞の密度に懸濁した。0.1mlのPBS中の5×105個の細胞を各ラットに注射した。それぞれ各8匹2群のラットを用いた(それぞれ±VPA)。ラットは原発癌を増殖させるべく21日間放置した。VPAナトリウム塩を155mM(等張)の水に溶かした。pHを少量の塩酸により6から7に調整し、得られた溶液を滅菌ろ過した。化合物は腹腔内注射で施用した。投与量は250gのラット当たり2mlであり、これはkgBW(体重)及び1投与量当たり1.25mmol VPAに相当する。1日2投与を適用した。対照動物には同量の無菌等張塩化ナトリウム溶液を投与した。
原発腫瘍の成長を腫瘍体積を測ることにより追跡し、VPAが腫瘍の拡大を遅らせることが分かった。実験は、対照群のラットの1匹における腫瘍サイズが法で定められている限度の50mmに達した時点で終了とした。その時点で肺転移を評価するため、試験に用いた全てのラットを剖検した。対照群のすべてのラットで転移が顕著に進行していた。代表例を図10に示した。転移はVPA処置ラット8匹中7匹にも見られた。しかし、転移のサイズと数はNaCl処置ラットと比較してずっと小さかった。代表例を図10に示した。投与量決定のための実験は、上記で選んだ投与量プロトコルは高い初期血清レベルを生じる(例えば、腹腔内注射後1時間で3.6mM)が、それは急速に低下し(例えば、腹腔内注射後4時間で0.25mMなど)、ヒトでのてんかん治療中に維持されるレベルより低くなることを示した。要約すると、実験は、VPAはゲッ歯類の血清からのクリアランスが早く、有効範囲として期待される0.5mM以上のVPA血清レベルを維持することはできないにもかかわらず、VPA処置によりMT450ラット乳癌モデルにおいて原発腫瘍の増殖及び肺転移を顕著に抑制した。したがって、微小残存腫瘍症を抑制するために用いられてもよい。
〔実施例10〕
VPA処置は対照細胞の分化に影響を与えなかったが、一方、PML−RARの発現は強力な分化遮断を起し(図11)、それは上記のサイトカインのみによる処置では克服できなかった。しかしながら、VPA(1mM,右区画、PML−RARと表記したもの)は、PML−RARにより課された分化遮断をほぼ完全に回復させた(図11)。このように、RAなしで、VPAは、恐らくはPNL−RARにより対象遺伝子に動員されたHDAC複合体の作用を阻害することによって、細胞を分化が許容される状態に再建及び増感し、ついで、サイトカインにより分化が誘導される。このVPAの活性は相乗的な活性と見なされなければならない。なぜなら、上記のようにサイトカイン類のみでの処置では、これらのPML−RAR細胞における分化遮断を相当程度解き放つことには至らなかったからである。
ネズミの造血前駆細胞を、129匹のマウスの骨髄から系統分化マーカーがないこと(lin−)を基礎に精製した。Lin−細胞は、IL−3(20ng/ml)、IL−6(20ng/ml)、SCF(100ng/ml)の存在下で48時間培養し、ついでレトロネクチン(宝酒造)でコートした組織培養に未使用のプレートに付着させた。細胞は次いで、対照レトロウイルス性ベクターPINCO、又はPINCO−PML−RARで一過性に遺伝子導入したフェニックス同種指向性パッケージング細胞からの上澄み(上記のように新鮮血清及びIL−3、IL−6及びSCFで補充されている)と培養することにより形質導入した。60時間後、GFP+細胞をFACSにより選別し、上記と同様のものに加えてG−CSF(60ng/ml)及びGM−CSF(20ng/ml)を補充したメチルセルロースプレートに播種した。それと表示したものでは、バルプロ酸ナトリウム(VPA,左から右へ0.2あるいは1mM)を分化用媒質に加えた。8〜10日後、細胞を骨髄球分化マーカーMac−1の存在についてFACSで分析した。VPAは1mMまでの濃度は、Mac−1+細胞の数にも対照細胞のコロニーの数にも顕著な変化をもたらさなかった。より高い濃度(>3mM)では、コロニー数の減少が観察され、これは恐らく細胞死の導入による(データ示さず)。対照として、赤血球系の分化マーカー(Ter−119)を有するVPA−処置細胞を調べたが陽性細胞は見つからなかった(データ示さず)。対照ベクターのPINCOベクターの影響を受けていない細胞と影響を受けた細胞の反応パターンは同一であった(データ示さず)。
〔実施例11〕
X線処置(2グレイ)によって、造血前駆細胞は生存ポテンシャルの大幅な減少を示し、アポトーシスを受ける。半固体培養条件(メチルセルロース基盤媒質)では野生型細胞培養物をX線処理したものにおいては、ほとんど完全にコロニー(コロニー形成細胞、CFC由来)が存在しなくなる。このことは、未分化細胞はこの処置に非常に感受性であることを示す(図12)。際だっているのは、PML−RARの発現により、救助率>50%と対象細胞のX線感受性を著しく低下させたことである(図12)。同条件下で、VPA(1mM)は野生型細胞の感受性をわずかに減少させた。しかしながら、VPAはVPA処理細胞においてコロニーの完全かつ明らかに相乗的なコロニーの消滅を伴うPML−RAR発現細胞の再感作をもたらした(図12)。したがって、VPAはX線との組合せにおいて、X線のみの処置に耐性となった(例えば、癌原性融合タンパク質の発現を介して)細胞の感受性を救い、相乗的な治療成功率をもたらすことができよう。
図11について記載したと同様にしてLin−細胞に形質導入し(詳細については実施例10での方法を参照)、そしてFACSにより選別した。選別の12時間後、細胞をPBSで洗浄し、サイトカインを含む媒質(図12に提示したデータはIL−3(20ng/ml)、L−6(20ng/ml)、SCF(100ng/ml)、G−CSF(60ng/ml)、GM−CSF(20ng/ml)を用いている)中、又はサイトカインを含まない媒質(データ示さず)中で8〜12時間培養した。培養の終わりに、細胞をX線源にさらし(合計曝露量2Gy)、さらにVPAの存在下あるいは非存在下で12〜16時間培養した。最後に、細胞をサイトカイン(IL3、IL6、SCF、G−及びGM−CSF)存在下でメチルセルロースを含む媒質(StemCell Technologies)中に播種し、8〜10日間培養した。
〔実施例12〕
最も顕著であったのは、レチノイン酸とVPAの併用で最も長い生存延長を示し、末梢血中及び調べた内臓(骨髄、脾臓)中に、全処置期間中を通して白血病芽細胞を認めなかったことであった(図13)。白血病のインビボモデルにおけるレチノイン酸とVPAの併用の際のこの印象的な結果は、VPAを、白血病の処置において、少なくとも相加的な、しかし相乗的である可能性がより高そうな、治療的生物学的応答を誘起するために、分化誘導剤(例えばレチノイン酸)との併用で投与できるであろうことを示す。
PML−RARを発現している細胞の接種後に白血病を発病したマウスを屠殺した。脾細胞の単細胞懸濁液を調製し、第二次の宿主マウスに107個の細胞を再接種した。白血病芽細胞の存在が末梢血中に明らかであり、内臓(骨髄、脾臓)が既に顕微鏡的に見ると白血病細胞により侵された時点で、VPA(400mg/kg)の12時間置きの腹腔内注射、及びレチノイン酸の徐放性ペレットの皮下への埋め込みによる治療を開始した。VPA処置は、次のスケジュール、すなわち5日×2回と2日間欠期を3連続週行なった。
〔実施例13〕
細胞を3日間VPAで前処置し、次いで48時間まで、VPA+AD、VPA+FU又はVPA+ITによる同時処置を行った(図17A〜C)。驚くべきことに、VPAによる前処置により、どの化学療法剤に晒された細胞においても目覚しい相乗的なアポトーシスの亢進を引き起した(図17A〜C)。並行して行った実験で、VPAを除去(3日の前処置につづき24時間の洗い流し)した場合には、感受性は失われた。このことは、VPAがその感作作用を達成するにはVPAが化学療法剤と同時に投与されなければならないことを示している(図17A〜C)。これらの結果を総合すると、VPAの使用は腫瘍細胞を抗腫瘍活性を有するいくつかの薬剤の作用に対し感作させ、そのような組合せ治療処置における相乗的な活性をもたらす可能性があることを示している。
図14については、細胞は6ウェル培養皿に100,000細胞/ウェルで播種した。次の日、それらを表示した薬剤(5−FU:2μM 5−フルオロウラシル、AD:20ng/ml アドリアマイシン、IT:3μM イリノテカン)で処置し、さらに48時間培養した。処置の24時間及び48時間後に細胞数を数えた。全てのアッセイは3回ずつ行った。
細胞周期分析の結果はアポトーシス細胞の割合(%)で表わしてある。全てのアッセイは3回ずつ行った。
上部パネルは生存細胞の3回計数の結果である。下部パネルでは、細胞は透過化処理及び固定後にヨウ化プロピディウムで染色された。細胞周期分析の結果はアポトーシス細胞の割合(%)で表わしてある。全てのアッセイは3回ずつ行った。

Kasumi−1細胞(図20)及びMO、M2及びM4 FABのサブタイプに属するAML芽細胞(図22)において、我々はVPAが単独薬剤として部分的な骨髄球分化を誘導すること、そしてRAとの併用で、後骨髄球様又は好中球様形態を有する細胞の出現及びNBT染料還元アッセイにおける陽性細胞の数の増加(65%まで)(Kasumi−1細胞については図20及び21を、AML芽細胞については図22及び24を参照)により明らかになった、完全でそれゆえ相乗的な骨髄球分化を誘発することを見出した。加えて、Kasumi−1細胞において細胞周期分析から、VPAとRAの組合せを用いた処置により、VPA又はRA単独での処置と比べて、G1期停止への細胞周期の移行が促進されていることが分かった(図23)。更に興味深いことには、AML芽細胞において、VPAとRAの組合わせによる処置は、特定の遺伝学的損傷の存在とは独立して骨髄球の分化に影響を与えた(図22)。
