Description
【特許請求の範囲】
【請求項1】 患者の自己免疫および慢性炎症性状態を処置するための組成物であって、CD30LへのCD30の結合を阻害できる剤を含み、ここで前記剤は、患者の状態の重度を反映する少なくとも一つの指標における持続された改善を誘導するために適している用量および頻度の投与計画に従って投与され、もし少なくとも1日離れた少なくとも二つの時点で患者が改善を示すとすれば、改善は持続されたと考えられる、前記組成物。
【請求項2】 患者がヒトである、請求項1に記載の組成物。
【請求項3】 前記剤が以下の(a)−(c):
(a)CD30Lに特異的である抗体;
(b)CD30に特異的である非アゴニスト的抗体;並びに
(c)以下の(i)−(iii):
(i)配列番号6のヒトCD30ポリペプチドのアミノ酸19−390;および
(ii)(i)の配列の断片、ここで該断片はCD30Lを結合できる;
(iii)配列番号6のアミノ酸19−390と少なくとも90%の同一性を持っているCD30−結合ポリペプチド
からなる群より選択されるアミノ酸配列を含む可溶性CD30ポリペプチド
からなる群より選択される、請求項1に記載の組成物。
【請求項4】 前記剤が、配列番号6のアミノ酸19−390またはそれらのCD30L−結合断片を含む可溶性ヒトCD30ポリペプチドであり、該ポリペプチドがさらにヒト免疫グロブリンのFc領域を含む、請求項2に記載の組成物。
【請求項5】 状態が関節炎である、請求項2に記載の組成物。
【請求項6】 状態がリューマチ性関節炎である、請求項5に記載の組成物。
【請求項7】 前記剤が、CD30Lに特異的である抗体、配列番号6のアミノ酸19−390を含む可溶性ヒトCD30ポリペプチド、配列番号6のアミノ酸19−390のCD30L−結合断片、配列番号6のアミノ酸19−390およびヒト免疫グロブリン蛋白質のFc領域を含む融合蛋白質、並びに配列番号6のアミノ酸19−390のCD30L−結合断片およびヒト免疫グロブリン蛋白質のFc領域を含む融合蛋白質、からなる群から選択される、請求項6に記載の組成物。
【請求項8】 前記剤が、TNFα、IL−1α、IL−1βまたはIL−4のアンタゴニストである第二の剤と同時に投与される、請求項3に記載の組成物。
【請求項9】 患者の状態が、多発性硬化症、全身性硬化症、急性炎症性脱髄性多発性神経症、急性運動軸索神経症、急性運動感覚軸索神経症およびフィッシャー症候群からなる群より選択される、請求項2に記載の組成物。
【請求項10】 患者の状態が全身性紅斑性狼瘡である、請求項2に記載の組成物。
【請求項11】 CD30/CD30L相互作用を遮断できる前記剤がCD30L特異的抗体であり、そして前記第二の剤がIL−4のアンタゴニストである、請求項8に記載の組成物。
【請求項12】 IL−4アンタゴニストが、IL−4特異的抗体、IL−4R特異的抗体、並びに配列番号16のアミノ酸1−207または2−207を含む可溶性IL−4レセプター、からなる群より選択される、請求項11に記載の組成物。
【請求項13】 患者の状態が全身性紅斑性狼瘡、強皮症および尋常性天疱瘡からなる群より選択される、請求項12に記載の組成物。
【請求項14】 TNFα阻害剤での処置に抵抗性である自己免疫または慢性炎症状態を処置するための組成物であって、CD30LへのCD30の結合、またはIL−R1へのIL−1αまたはIL−1βの結合を阻害し、それによりCD30またはIL−1によるシグナル伝達を遮断することのできる剤を含み、ここで、前記剤は、患者の状態の重度を反映する少なくとも一つの指標における持続された改善を誘導するために適している用量および頻度の投与計画に従って投与され、もし少なくとも1日離れた少なくとも二つの時点で患者が改善を示すとすれば、改善は持続されたと考えられる、前記組成物。
【請求項15】 前記剤がCD30、CD30L、IL−1α、IL−1βまたはIL−1R1に特異的である抗体であり、そしてさらにここで、該抗体がポリクローナル抗体、モノクローナル抗体、ヒト化抗体及びヒト抗体からなる群より選択される、請求項14に記載の組成物。

【請求項18】 治療剤をスクリーニングするための動物モデルであって、該動物モデルは以下の(a)−(b):
(a)そのp55およびp75 TNFαレセプター蛋白質を不活性化する遺伝子修飾を保持している;および
(b)実験的に誘導される関節炎に対して遺伝的に感受性である。
により特徴付けられている、前記動物モデル。
【請求項19】 動物モデルが遺伝的に感受性である関節炎が、コラーゲン−誘導関節炎である、請求項18に記載の動物モデル。
【請求項20】 動物モデルが、DBA/1、BUBおよびB10.Qからなる群から選択されるマウスの系統、あるいはDA、BB−DRおよびLEWからなる群から選択されるラットの系統である、請求項19に記載の動物モデル。
【請求項21】 動物モデルがマウスの系統であり、およびマウスの系統がDBA/1であり、そしてさらにここで、マウスのDBA/1系統がそのp55およびp75 TNFαレセプター遺伝子に二重ヌル突然変異を持っている、請求項20に記載の動物モデル。
【請求項22】 TNFα阻害剤での処置に抵抗性である自己免疫または慢性炎症性状態の処置における効力(efficacy)を決定するための候補治療剤をスクリーニングするために、請求項18の動物モデルを使用する方法であって、該方法は請求項18の動物モデルに関節炎を誘導し、該動物に候補治療剤を投与し、そして、もし候補剤が投与された後に該動物の関節炎の重度が軽減されるなら前記剤は有効であることを決定する、ことを含む、前記方法。
【請求項23】 動物モデルが、コラーゲン−誘導関節炎に対して感受性であるマウスまたはラットの系統である、請求項22に記載の方法であって、ここで関節炎はコラーゲンを注射することにより誘導され、そしてさらにここで、候補治療剤を投与した後、関節炎の重度は動物の足における紅斑および浮腫の量を観察することにより評価される、前記方法。
【請求項24】 動物モデルがDBA/1マウス、BUBマウス、B10.Qマウス、DAラット、BB−DRラットおよびLEWラットからなる群より選択される、請求項23に記載の方法。
【請求項25】 動物モデルが、そのp55およびp75 TNFαレセプター遺伝子に二重ヌル突然変異を保持するDBA/1マウスである、請求項24に記載の方法。
【請求項1】 患者の自己免疫および慢性炎症性状態を処置するための組成物であって、CD30LへのCD30の結合を阻害できる剤を含み、ここで前記剤は、患者の状態の重度を反映する少なくとも一つの指標における持続された改善を誘導するために適している用量および頻度の投与計画に従って投与され、もし少なくとも1日離れた少なくとも二つの時点で患者が改善を示すとすれば、改善は持続されたと考えられる、前記組成物。
【請求項2】 患者がヒトである、請求項1に記載の組成物。
【請求項3】 前記剤が以下の(a)−(c):
(a)CD30Lに特異的である抗体;
(b)CD30に特異的である非アゴニスト的抗体;並びに
(c)以下の(i)−(iii):
(i)配列番号6のヒトCD30ポリペプチドのアミノ酸19−390;および
(ii)(i)の配列の断片、ここで該断片はCD30Lを結合できる;
(iii)配列番号6のアミノ酸19−390と少なくとも90%の同一性を持っているCD30−結合ポリペプチド
からなる群より選択されるアミノ酸配列を含む可溶性CD30ポリペプチド
からなる群より選択される、請求項1に記載の組成物。
【請求項4】 前記剤が、配列番号6のアミノ酸19−390またはそれらのCD30L−結合断片を含む可溶性ヒトCD30ポリペプチドであり、該ポリペプチドがさらにヒト免疫グロブリンのFc領域を含む、請求項2に記載の組成物。
【請求項5】 状態が関節炎である、請求項2に記載の組成物。
【請求項6】 状態がリューマチ性関節炎である、請求項5に記載の組成物。
【請求項7】 前記剤が、CD30Lに特異的である抗体、配列番号6のアミノ酸19−390を含む可溶性ヒトCD30ポリペプチド、配列番号6のアミノ酸19−390のCD30L−結合断片、配列番号6のアミノ酸19−390およびヒト免疫グロブリン蛋白質のFc領域を含む融合蛋白質、並びに配列番号6のアミノ酸19−390のCD30L−結合断片およびヒト免疫グロブリン蛋白質のFc領域を含む融合蛋白質、からなる群から選択される、請求項6に記載の組成物。
【請求項8】 前記剤が、TNFα、IL−1α、IL−1βまたはIL−4のアンタゴニストである第二の剤と同時に投与される、請求項3に記載の組成物。
【請求項9】 患者の状態が、多発性硬化症、全身性硬化症、急性炎症性脱髄性多発性神経症、急性運動軸索神経症、急性運動感覚軸索神経症およびフィッシャー症候群からなる群より選択される、請求項2に記載の組成物。
【請求項10】 患者の状態が全身性紅斑性狼瘡である、請求項2に記載の組成物。
【請求項11】 CD30/CD30L相互作用を遮断できる前記剤がCD30L特異的抗体であり、そして前記第二の剤がIL−4のアンタゴニストである、請求項8に記載の組成物。
【請求項12】 IL−4アンタゴニストが、IL−4特異的抗体、IL−4R特異的抗体、並びに配列番号16のアミノ酸1−207または2−207を含む可溶性IL−4レセプター、からなる群より選択される、請求項11に記載の組成物。
【請求項13】 患者の状態が全身性紅斑性狼瘡、強皮症および尋常性天疱瘡からなる群より選択される、請求項12に記載の組成物。

【請求項15】 前記剤がCD30、CD30L、IL−1α、IL−1βまたはIL−1R1に特異的である抗体であり、そしてさらにここで、該抗体がポリクローナル抗体、モノクローナル抗体、ヒト化抗体及びヒト抗体からなる群より選択される、請求項14に記載の組成物。
