Abstract
【解決手段】角層中のタンパク質の酸化を抑制することにより、角層の透明性及び/又は保水性を保持・向上させる。
【効果】角層タンパク質の酸化が及ぼす肌への影響の解明により、角層酸化タンパク質を指標としたスキンケア手段の提供が可能となり、皮膚学的・化粧学的側面において極めて有益であると予測される。

Description
別の観点からすると、本発明は、角層の透明性及び/又は保水性を指標とする、角層中のタンパク質の酸化度を検出するための方法を提供する。
さらには、本発明は角層中のタンパク質の酸化を抑制することにより、角層の透明性及び/又は保水性を保持・向上させる方法を提供する。
角層又は肌の「透明性」が高いと、あたかも肌が透き通ってみえるような状態になり、美容上きわめて好ましい外観を示すことになる。また、角層又は肌の「保水性」が高い皮膚では肌がみずみずしく、潤いのある健康な状態になる。そのため肌が「透明性」・「保水性」を有することは美容学上極めて重要であり、一般に角層や肌の「透明性」や「保水性」は美容技術者などの美容専門家の主観を頼りとした官能テストや、面倒な操作、高価な装置・設備を必要とする皮膚試料の光学的検査や水分検査などにより行われていた。本発明は角層中の酸化タンパク質を指標とすることで、より詳しくは角層中の酸化タンパク質の存在量、分布状態等を検出することで、角層、ひいては肌の「透明性」・「保水性」をより簡単かつ客観的な手段で調べることを可能とする。

を有するものが好ましい。そのような蛍光物質の例には、フルオレセイン−5−チオセミカルバジド、テキサスレッドヒドラジドなどが挙げられる。
(1)角層試料を、例えばテープストリッピングにより、採取する;
(2)これに適当な緩衝液(例えば100mMのMES-Na緩衝液(pH5.5))中のヒドラジノ基含有蛍光物質を室温にて数時間(例えば1時間)反応させる;
(3)反応終了後に適当な生理溶液(例えば緩衝液リン酸緩衝生理食塩液(PBS))にて十分に洗浄した後、蛍光顕微鏡にて酸化タンパク質を検出する;
(4)任意的に、蛍光顕微鏡撮影する。

(2)これに適当な緩衝液(例えば100mMのMES-Na緩衝液(pH5.5))中のビオチンヒドラジドを室温にて数時間(例えば1時間)反応させる;
(3)反応終了後に適当な生理溶液(例えば緩衝液リン酸緩衝生理食塩液(PBS))にて十分に洗浄した後、蛍光標識アビジンを室温にて数時間(例えば1時間)反応させる;
(4)蛍光顕微鏡にて酸化タンパク質を検出する;
(5)任意的に、蛍光顕微鏡撮影する。
(1)角層試料を、例えばテープストリッピングにより、採取する;
(2)これに適当な緩衝液(例えば100mMのMES-Na緩衝液(pH5.5)中のジニトロフェニルヒドラジン(DNPH)を室温にて数時間(例えば1時間)反応させる;
(3)反応終了後に適当な生理溶液(例えば緩衝液リン酸緩衝生理食塩液(PBS))にて十分に洗浄した後、抗DNP抗体、例えばウサギDNP抗体(ZYMED社製)の同生理溶液を室温にて数時間(例えば1時間)反応させる;
(4)反応終了後に同生理溶液にて十分に洗浄し、上記抗DNP抗体に特異的な蛍光標識二次抗体、例えばフルオレセイン標識抗ウサギIg(アマシャムファルマシアバイオテク社製)等を室温にて数時間(例えば1時間)反応させる;
(5)蛍光顕微鏡にて酸化タンパク質を検出する;
(6)任意的に、蛍光顕微鏡撮影する。
乾燥豚皮(アロアスク、大鵬薬品工業株式会社)を水に3日間浸漬した。豚皮を取り出し、0、1、10、100mMのアクロレイン(東京化成工業株式会社)溶液に3時間浸漬して酸化処理した。アクロレインはタンパク質に作用することで酸化させ、アルデヒドタンパク質付加体を生成する。酸化処理後、水で1時間洗浄した。洗浄後、角層側を上に向けてシャーレ上で乾燥(湿度50%、温度25℃)を開始し、15分後の角層水分量をスキコン200(アイ・ビイ・エス株式会社)で測定した。
黒豚皮膚を10mMアクロレイン(東京化成工業株式会社)溶液で1時間、酸化処理した。水で1時間洗浄した後、水分をよく拭き取り、角層水分量の変化をスキコン200によって経時で測定した。
黒豚皮膚を20mMの次亜塩素酸で1時間処理した。水で1時間洗浄した後、水分をよく拭き取り、角層水分量の変化をスキコンによって経時で測定した。
ヒト腕皮膚をPBS又は20mMの次亜塩素酸で1時間、閉塞処理した。水で1時間洗浄後、水分をよく拭き取り、角層水分量の変化をスキコンによって経時で測定した。
乾燥豚皮(アロテスク、大鵬薬品工業株式会社)を水に3日間浸漬した。豚皮を取り出し、0、1、10、100mMのアクロレイン(東京化成工業株式会社)溶液に3時間浸漬して酸化処理した。酸化処理後、水で1時間洗浄した。洗浄後、角層側を上に向けてシャーレ上で乾燥(湿度50%、温度25℃)を開始し、24時間かけて乾燥させた豚皮の外観を目視観察した。

- 角層中の酸化タンパク質を指標とする、角層の透明性及び/又は保水性を評価するための方法。
- 皮膚から採取した角層試料中の酸化タンパク質のカルボニル基を特異的に蛍光標識し、その蛍光を検出することで角層酸化タンパク質の検出を行う、請求項1記載の角層の透明性及び/又は保水性を評価するための方法。
- 角層の透明性及び/又は保水性を指標とする、角層中のタンパク質の酸化度を検出するための方法。
- 角層中のタンパク質の酸化を抑制することにより、角層の透明性及び/又は保水性を保持・向上させる方法。
