Abstract
【解決手段】 微小流路アレイ6に、回収対象である生体分子7含む試料溶液8を送液し、該生体分子7を、微小流路の壁面にスポット状に固定されており、かつ生体分子7との高い親和性を有するプローブ分子と結合させた後、周期的な突起物を有する第2の基板を微小流路アレイ6から剥がす。微小流路アレイ6にある第1の基板の回収対象である生体分子7を結合しているプローブ分子が固定されているスポットが含まれる部位を、鋭利な切出刃等を用いて切出し、生体分子7を結合したプローブ分子を表面に保持している小片を得る。

Description
図1〜図3は、多種類のプローブ分子を固定したスポットと周期的な突起物を併せ持つ流路構造物の製作方法を示す模式図であり、図4は、多種類のプローブ分子を固定したスポットと周期的な突起物を併せ持つ微小流路アレイの模式図である。
Claims (5)
- 軟質材質でできた第1の基板上に微小流路が設けられ、該微小流路の下面及び/又は壁面に多種類のプローブ分子が所望の配列で化学結合によりスポット状に固定された微小流路アレイ部品。
- 軟質樹脂材質でできた第1の基板上に微小流路が設けられ、該微小流路の下面及び/又は壁面に多種類のプローブ分子が所望の配列で化学結合によりスポット状に固定された微小流路アレイ部品と、該微小流路アレイ部品の微小流路にはめ込むことが可能な大きさを有する周期的な突起物を有する第2の基板を張り合わせて製作された、該微小流路に多種類のプローブ分子が固定されたスポットと周期的な突起物を併せ持つ生体分子回収用微小流路アレイ。
- 請求項2に記載の、多種類の生体分子を固定したスポットと周期的な突起物を併せ持つ微小流路アレイの微小流路に、回収対象である生体分子を含む試料溶液を送液する工程と、該試料溶液に含まれる回収対象である生体分子を、微小流路の下面及び/又は壁面にスポット状に固定され、かつ該生体分子との高い親和性を有するプローブ分子と迅速に結合させる工程とを含むことを特徴とする生体分子の回収方法。
- 請求項3に記載の生体分子の回収方法において、該試料溶液に含まれる回収対象である生体分子を、微小流路の下面及び/又は壁面にスポット状に固定され、かつ該生体分子との高い親和性を有するプローブ分子と迅速に結合させる工程の後、周期的な突起物を有する基板を微小流路アレイ部品から剥がし、該微小流路アレイ部品の微小流路の、回収対象である生体分子を結合しているプローブ分子が固定されている部位を、鋭利な工具を用いて切出す工程とを含むことを特徴とする生体分子の回収方法。
- 請求項4に記載の生体分子の回収方法において、該微小流路アレイ部品の微小流路の、回収対象である生体分子を結合しているプローブ分子が固定されている部位を、鋭利な工具を用いて切出す工程の後、切出した基板の小片に化学的な処理を施し、小片の表面に固定されたプローブ分子と回収対象である生体分子の結合を切断することにより、回収対象である生体分子を回収する工程を含むことを特徴とする生体分子の回収方法。
