Abstract
【解決手段】 3−エチニルチオフェン類を酸触媒の存在下で水和して3−アセチルチオフェン類を製造する。原料3−エチニルチオフェン類は、3−ハロゲノチオフェン類とアセチレン化合物のカップリング反応によって得られるチエニルアセチレン化合物を脱保護することによって得ることができる。

Description
チエニルアセチレン化合物は、3−ハロゲノチオフェン類とアセチレン化合物のカップリング反応によって得ることができる。この反応は既知の方法により、あるいはそれに準じて行なうことができる。かかる方法の代表例としては、3−ハロゲノチオフェンと2−メチルー3−ブチン−2−オールとを、パラジウム触媒、銅塩及び塩基の存在下に反応させる方法(Synthesis、1996年、p.589やJ.Chem.Soc.Perkin Trans.1、1998年、p.4141)を挙げることができるが、これらに限定されるものではない。
本発明における3−アセチルチオフェン類の製造の原料となる3−エチニルチオフェン類は、上記方法によって得られるチエニルアセチレン化合物をアルカリ触媒下で脱保護することによって得ることができる。この脱保護反応は、前記したSynthesis、1996年、p.589やJ.Chem.Soc.Perkin Trans.1、1998年、p.4141に記載されているような既知の方法によって行なうことができるが、かかる方法のみに限定されるものではない。
本発明においては、上記方法によって製造可能な3−エチニルチオフェン類の水和反応によって3−アセチルチオフェン類を製造するものである。一般にアルキンの水和反応においては、酸と水銀塩を触媒に使用することが多いが、本発明の反応においては毒性の強い水銀塩を使用することなく酸触媒のみを用いて、高収率で3−アセチルチオフェン類を製造することが可能である。酸触媒としては硫酸の使用が好ましく、例えば1〜95%程度の水溶液として使用することができる。酸触媒の使用量は、3−エチニルチオフェン類1モルに対して0.01〜20モル、とくに0.1〜10モルの範囲とするのが好ましい。
2−メチル−4−(3’−チエニル)−3−ブチン−2−オールの合成
攪拌機、温度計及び還流冷却器を備えた100mlガラス製フラスコに、3−ブロモチオフェン3.00g(18.4ミリモル)、炭酸カリウム6.36g(46.0ミリモル)、ジメトキシエタン30ml及び水30mlを仕込み、窒素ガスをバブリングしながら、この混合液に、トリフェニルホスフィン386mg(1.47ミリモル)、含水10%パラジウム/炭素783mg(0.37ミリモル)、沃化銅(I)140mg(0.74ミリモル)を加えて、窒素雰囲気下、35℃で30分間攪拌した。次いで80℃に昇温し、同温度で2−メチル−3−ブチン−2−オール3.87g(46.0ミリモル)を30分かけて滴下し、滴下終了後、80℃で15時間攪拌した。反応終了後、20℃まで冷却し、不溶物を濾過した後、濾液に酢酸エチル及び氷水を加えて分液した。酢酸エチル層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥した後、酢酸エチル溶液を濃縮して、2−メチル−4−(3’−チエニル)−3−ブチン−2−オールの薄黄色固体3.5gを得た。
3−エチニルチオフェンの合成(1)
温度計を備えた50mlガラス製フラスコに、窒素雰囲気下で、参考例1で得られた2−メチル−4−(3’−チエニル)−3−ブチン−2−オール2.0g(12.0ミリモル)、粉砕した水酸化ナトリウム0.11g(2.65ミリモル)及び流動パラフィン2.0mlを仕込み、減圧単蒸留できるように、留出管、冷却器及び受器を装着した。次に減圧下でオイルバスにより加熱し、生成した3−エチニルチオフェンとアセトンを留出させながら反応を行なった。オイルバス温度を130℃に保って1時間反応を継続した時点で、留出が認められなくなり、反応を終了させた。
3−アセチルチオフェンの合成(1)
攪拌機、温度計及び還流冷却器を備えた50mlガラス製フラスコに、参考例2で得られた3−エチニルチオフェン1.0g(8.91ミリモル)、30%の硫酸水溶液3.5ml及びイソプロパノール5mlを仕込み、80℃で1時間攪拌した。反応終了後、20℃まで冷却し、飽和重曹水を加えて中和し、酢酸エチル30mlを加えて、抽出、分液した。酢酸エチル層を飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥した後に、得られた酢酸エチル溶液を濃縮して、3−アセチルチオフェンの薄黄色結晶1.10gを得た。さらに酢酸エチルとヘプタンの混合溶媒(1/5)で再結晶を行ない、白色の3−アセチルチオフェン0.96gを得た。高速液体クロマトグラフィを用いて分析したところ、純度は99.7%であり、3−エチニルチオフェンからの収率は、85.1モル%であった。
3−エチニルチオフェンの合成(2)
攪拌器、温度計及びディーンスターク(Dean−Stark)トラップを装着した100mlガラスフラスコに、粉砕した水酸化ナトリウム0.35g(8.75ミリモル)トルエン50ml及び参考例1と同様に調製した2−メチル−4−(3’−チエニル)−3−ブチン−2−オール6.0gを仕込んだ。ディーンスタークトラップには、数mlの水を入れておき、副生するアセトンを吸収させるようにして、前記混合物をトルエン還流下に1時間反応させた。反応終了後、20℃まで冷却して濾過により水酸化ナトリウムを除去し、高速液体クロマトグラフィによる純度94.4%の3−エチニルチオフェンを含むトルエン溶液を得た。この溶液を実施例2の原料として使用した。
3−アセチルチオフェンの合成(2)
攪拌機、温度計及び還流冷却器を備えた50mlガラス製フラスコに、参考例3で得られた3−エチニルチオフェンを含むトルエン溶液及び30%の硫酸水溶液3.5mlを仕込み、80℃で1時間攪拌した。反応終了後、20℃まで冷却し、飽和重曹水を加えて中和後、分液した。トルエン層を飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥した後、得られたトルエン溶液を濃縮して、3−アセチルチオフェンの薄黄色結晶3.92gを得た。さらに酢酸エチルとヘプタンの混合溶媒(1/5)で再結晶を行ない、淡黄色の3−アセチルチオフェン3.32gを得た。高速液体クロマトグラフィを用いて分析したところ、純度は97.2%であり、2−メチル−4−(3’−チエニル)−3−ブチン−2−オールからの収率は、70.9モル%であった。
