本発明に関連して、本発明者らは、正常被験者およびリウマチ性関節炎(RA)患者の滑膜における肥満細胞サブセットの分布およびその密度を調べた。正常な滑膜では、肥満細胞の大多数がMCTCサブセットに属し、MCT細胞の数を5:1の比率で上回った。肥満細胞の平均密度は、正常滑膜と比較してRA滑膜では有意に増加した。RAでは、双方のサブセットが増加して、これらは浸潤性炎症細胞または高度に細胞性の繊維様組織領域(主にMCTC)に関連した。さらに、リウマチ様疾患の活動性または進行の臨床パラメータとMCTC細胞の密度、特に、滑膜表層の密度とのあいだに相関を認めた。

治療プロトコール
関節炎を誘導する前に、マウスに様々な濃度の化合物を2日間(第2日、第1日)前処置した。関節炎は、0日目および2日目に血清150μlの腹腔内注射によって誘導した。異なる濃度の化合物による処置は、疾患の経過のあいだ継続した。対照マウスには、関節炎の誘導前および疾患の経過のあいだ酸PBSを注射した。足首の厚みおよび関節炎スコアを12日間評価した。関節炎スコア:それぞれの四肢のスコアの総和(0、疾患なし;0.5、足または指数本の軽度の腫脹;1、明確な関節の炎症;最大スコア=4)。結果を図1および2に示す。表1は、本試験において用いたマウスの数を示す。

- チロシンキナーゼ阻害剤を、そのような治療を必要とする哺乳類に投与する段階を含む、炎症性疾患を治療する方法。
- チロシンキナーゼ阻害剤が、IL-3の存在下で培養したIL-3依存的細胞の死を促進することができない、請求項1記載の方法。
- c-kit阻害剤を、そのような治療を必要とする哺乳類に投与する段階を含む、炎症性疾患を治療する方法。
- c-kit阻害剤が非毒性で選択的かつ強力なc-kit阻害剤である、請求項3記載の方法。
- 阻害剤が、インドリノン、ピリミジン誘導体、ピロロピリミジン誘導体、キナゾリン誘導体、キノキサリン誘導体、ピラゾール誘導体、ビス単環式、二環式、または複素環式アリール化合物、ビニレン-アザインドール誘導体およびピリジル-キノロン誘導体、スチリル化合物、スチリル置換ピリジル化合物、セレオインドール、セレニド、三環式ポリヒドロキシル化合物、ならびにベンジルホスホン酸化合物からなる群より選択される、請求項4記載の方法。
- 阻害剤が以下からなる群より選択される、請求項4記載の方法:
‐ピリミジン誘導体、より詳細にはN-フェニル-2-ピリミジン-アミン誘導体、
‐インドリノン誘導体、より詳細にはピロール置換インドリノン、
‐単環式、二環式アリール、およびヘテロアリール化合物、
‐ならびにキナゾリン誘導体。 - 阻害剤が、4-(4-メチルピペラジン-1-イルメチル)-N-[4-メチル-3-(4-ピリジン-3-イル)ピリミジン-2-イルアミノ)フェニル]ベンズアミドである、請求項7記載の方法。
- c-kit阻害剤がIL-3の存在下で培養したIL-3依存的細胞の死を促進することができない、請求項3から8のいずれか一項に記載の方法。
- c-kit阻害剤が活性化c-kitの阻害剤である、請求項3から9のいずれか一項に記載の方法。
- 活性化c-kit阻害剤がSCF活性化c-kitを阻害することができる、請求項10記載の方法。
- 阻害剤が構成的に活性化された変異体c-kitを阻害することができる、請求項10記載の方法。
- 以下を含むスクリーニング法によって得ることができる、選択的で強力かつ非毒性の活性化c-kit阻害剤である化合物を、そのような治療を必要とする哺乳類に投与する段階を含む、炎症性疾患を治療する方法:
a)(i)活性化c-kit、および(ii)試験する少なくとも一つの化合物を、成分(i)と成分(ii)とが複合体を形成することができる条件下で接触させる段階、
b)活性化c-kitを阻害する化合物を選択する段階、
c)IL-3の存在下で培養したIL-3依存的細胞の死を促進することができない、段階b)において同定された化合物のサブセットを試験して選択する段階。 - スクリーニング法が、SCF活性化野生型c-kitを同様に阻害することができる変異体活性化c-kitの阻害剤である、段階b)において同定された化合物のサブセットを試験して選択することからなる段階をさらに含む、請求項13記載の方法。
- 活性化c-kitが、段階a)のSCF活性化野生型c-kitである、請求項13記載の方法。
- 推定の阻害剤が、段階a)において10μMより高い濃度で試験される、請求項13から15のいずれか一項に記載の方法。

a)永続的に活性化されたc-kitである変異体c-kit(例えば、トランスホスホリラーゼドメインにおける変異)を発現する細胞を用いて、細胞死の程度を測定することによって、それぞれがIC50<10μMを有する、活性化c-kitを標的とする候補化合物のサブセットを同定するために、複数の被験化合物について増殖アッセイを行う段階、
b)c-kitを特異的に標的とする候補化合物のサブセットを同定するために、IL-3の存在下で培養したIL-3依存的細胞である野生型c-kitを発現する細胞を用いて、段階(a)において同定された候補化合物のサブセットについて増殖アッセイを行う段階、
