Claims (1)
- 【特許請求の範囲】 1.インフルエンザ菌(Haemophilus influenzae)(H.influenzae)のNucAタ ンパク質またはそのエピトープ(1個もしくは複数)を含むNucAタンパク質 のペプチドをコードするDNA配列を含んで成る、単離されかつ精製されたDN A配列。 2.標準的な高緊縮サザンハイブリダイゼーション条件下で、配列番号2のアミ ノ酸26〜603のアミノ酸配列もしくはその生物学的に同等なアミノ酸配列を有す るインフルエンザ菌(H.influenzae)のNucAタンパク質をコードするDNA 配列とハイブリダイゼーションするDNA配列を含んで成る、単離されかつ精製 されたDNA配列。 3.前記DNA配列が、標準的な高緊縮サザンハイブリダイゼーション条件下で 、配列番号1のヌクレオチド304〜2037のヌクレオチド配列を有するDNA配列 とハイブリダイゼーションする、請求の範囲2の単離されかつ精製されたDNA 配列。 4.DNA配列が、標準的な高緊縮サザンハイブリダイゼーション条件下で、イ ンフルエンザ菌(H.influenzae)のNucAタンパク質についての生物学的に同 等なアミノ酸配列をコードするDNA配列とハイブリダイゼーションする、請求 の範囲2の単離されかつ精製されたDNA配列であって、配列番号2のアミノ酸 26〜603のアミノ酸配列が、K79E、N186K、S262G、V294A、 E305Q、K327R、T337A、D360Y、R376HもしくはV43 6Iから成る群から選択されるアミノ酸残基の変化の1個もしくはそれ以上によ り改変される配列。 5.請求の範囲1のDNA配列を含んで成る、単離されかつ精製された、 インフルエンザ菌(H.influenzae)のDNA配列を含有するプラスミド。 6.プラスミドが、標準的な高緊縮サザンハイブリダイゼーション条件下で、配 列番号2のアミノ酸26〜603のアミノ酸配列もしくはその生物学的に同等なアミ ノ酸配列を有するインフルエンザ菌(H.influenzae)のNucAタンパク質をコ ードするDNA配列とハイブリダイゼーションするDNA配列を含有する、請求 の範囲5のプラスミド。 7.プラスミドが、標準的な高緊縮サザンハイブリダイゼーション条件下で、配 列番号1のヌクレオチド304〜2037のヌクレオチド配列を有するDNA配列とハ イブリダイゼーションするDNA配列を含有する、請求の範囲6のプラスミド。 8.プラスミドが、標準的な高緊縮サザンハイブリダイゼーション条件下で、イ ンフルエンザ菌(H.influenzae)のNucAタンパク質について生物学的に同等 なアミノ酸配列をコードするDNA配列とハイブリダイゼーションするDNA配 列を含有する、請求の範囲6のプラスミドであって、配列番号2のアミノ酸26〜 603のアミノ酸配列が、K79E、N186K、S262G、V294A、E3 05Q、K327R、T337A、D360Y、R376HもしくはV436I から成る群から選択されるアミノ酸残基の変化の1個もしくはそれ以上により改 変されるプラスミド。 9.プラスミドがpPX691と称されるものである、請求の範囲7のプラスミ ド。 10.請求の範囲5のプラスミドで形質転換された宿主細胞。 11.宿主細胞が大腸菌(Escherichia coli)株InvaF’である、請求の範囲 10の宿主細胞。 12.プラスミドがpPX691(ATCC 98104)と称されるものであ る、請求の範囲11の宿主細胞。 13.宿主細胞を請求の範囲5のプラスミドで形質転換もしくはトランスフェク ションすること、および、宿主細胞による前記NucAタンパク質の発現を許す 条件下で宿主細胞を培養することを含んで成る、インフルエンザ菌(H.influenz ae)のNucAタンパク質の産生方法。 14.単離されかつ精製された、インフルエンザ菌(H.influenzae)のNucA タンパク質、または、そのエピトープ(1個もしくは複数)を含んで成るNuc Aタンパク質のペプチド。 15.配列番号2のアミノ酸26〜603のアミノ酸配列もしくはその生物学的に同 等なアミノ酸配列を有する、請求の範囲14の単離されかつ精製された、インフ ルエンザ菌(H.influenzae)のNucAタンパク質。 16.(a)12%SDS−PAGEゲル上で測定されるようなおよそ63,000ダル トンの分子量;および (b)5’−ヌクレオチダーゼ活性 を有する、請求の範囲14の単離されかつ精製された、インフルエンザ菌(H.in fluenzae)のNucAタンパク質。 17.インフルエンザ菌(H.influenzae)のNucAタンパク質について生物学 的に同等なアミノ酸配列を有する、請求の範囲15の単離されかつ精製されたN ucAタンパク質であって、配列番号2のアミノ酸26〜603のアミノ酸配列が、 K79E、N186K、S262G、V294A、E305Q、K327R、T 337A、D360Y、R376HもしくはV436Iから成る群から選択され るアミノ酸残基の変化の1個もしくはそれ以上により改変されるタンパク質。 18.単離されかつ精製された、インフルエンザ菌(H.influenzae)のNucA タンパク質またはそのエピトープ(1個もしくは複数)を含んで成るNucAタ ンパク質のペプチドを含んで成るワクチン組成物であって、前記ワクチン組成物 が噛乳動物宿主で防御免疫応答を導き出す組成物。 19.NucAタンパク質が、配列番号2のアミノ酸26〜603のアミノ酸配列も しくはその生物学的に同等なアミノ酸配列を有する、請求の範囲18のワクチン 組成物。 20.NucAタンパク質が、 (a)12%SDS−PAGEゲル上で測定されるようなおよそ63,000ダルトンの 分子量;および (b)5’−ヌクレオチダーゼ活性 を有する、請求の範囲18のワクチン組成物。 21.NucAタンパク質が、インフルエンザ菌(H.influenzae)のNucAタ ンパク質について生物学的に同等なアミノ酸配列を有する、請求の範囲19のワ クチン組成物であって、配列番号2のアミノ酸26〜603のアミノ酸配列が、K7 9E、N186K、S262G、V294A、E305Q、K327R、T33 7A、D360Y、R376HもしくはV436Iから成る群から選択されるア ミノ酸残基の変化の1個もしくはそれ以上により改変される組成物。 22.アジュバント、希釈剤もしくは担体をさらに含んで成る、請求の範囲18 のワクチン組成物。 23.アジュバントが、水酸化アルミニウム、リン酸アルミニウム、QS−21 、3−O−デアシル化モノホスホリルリピドAおよびIL−1 2から成る群から選択される、請求の範囲22のワクチン組成物。 24.請求の範囲18のワクチン組成物の免疫原性の量を哺乳動物宿主に投与す ることを含んで成る、インフルエンザ菌(H.influenzae)に対する免疫方法。 25.単離されかつ精製された、インフルエンザ菌(H.influenzae)のNucA タンパク質またはそのエピトープ(1個もしくは複数)を含んで成るNucAタ ンパク質のペプチドを、ワクチン組成物が哺乳動物宿主で防御免疫応答を導き出 すように十分な量で包含することを含んで成る、前記ワクチン組成物の調製方法 。
