Claims (1)
- 【特許請求の範囲】 1.酵素活性の特異的阻害剤の存在および不在下でサンプル中の酵素活性を測 定することからなる、サンプル中の酵素活性を測定するアッセイの感度および特 異性を増強する方法。 2.以下の工程: (a)カルボキシペプチダーゼA特異的阻害剤の存在および不在下でカルボキ シペプチダーゼA基質に生物学上の流体を接触させ;そして (b)カルボキシペプチダーゼA特異的阻害剤の存在および不在下で生物学上 の流体中のカルボキシペプチダーゼAによりカルボキシペプチダーゼA基質の加 水分解によりもたらされる光学密度の変化を測定すること からなる、生物学上の流体中のカルボキシペプチダーゼAレベルを測定する方法 。 3.以下の工程: (a)カルボキシペプチダーゼA特異的阻害剤の存在および不在下で生物学上 の流体中のあらゆるカルボキシペプチダーゼAによるカルボキシペプチダーゼA 基質の加水分解によりもたらされる光学密度の変化を検出することにより患者か らの生物学上の流体中のカルボキシペプチダーゼAレベルを測定し;そして (b)患者の生物学上の流体中のカルボキシペプチダーゼAの測定されたレベ ルが、健康な対照集団からの生物学上の流体中のレベル以上に上昇したか否かを 測定すること からなる、急性膵臓炎を罹患する疑いのある患者における急性膵臓炎の診断方法 。 4.以下の工程: (a)クロストリパインの添加により生物学上の流体中のプロカルボキシペプ チダーゼAをカルボキシペプチダーゼAに変換し; (b)カルボキシペプチダーゼA特異的阻害剤の存在および不在下でカルボキ シペプチダーゼA基質に生物学上の流体を接触させ;そして (c)カルボキシペプチダーゼA特異的阻害剤の存在および不在下で生物学上 の流体中のカルボキシペプチダーゼAによるカルボキシペプチダーゼA基質の加 水分解によりもたらされる光学密度の変化を測定すること からなる、生物学上の流体中の全カルボキシペプチダーゼAレベルを測定する方 法。 5.以下の工程: (a)クロストリパインの添加により、患者から得られた生物学上の流体中の プロカルボキシペプチダーゼAをカルボキシペプチダーゼAに変換し; (b)カルボキシペプチダーゼA特異的阻害剤の存在および不在下で生物学上 の流体中のカルボキシペプチダーゼAによるカルボキシペプチダーゼA基質の加 水分解によりもたらされる光学密度の変化により検出される生物学上の流体中の 全カルボキシペプチダーゼAレベルを測定し; (c)患者の生物学上の流体中の測定された全カルボキシペプチダーゼAレベ ルが、生物学上の流体中の上昇したプロカルボキシペプチダーゼAのために、健 康な集団中の全カルボキシペプチダーゼAレベルに比較して増加したか否かを決 定すること からなる、患者中の初期段階の膵臓癌を診断する方法。
