Claims (1)
- 【特許請求の範囲】 1.下記の段階を含む、蛋白質の第1の非病原性構造および蛋白質の第2の病原性 構造を含む試料において選択された蛋白質の病原性型の存在を決定する方法: 試料の第1の部分を、第2の構造よりも第1の構造に高い親和性を持つ結合パー トナーに接触させて、第1の濃度を決定する段階; 試料の第2の部分を、結合パートナーに対する第2の構造の結合親和性を上昇さ せるように処理する段階; 試料の処理された第2の部分を結合パートナーと接触させて、第2の濃度を決定 する段階; 調整濃度を提供するために、第1の構造の蛋白質の結合パートナーに対する親 和性の処理による増加分を補正するように、第2の濃度を調整する段階;および 第1の濃度を調整濃度と比較して、第2の病原性構造を取る蛋白質の存在を決定 する段階。 2.第1の濃度および第2の濃度が時間分解、解離増強蛍光法で決定され; さらに蛋白質の第2の病原性構造が1x103粒子/ml以下の濃度で試料中に存在し; さらに該蛋白質が、βA4蛋白質、PrP蛋白質、およびトランスチレチンからなる 群より選択される、 疾病の症状を示していない動物から試料を得る、請求項1記載の方法。 3.蛋白質が固体支持体に結合され、 処理が、第2の型の任意の蛋白質の少なくとも2%を結合型に転換するために十分 な、熱、圧力、および化学的変性からなる群より選択される処理を試料に加える ことを含み; 結合パートナーが、第2の型に対する親和性よりも第1の型に対して少なくとも 10倍高い親和性を持つ標識抗体を含む、請求項1記載の方法。 4.下記の段階を含む、結合パートナーに対する結合親和性の異なる第1の構造 および第2の構造で存在する、選択された蛋白質の病原性型の存在を、非病原性 構造を取る蛋白質の存在下で決定する方法: 蛋白質の第2の構造を、結合パートナーに対して第2の構造よりも高い親和性を 持つ結合型に転換するために試料を処理する段階; 濃度を決定するために、処理した試料を結合パートナーに接触させる段階; 調整濃度を提供するために、蛋白質の第1の構造の結合パートナーに対する親 和性の処理による増加分を補正するように、濃度を調整する段階; 該調整第1濃度を、対照濃度および事前に決定された基準濃度からなる群より 選択される既知の濃度と比較して、試料中の第2の構造の蛋白質の存在を決定す る段階。 5.結合パートナーが標識抗体を含み、濃度がフローサイトメトリーを用いて決 定され; 調整濃度が、処理済み非感染対照試料から決定された既知の濃度または非感染 群の処理済み試料からあらかじめ決定された既知の濃度と比較され; さらに抗体が3F4であり; さらに第2の構造の蛋白質が、1mlあたり1x103蛋白質分子以下の濃度で試料中 に存在し、且つ第1の構造の蛋白質が、1mlあたり1x106蛋白質分子以上の濃度で 試料中に存在する、請求項4記載の方法。 6.下記の段階を含む、立体構造が異なる第1の型と第2の型とを有し該型のうち の1つがプリオンが仲介する疾病と関連している蛋白質により特徴づけられる、 プリオンが仲介する疾病に対する治療活性を有する化合物の同定方法: プリオンが仲介する疾病に感受性の動物を提供する段階; 試験化合物を該動物に投与する段階; 該プリオンが仲介する疾病を誘導する段階; 該動物から試料を得る段階; 該蛋白質を含む試料の第1の部分を、該第2の型よりも該蛋白質の該第1の型に 対する親和性が高い結合パートナーに接触させ、第1の濃度を決定する段階; 該第2の型を該結合パートナーに対する親和性のより高い結合型に転換させる ために、試料の第2の部分を処理する段階; 第2の濃度を決定するために、処理済みの第2の部分を結合パートナーに接触さ せる段階; 調整濃度を提供するために、第1の型の蛋白質の結合パートナーに対する親和 性の処理による増加分を補正するように、第2の濃度を調整する段階;および 第1の濃度を調整濃度と比較して、第2の型の蛋白質の構造を決定し、第2の型 の蛋白質濃度に対する試験化合物の効果を推定する段階。 7.下記の段階を含む、試料中の病原性蛋白質の株を決定する方法: 試料中の蛋白質の病原性構造の濃度を決定する段階; 蛋白質の病原性構造を、結合パートナーに対する結合親和性が病原性構造より も高い構造に変換するように、試料を処理する段階; 病原性構造に対する蛋白質処理の効果を決定する段階;および 該決定された効果を、既知の濃度の既知の株についての既知の基準効果と比較 し、それによって試料中の蛋白質の病原性構造の株を推定する段階。 8.第2の型の蛋白質濃度を低下させることにより特徴付けられる、請求項6およ び7のいずれか一項記載の方法によって同定される化合物。 9.下記の段階を含む、試料中の病原性蛋白質の株を決定する方法: 試料中の蛋白質の病原性構造の濃度を決定する段階; 蛋白質の病原性構造を、結合パートナーに対する結合親和性が病原性構造より も高い構造に変換するように、試料を処理する段階; 病原性構造に対する蛋白質処理の効果を決定する段階;および 該決定された効果を、既知の濃度の既知の株についての既知の基準効果と比較 し、それによって試料中の蛋白質の病原性構造の株を推定する段階。 10.病原性構造の濃度および処理による効果が、結合パートナーとしての標識抗 体を用い、時間分解、解離増強蛍光法を用いて決定され; 蛋白質の病原性構造がPrPScで、PrPScがプロテイナーゼKで処理される、請求 項9記載の方法。
