Abstract

<B>MéTODO PARA DETERMINAR A PRESENçA DE UMA FORMA PATOGêNICA DE UMA PROTEìNA SELECIONADA NUMA AMOSTRA"<D>. Um método de ensaio é revelado que torna possível determinar a presença de conformação, relacionada com doença, de uma proteína (por exemplo, PrP^ Sc^ ou a forma de folha <225> de uma <225>A4) em uma amostra. Uma amostra é dividida em duas porções e a primeira porção é presa transversalmente a um primeiro apoio sólido e então contactada com um anticorpo rotulado que se aglutina a uma forma de não-doença da proteína com um grau mais elevado de afinidade (por exemplo, 4 para 30 vezes mais elevada) que a forma de doença da proteína, para mudar a conformação em uma forma com uma afinidade aglutinadora mais elevada para anticorpo rotulado. A segunda porção tratada é então aglutinada a um segundo apoio sólido e contactada com anticorpo rotulado. O nível do anticorpo rotulado que se aglutina a uma proteína nas primeira e segunda porção é determinado e as quantidades medidas em cada uma delas são comparadas. A diferença entre as duas medições é uma indicação de se a conformação relacionada com doença da proteína estava presente na amostra. O método pode também determinar a concentração da conformação relacionada com a doença e a cepa particular presente.
