Claims (1)
- 【特許請求の範囲】 1.ヘリコバクター菌(Helicobacter)抗原をコードする少なくとも1つの異種核 酸分子を含んでなる組換え弱毒化微生物病原体であって、標的細胞中で該核酸分 子を発現することができる、または該核酸分子の発現を引き起こすことができる 該病原体。 2.腸内細菌細胞、特にサルモネラ菌(Salmonella)細胞である、請求項1に記載 の病原体。 3.サルモネラ・アロ(Salmonella aro)突然変異細胞である、請求項1または2 に記載の病原体。 4.ヘリコバクター菌抗原がウレアーゼ、ウレアーゼサブユニット、その免疫学 的に反応性の断片、またはそのペプチドミモトープ(mimotope)である、請求項1 から3のいずれかに記載の病原体。 5.ヘリコバクター菌抗原がウレアーゼ、ヘリコバクター菌由来の分泌ポリペプ チド、その免疫学的に反応性の断片、またはそのペプチドミモトープである、請 求項1から3のいずれかに記載の病原体。 6.ヘリコバクター菌抗原が抗原AlpA、AlpB、その免疫学的に反応性の断片、ま たはそのペプチドミモトープからなる群から選択される、請求項1から3および 5のいずれかに記載の病原体。 7.ヘリコバクター菌抗原をコードする該核酸分子が相変異的に(phase variabl y)に発現されうる、請求項1から6のいずれかに記載の病原体。 8.ヘリコバクター菌抗原をコードする該核酸分子が、該病原体中で実質的に不 活性であり、かつ該病原体中の核酸再編成(reorganization)機構によって引き起 こされる核酸再編成によって活性化されうる発現シグナルの制御下にある、請求 項7に記載の病原体。 9.該発現シグナルがバクテリオファージプロモーターであり、そして該活性化 が該病原体中における対応するバクテリオファージRNAポリメラーゼの産生を もたらすDNA再編成によって引き起こされる、請求項8に記載の病原体。 10.免疫変調性(immunomodulatory)ポリペプチドをコードする少なくとも1つの 第2核酸分子をさらに含む請求項1から9のいずれかに記載の病原体であって、 該第2核酸分子を発現することができる該病原体。 11.請求項1から10のいずれかに記載の病原体を活性成分として含み、さらに場 合により薬学上許容される希釈剤、担体およびアジュバントを含む、医薬組成物 。 12.粘膜表面への投与、または非経口投与に適する生ワクチンである、請求項11 に記載の組成物。 13.薬学上有効量の請求項1から10のいずれかに記載の弱毒化病原体を薬学上許 容される希釈剤、担体および/またはアジュバントと共に製剤化することを含む 、生ワクチンの製造方法。 請求項1に記載の方法。 14.請求項1から10のいずれかに記載の組換え弱毒化病原体の製造方法であって 、下記の工程を含む該方法: a)ヘリコバクター菌抗原をコードする核酸分子を弱毒化病原体に挿入して、標 的細胞中で該核酸分子を発現することができる、または該核酸分子の発現を引き 起こすことができる組換え弱毒化病原体を獲得し;そして b)適切な条件下で該組換え弱毒化病原体を培養する。 15.ヘリコバクター菌抗原をコードする該核酸分子を染色体外プラスミド上に位 置させる、または染色体中に挿入する、請求項14に記載の方法。 16.哺乳動物宿主中に防御免疫応答を引き起こすヘリコバクター菌抗原を同定す る方法であって、下記の工程を含む方法: a)弱毒化病原体中にヘリコバクター菌の発現遺伝子ライブラリーを供給し、そ して; b)哺乳動物宿主にヘリコバクター感染に対する防御免疫を付与する能力につい て該遺伝子ライブラリーのクローンをスクリーニングする。
