Claims (1)
- 【特許請求の範囲】 1.配列番号4のアミノ酸配列を有する哺乳類B細胞を活性化できるポリペプチ ド、または前記ポリペプチドの生物活性を保持する前記ポリペプチドの断片、ま たは前記ポリペプチドの生物活性を保持する前記ポリペプチドのバリアント、ポ リペプチドの保存的に置換されたバリアント、または前記ポリペプチドの生物活 性を有するその断片またはバリアントの共役結合体。 2.配列番号4を有するポリペプチドまたはその保存的に置換されたバリアント 。 3.ポリペプチドが翻訳時および/または翻訳後修飾されている、請求項1また は2に記載のポリペプチド。 4.請求項1および2に記載のアミノ酸配列を有するポリペプチドをコードする 核酸配列を含む核酸分子。 5.配列番号4のアミノ酸配列を有するポリペプチドをコードする核酸配列を含 む核酸分子。 6.cDNAである、請求項4および5に記載の核酸分子。 7.ゲノムDNA配列である、請求項4および5に記載の核酸分子。 8.配列番号1のヌクレオチド配列を含むcDNA分子である、請求項5に記載 の核酸分子。 9.乳漿からポリペプチドを単離することを含む、配列番号4の配列を有するポ リペプチドまたは翻訳時および/または翻訳後修飾した形の前記タンパク質をウ シ初乳乳漿から調製する方法。 10.ポリペプチドの単離が清澄にしたウシ初乳乳漿からタンパク質を塩析する ことを含む、請求項9に記載の方法。 11.ポリペプチドの別の単離が陰イオン交換クロマトグラフィを含む、請求項 10に記載の方法。 12.ポリペプチドの別の単離が分子ふるいカラムクロマトグラフィを含む、請 求項11に記載の方法。 13.ポリペプチドの別の単離がハイドロキシアパタイトカラムクロマトグラフ ィを含む、請求項12に記載の方法。 14.配列番号4、配列番号5または配列番号6のアミノ酸配列を有するポリペ プチド、または前記ポリペプチドの生物活性を保持する前記ポリペプチドの断片 、または前記ポリペプチドの生物活性を保持する前記ポリペプチドのバリアント 、ポリペプチドの保存的に置換されたバリアント、または前記ポリペプチドの生 物活性を有するその断片またはバリアントの共役結合体を使用してB細胞を活性 化する方法。 15.ポリペプチドの三次元コンフォメーションが保持されているという条件で 、配列番号4、配列番号5または配列番号6のポリペプチドのアミノ酸の置換、 削除または添加により前記ポリペプチドのアナローグを得る、請求項14に記載 の方法。 16.ポリペプチドが配列番号4、配列番号5または配列番号6の保存的に置換 されたバリアントである、請求項15に記載の方法。 17.ポリペプチドが翻訳時および/または翻訳後修飾されている、請求項14 、 15または16に記載の方法。 18.請求項14、15、16または17に記載のポリペプチドを投与すること により、B細胞の活性化を必要としている哺乳類でB細胞を活性化する方法。 19.哺乳類がヒト対象である、請求項18に記載の方法。 20.ヒトが乳児である、請求項19に記載の方法。 21.このような活性化を必要とする哺乳類がCD40陰性またはCD40の不 足したB細胞を有する、請求項18に記載の方法。 22.哺乳類がヒト対象である、請求項21に記載の方法。 23.ヒトが乳児である、請求項22に記載の方法。 24.哺乳類がT細胞免疫不全またはアレルギー患者である、請求項18に記載 の方法。 25.哺乳類がCD40L陰性またはCD40Lの不足したT細胞を有する、請 求項24に記載の方法。 26.哺乳類がヒト対象である、請求項25に記載の方法。 27.ヒトが乳児である、請求項26に記載の方法。 28.配列番号4、配列番号5または配列番号6のアミノ酸配列を有するポリペ プチド、または前記ポリペプチドの生物活性を保持する前記ポリペプチドの断片 、または前記ポリペプチドの生物活性を保持する前記ポリペプチドのバリアント 、 ポリペプチドの保存的に置換されたバリアント、または前記ポリペプチドの生物 活性を有するその断片またはバリアントの共役結合体を使用して、B細胞の成長 と免疫グロブリン高率分泌細胞への分化を誘導する方法。 29.ポリペプチドの三次元コンフォメーションが保持されているという条件で 、配列番号4、配列番号5または配列番号6のポリペプチドのアミノ酸の置換、 削除または添加により前記ポリペプチドのアナローグを得る、請求項28に記載 の方法。 30.ポリペプチドが配列番号4、配列番号5または配列番号6の保存的に置換 されたバリアントである、請求項28に記載の方法。 31.ポリペプチドが翻訳時および/または翻訳後修飾されている、請求項28 、29または30に記載の方法。 32.このような活性化を必要とする哺乳類で、請求項28、29、30または 31に記載のポリペプチドを投与することによりB細胞の成長および分化を誘導 する方法。 33.哺乳類がヒト対象である、請求項32に記載の方法。 34.ヒトが乳児である、請求項31に記載の方法。 35.配列番号4のアミノ酸配列を有する純粋なタンパク質または保存的に置換 されたバリアントを含めた翻訳時および/または翻訳後修飾された形の前記タン パク質を調製する方法であって、 (i)培養条件下で、前記タンパク質または翻訳時および/または翻訳後修飾 された形の前記タンパク質を発現できる原核生物または真核生物宿主を培養する ステップ、 (ii)前記タンパク質または翻訳時および/または翻訳後修飾された形の前 記タンパク質を発現するステップ、および (iii)前記タンパク質を培養物から回収するステップ を含む方法。 36.B細胞活性化を必要とする哺乳類でB細胞を活性化するための医薬品を製 造するための請求項14、15、16または17に記載のポリペプチドの使用。 37.請求項14、15、16または17に記載のポリペプチドをフォーミュラ に混合することを含む、乳児用フォーミュラの製造法。 38.請求項14、15、16または17に記載のポリペプチドをワクチン処方 に混合することを含む、ワクチンの製造法。 39.抗原と請求項14、15、16または17に記載のポリペプチドを結合さ せてワクチン処方と混合することを含む、請求項38に記載の方法。 40.抗原と請求項14、15、16または17に記載のポリペプチドをワクチ ン処方に混合するステップをさらに含む、請求項38に記載の方法。 41.抗原と請求項14、15、16または17に記載のポリペプチドを患者に 投与することを含む、患者にワクチン接種する方法。 42.抗原と請求項14、15、16または17に記載のポリペプチドを1つの ステップで投与する、請求項41に記載の方法。 43.抗原と請求項14、15、16または17に記載のポリペプチドを別々の ステップで投与する、請求項41に記載の方法。 44.所定量の抗原と請求項14、15、16または17に記載の所定量のポリ ペプチドを含む、ワクチン調製用キット。 45.配列番号5のアミノ酸配列を有するポリペプチド、または前記ポリペプチ ドの生物活性を保持する前記ポリペプチドの断片、または前記ポリペプチドの生 物活性を保持する前記ポリペプチドのバリアントに対する抗体を必要とする哺乳 類に、前記ポリペプチドまたはその断片またはバリアントに結合できるポリペプ チドの保存的に置換されたバリアントを投与し、それによって、そうしない場合 に配列番号5のアミノ酸配列を有するポリペプチドまたはその機能的誘導体の血 清濃度が上昇するために過剰に活性化されるヒトB細胞の活性を低下または阻害 する方法。 46.哺乳類がヒト対象である、請求項45に記載の方法。 47.生じさせようとする抗体に対する抗原に合わせて、最適下濃度で、請求項 14、15、16または17に従ってB細胞を活性化することを含む、所望の抗 原特異性を有するモノクローナル抗体を分泌する哺乳類B細胞を増加させる方法 。 48.哺乳類B細胞がヒト起源である、請求項47に記載の方法。 49.哺乳類B細胞がネズミ起源である、請求項47に記載の方法。
