Claims (1)
- 【特許請求の範囲】 1.(a)アッセイしようとする液状検体を受け入れるのに適した検体調製手段 を含む第1の対置成分と; (b)検出しようとする分析対象物に特異的に結合しクロマトグラフィ媒体上 の検出ゾーンに結合された少なくとも1種類の試薬を有する前記クロマトグラフ ィ媒体を含み前記第1の対置成分に取り付けられる第2の対置成分と; (c)前記第2の対置成分に取り付けられた導電バリアとを含み、 前記第1と第2の対置成分をこれらが対置していない位置から対置させて前記 検体調製手段に検査しようとする前記液状検体を前記導電バリア経由で前記クロ マトグラフィ媒体へ添着できるようにしまた前記クロマトグラフィ媒体を通して 流すことができるようにし、クロマトグラフィ・アッセイが前記クロマトグラフ ィ媒体内部での前記検体の移動の結果として行なわれて、検出しようとする分析 対象物に特異的に結合する標識試薬の結合による前記移動の結果として前記分析 対象物が検出され、前記検体が前記クロマトグラフィ媒体に添着された位置とは 異なる位置で前記分析対象物が検出され、前記検出ゾーンに結合した前記分析対 象物に対する前記標識試薬の結合により移動後に前記クロマトグラフィ媒体上で 前記分析対象物が検出されるようにしてあることを特徴とするクロマトグラフィ ・アッセイ装置。 2.前記検体調製手段は前記検体を前記クロマトグラフィ媒体に添着する前に前 記検体の処理用に少なくとも1つの試薬を含むことと、前記検体処理用の前記試 薬は前記検体から分析対象物を抽出する抽出試薬であることを特徴とする請求の 範囲1に記載の装置。 3.前記検出ゾーンは前記クロマトグラフィ媒体より実質的に小さいことと、前 記検出ゾーンはこれに不動化した前記分析対象物に体する第1の特異的結合パー トナーを含むことを特徴とする請求の範囲1に記載のクロマトグラフィ・アッセ イ装置。 4.前記分析対象物は抗原またはハプテンであり、前記第1の特異的結合パート ナーは前記抗原またはハプテンに体する抗体であることを特徴とする請求の範囲 3に記載のクロマトグラフィ・アッセイ装置。 5.前記分析対象物はヒトヘモグロビンであり前記第1の特異的結合パートナー は抗ヒトヘモグロビン抗体であることを特徴とする請求の範囲4に記載のクロマ トグラフィ・アッセイ装置。 6.前記分析対象物は抗体であり前記第1の特異的結合パートナーは前記抗体に より特異的に結合され得るハプテンまたは抗原であることを特徴とする請求の範 囲3に記載のクロマトグラフィ・アッセイ装置。 7.前記クロマトグラフィ媒体はさらに前記クロマトグラフィ媒体より実質的に 小さい対照ゾーンを含むことを特徴とする請求の範囲1に記載のクロマトグラフ ィ・アッセイ装置。 8.前記対照ゾーンはこれに不動化した分析対象物を含むことを特徴とする請求 の範囲7に記載のクロマトグラフィ・アッセイ装置。 9.前記クロマトグラフィ媒体の前記第2の端部と動作的に接触する吸収手段を さらに含むことを特徴とする請求の範囲1に記載のクロマトグラフィ・アッセイ 装置。 10.前記検体調製手段は前記検体調製手段へ液体の添加により再溶解可能な形 で検出可能な標識で標識した前記分析対象物に体する特異的結合パートナーをさ らに含むことを特徴とする請求の範囲1に記載のクロマトグラフィ・アッセイ装 置。 11.前記第1と第2の対置成分はこれらが対置していない位置から直接用手的 に閉じることで対置させ得ることを特徴とする請求の範囲1に記載のクロマトグ ラフィ・アッセイ装置。 12.(a)請求の範囲1の前記クロマトグラフィ・アッセイ装置と、 (b)前記クロマトグラフィ・アッセイ装置で使用され、検出可能な標識で標 識され前記分析対象物に対する特異的結合パートナーと、 を別々のパッケージに含むことを特徴とする分析対象物の検出および/または 判定のための検査キット。 13.(a)各々がアッセイしようとする検体を受け入れるのに適した横方向に 分離された複数の検体調製手段を含む第1の対置成分と、 (b)前記第1の対置成分に取り付け可能で前記第1の対置成分上の各検体調 製手段用のクロマトグラフィ媒体を含み、前記クロマトグラフィ媒体が横方向に 分離されている第2の対置成分と、 (c)各クロマトグラフィ媒体に1つづつ各々が前記第2の対置成分に取り付 けられた複数の導電バリアとを含み、 前記第1と第2の対置成分が各検体調製手段で検査しようとする各検体をこれ に対応する前記導電バリアを介してこれに対応する前記クロマトグラフィ媒体へ 添着するように対置できることを特徴とする少なくとも1つの分析対象物の検出 および/または判定のためのクロマトグラフィ・アッセイ装置。 14.各クロマトグラフィ媒体はさらに前記クロマトグラフィ媒体より実質的に 小さい検出ゾーンを含むことと、各検出ゾーンはアッセイしようとする前記分析 対象物に対する第1の特異的結合パートナーをそのクロマトグラフィ媒体上に含 むことを特徴とする請求の範囲13に記載のクロマトグラフィ・アッセイ装置。 15.少なくとも1つの検体調製手段は検体を含む器具を受け入れるのに適した 折り畳みウェルを含むことを特徴とする請求の範囲13に記載のクロマトグラフ ィ・アッセイ装置。 16.前記第1の対置成分は前記第1の対置成分と前記第2の対置成分を対置さ せた時に前記第2の対置成分上に折り曲がる蝶番式に折り曲げ自在な羽根をさら に含むことを特徴とする請求の範囲15に記載のクロマトグラフィ・アッセイ装 置。 17.(a)請求の範囲13記載の前記クロマトグラフィ・アッセイ装置と、 (b)各々の特異的結合パートナーが検出可能な標識で標識してあり、各々の 特異的結合パートナーは別々にパッケージされ、各々の特異的結合パートナーが 前記クロマトグラフィ・アッセイ装置で使用され、検査しようとする各分析対象 物のための特異的結合パートナーと、 を別々のパッケージに含むことを特徴とする少なくとも1つの分析対象物の検 出および/または判定のための検査キット。 18.分析対象物の検出および/または判定のためのクロマトグラフィ・アッセ イ装置であって、 (a)(i)検体調製手段と (ii)前記検体調製手段と動作的に接触するクロマトグラフィ媒体と を含む第1の対置成分と、 (b)前記第1の対置成分に取り付け可能で、液体の添加により再溶解可能な 形で検出可能な標識により標識された前記分析対象物に対する特異的結合パート ナーを含む添着手段を含む第2の対置成分と、 (c)前記第1の対置成分に取り付けられた導電バリアとを含み、 前記第1と第2の対置成分を対置させることで前記添着手段を前記導電バリア と接触させ、前記分析対象物に対する前記標識特異的結合パートナーが前記導電 バリアを通る液体により溶解されることを特徴とするクロマトグラフィ・アッセ イ装置。 19.前記第1の対置成分は導電手段をさらに含み、前記検体調製手段と前記ク ロマトグラフィ媒体の間の動作的接触が前記検体調製手段と前記クロマトグラフ ィ媒体の両方を前記導電手段と動作的に接触させることで実現することを特徴と する請求の範囲18に記載のクロマトグラフィ・アッセイ装置。 20.前記クロマトグラフィ媒体は第1と第2の端部を有し前記導電手段は前記 クロマトグラフィ媒体の前記第1の端部と動作的に接触し、前記第1の対置成分 は前記クロマトグラフィ媒体の前記第2の端部と動作的に接触する吸収手段をさ (b)前記クロマトグラフィ・アッセイ装置へ挿入するための複数の乾燥試料 を含むことのできる検査カードと、 (c)アッセイしようとする前記分析対象物について前記検査カードに添着さ れる抽出試薬とを別々のパッケージに含むことを特徴とする分析対象物の検出お よび/または判定のための検査キット。 33.検体中の分析対象物の検出および/または判定のためのクロマトグラフィ ・アッセイ装置であって、 (a)(i)第1と第2の端部を有するクロマトグラフィ媒体と (ii)前記クロマトグラフィ媒体の前記第1の端部と動作的に接触し 前記分析対象物の検出用に少なくとも1種類の試薬を含む検出器添着パッドと (iii)前記検出器添着パッドと動作的に接触し前記クロマトグラフ ィ媒体の前記第1の端部と間接接触して液体を通すことのできる導体と (iv)前記クロマトグラフィ媒体の前記第2の端部と動作的に接触し て液体を吸収するためのアブソーバと を含む第1の対置成分と、 (b)検査しようとする検体を受け入れるための検体調製ゾーンを含む第2の 対置成分であって、前記第1と第2の対置成分が対置されて液体が前記第2の対 置成分から前記第1の対置成分へ移し換えられるように前記第1の対置成分に取 り付け自在にしてある第2の対置成分と、 (c)前記第1の対置成分に取り付けられた導電バリアとを含み、 前記第1と第2の対置成分が前記第1と第2の対置成分が対置されない時に前 記第2の対置成分上の前記検体調製ゾーンへ検体を添着できるように、また前記 第1と第2の対置成分を対置させることで前記検体調製ゾーンが前記導体と間接 接触して前記検査しようとする検体を前記導電バリア経由で前記導体へ添着させ て前記導体を通しさらに前記検出器添着パッド経由で前記クロマトグラフィ媒体 の前記第1の端部へ添着して前記検体に対して前記分析対象物の検出用試薬を追 加し、前記導体から前記検出器添着パッドを通り前記クロマトグラフィ媒体の前 記第1の端部への流れが前記アブソーバによる液体の吸収で支援されることを特 徴とするクロマトグラフィ・アッセイ装置。 34.(a)請求の範囲33記載の前記クロマトグラフィ・アッセイ装置と (b)前記検出器添着パッドへ適用される検出可能な標識で標識された前記分 析対象物に対する特異的結合パートナーと を別々の容器に含むことを特徴とする検体中の分析対象物の検出および/また は判定のための検査キット 35.検体中の分析対象物の検出および/または判定のためのクロマトグラフィ ・アッセイ装置であって、 (a)(i)第1と第2の端部を有するクロマトグラフィ媒体と (ii)前記クロマトグラフィ媒体の前記第1の端部と動作的に接触し 液体の添加により再溶解可能な形で前記分析対象物に対する第1の特異的結合パ ートナーを含み、前記第1の特異的結合パートナーを検出可能な標識で標識して ある第1の検出器添着パッドと (iii)前記第1の検出器添着パッドと動作的に接触して液体を通過 させる導体であって、前記第1の検出器添着パッドが前記導体と前記クロマトグ ラフィ媒体の前記第1の端部とを架橋して前記導体から前記第1の検出器添着パ ッドを通り前記クロマトグラフィ媒体の前記第1の端部へ液体が流れるようにす るための導体と (iv)前記クロマトグラフィ媒体の前記第2の端部と動作的に接触し て液体を吸収するためのアブソーバと を含む第1の対置成分と、 (b)前記第1と第2の対置成分を当接させて液体が前記第2の対置成分から 前記第1の対置成分へ移し換えるように前記第1の対置成分に取り付け自在な第 2の対置成分であって、 (i)アッセイしようとする検体を受け入れるための検体調製ゾーンと (ii)前記検体調製ゾーンと動作的に接触し、前記検体調製ゾーンへ 検体の添加により再溶解可能な形で前記分析対象物に対する第2の特異的結合パ ートナーを含み、前記第2の特異的結合パートナーを検出可能な標識で標識して あり、前記検体調製ゾーンへの前記検体の添加で前記第2の特異的結合パートナ ーを溶解できるように前記第2の対置成分上の前記検体調製ゾーンに隣接して配 置されて前記検体調製ゾーンが前記検体と前記第2の特異的結合パートナーの混 合物を含むようにずる第2の検出器添着パッドと、 (c)前記第1の対置成分に取り付けられた導電バリアと、 前記第1と第2の対置成分は前記第1と第2の対置成分が対置されていない時 に前記第2の対置成分上の前記検体調製ゾーンへ検体を添着でき、また前記第1 と第2の対置成分を対置させることによって前記第2の対置成分上の前記検体調 製ゾーンが前記導電バリアを介して前記第1の対置成分上の前記導体と動作的間 接的に接触して検査しようとする前記検体と前記第2の特異的結合パートナーを 前記導体に添着させ、前記導体経由でさらに前記第1の検出器添着パッドを通り 前記クロマトグラフィ媒体の前記第1の端部へ流して前記第1の特異的結合パー トナーを前記検体と前記第2の特異的結合パートナーに追加し、前記導体から前 記第1の検出器添着パッド経由で前記クロマトグラフィ媒体の前記第1の端部へ の流れが前記アブソーバによる液体の吸収によって支援されることを特徴とする クロマトグラフィ・アッセイ装置。 36.前記クロマトグラフィ媒体は前記クロマトグラフィ媒体より面積が実質的 に小さい検出ゾーンをさらに含み、前記検出ゾーンは不動化した前記分析対象物 に対する第3の特異的結合パートナーを含み、前記検体中に分析対象物が存在し ている場合に(1)前記第1と第2の特異的結合パートナーの一方と、(2)前 記分析対象物と、(3)前記不動化した第3の特異的結合パートナーとを含む三 次元複合体が前記検出ゾーンに形成されることを特徴とする請求の範囲33に記 載のクロマトグラフィ・アッセイ装置。 37.(a)請求の範囲35記載の前記クロマトグラフィ・アッセイ装置と、 (b)前記分析対象物に対する前記第1の特異的結合パートナーと前記分析対 象物に対する前記第2の特異的結合パートナーの少なくとも一方を溶解し、前記 第1の対置成分上の前記第1の検出器添着パッドと前記第2の対置成分上の前記 第2の検出器添着パッドの少なくとも一方に添着される液体と を別々の容器に含むことを特徴とする検体中の分析対象物の検出および/また は判定のための検査キット。 38.検体中の分析対象物の検出および/または判定のためのクロマトグラフィ ・ アッセイ装置であって、 (a)(i)第1と第2の端部を有するクロマトグラフィ媒体と (ii)前記クロマトグラフィ媒体の前記第1の端部と動作的に接触し て液体を通過させる導体と (iii)前記クロマトグラフィ媒体の前記第2の端部と動作的に接触 して液体を吸収するためのアブソーバと を含む第1の対置成分と、 (b)前記第1の対置成分に取り付けられる第2の対置成分であって、前記第 1と第2の対置成分を対置させて液体を前記第2の対置成分から前記第1の対置 成分へ移し換えできるようにし、前記第1と第2の対置成分を対置させた時に前 記第1の対置成分上の前記導体へ液体を添着するためのアプリケータを含み、前 記アプリケータが、 (i)前記第1と第2の対置成分が対置されていない時に前記アプリケータ へ液体の添加により再溶解可能な形で前記分析対象物に対する第1の特異的結合 パートナーを含み前記第1の特異的結合パートナーが検出可能な標識で標識され る第1のセクタと (ii)前記分析対象物に対する第1の特異的結合パートナーを欠如する第 2のセクタと の2つのセクタに分割されている前記第2の対置成分と、 (c)前記第1の対置成分に取り付けてある導電バリアと、 を含み、前記第1と第2の対置成分は前記第1と第2の対置成分を対置させる ことで前記第2の対置成分上の前記アプリケータの前記第2のセクタではなく前 記第1のセクタを前記導電バリア経由で前記第1の対置成分上の前記導体と間接 接触させ、前記アプリケータの前記第2のセクタは前記第1のセクタ経由で前記 導体と間接接触して、前記アプリケータの前記第1のセクタの内容を前記クロマ トグラフィ媒体に添着し、前記アプリケータの前記第1のセクタの内容の前記ク ロマトグラフィ媒体への添着に続けて、前記アプリケータの前記第2のセクタの 内容を前記クロマトグラフィ媒体へ添着させ、前記アブソーバが前記クロマトグ ラフィ媒体から液体を排出させて前記アプリケータから前記導体と前記クロマト グラフィ媒体を通る液体の流れを支援するようにしてあることを特徴とするクロ マトグラフィ・アッセイ装置。 39.前記クロマトグラフィ媒体は前記クロマトグラフィ媒体より面積が実質的 に小さい検出ゾーンをさらに含み、前記検出ゾーンはこれに不動化した前記分析 対象物に対する第2の特異的結合パートナーを含み、分析対象物が前記検体中に 存在する場合には前記第1の特異的結合パートナーと、前記分析対象物と、前記 第2の特異的結合パートナーとを含む三次元複合体が前記検出ゾーンに形成され ることを特徴とする請求の範囲23,25,27,33,または38のいずれか 1つに記載のクロマトグラフィ・アッセイ装置。
