Claims (1)
- 【特許請求の範囲】 1.選択物質を検出するためのバイオディテクターであって、 (a)細胞外リガンド特異的部分が選択物質を選択的に認識し、その認識が細 胞内シグナル変換ドメインを活性化する、細胞外リガンド特異的部分と細胞内シ グナル変換ドメインを含むシグナル転換要素、 (b)互いに別個の不活性形態および活性形態を持ち、かつ該活性化細胞内シ グナル変換ドメインがその不活性形態を活性形態へと転換する、トランスデュー サー、および (c)該トランスデューサーの該活性形態により活性化されて、検出可能なシ グナルを生じる、応答要素 を含む、バイオディテクター。 2.該応答要素が、該トランスデューサーの活性形態により活性化される転写 活性化要素を含む、請求の範囲第1項記載のバイオディテクター。 3.該応答要素が、更に、1または複数の遺伝子産物をコードする核酸を含み 、遺伝子産物は検出可能なシグナルを発生するものであり、該核酸は該転写活性 化要素に作動可能に連結している、請求の範囲第2項記載のバイオディテクター 。 4.該検出可能なシグナルが光である、請求の範囲第3項記載のバイオディテ クター。 5.該遺伝子産物が、バイオ発光、比色性反応または蛍光からなる群から選択 される手段により検出可能である、請求の範囲第3項記載のバイオディテクター 。 6.該核酸がルシフェラーゼオペロンを含む、請求の範囲第3項記載のバイオ ディテクター。 7.該細胞内シグナル変換要素が膜シグナルトランスデューサーである、請求 の範囲第6項記載のバイオディテクター。 8.該膜シグナルトランスデューサーが、細菌性二成分制御系、真核生物レポ ーター媒介シグナルトランスデューサー、原核生物レポーター媒介シグナルトラ ンスデューサーからなる群から選択される、請求の範囲第7項記載のバイオディ テクター。 9.該物質が、微生物、ウイルス、レトロウイルス、タンパク質、糖、イオン からなる群から選択される、請求の範囲第6項記載のバイオディテクター。 10.選択物質の検出法であって、 (a)(i)細胞外リガンド特異的部分が選択物質を選択的に認識し、その認 識が細胞内シグナル変換ドメインを活性化する、細胞外リガンド特異的 部分と細胞内シグナル変換ドメインを含むシグナル転換要素 (ii)別個の不活性形態および活性形態を持ち、かつ該不活性形態が該 活性化細胞内シグナル変換ドメインにより該活性形態へと転換される、 トランスデューサー、および (iii)該トランスデューサーの該活性形態により活性化されて、検出 可能なシグナルを生じる、応答要素 を含む、バイオディテクターを製造し、 (b)該バイオディテクターを試料に加え、 (c)該検出可能なシグナルを測定および定量し、 (d)該検出可能なシグナルのレベルを該物質の存在および量と相関させる、 ことを含む、方法。 11.該バイオディテクターの該応答要素が、該トランスデューサーの活性形 態により活性化される転写活性化要素を含む、請求の範囲第10項記載の方法。 12.該応答要素が、更に、1または複数の遺伝子産物をコードする核酸を含 み、遺伝子産物は該検出可能なシグナルを発生するものであり、該核酸は該転写 活性化要素に作動可能に連結している、請求の範囲第10項記載の方法。 13.該検出可能なシグナルが光である、請求の範囲第12項記載の方法。 14.該遺伝子産物が、バイオ発光、比色性反応または蛍光からなる群から選 択される手段により検出可能である、請求の範囲第12項記載の方法。 15.該核酸がルシフェラーゼオペロンを含む、請求の範囲第12項記載の方 法。 16.該物質が、微生物、ウイルス、レトロウイルス、タンパク質、糖、イオ ンからなる群から選択される、請求の範囲第12項記載の方法。 17.該検出可能なシグナルが光検出システムによって検出される、請求の範 囲第13項記載の方法。 18.該光検出システムが、照度計、分光光度計、蛍光計、CCD検出器から 選択される、請求の範囲第17項記載の方法。 19.バイオディテクターまたは試料が固相支持体上に固定されている、請求 の範囲第18項記載の方法。 20.試料中に含まれる様々な物質を検出できるように、更に、固相支持体上 に規則正しく並べて一連のバイオディテクターを固定することを含む、請求の範 囲第19項記載の方法。
