Claims (1)
- 【特許請求の範囲】 1.標的分子が、遺伝子ファミリーメンバー由来の配列を有するか否かを決定 するための方法であって、以下の工程: 少なくとも2つの異なる遺伝子ファミリーメンバーの各々について、該遺伝子 ファミリーメンバー由来の参照ヌクレオチド配列を規定する1セットのポリヌク レオチドプローブを含むポリヌクレオチドアレイを提供する工程; 該標的核酸分子と該セット中の該プローブとの間のハイブリダイゼーション反 応を行うこと、および該標的核酸分子と該セット中の各該プローブとの間のハイ ブリダイゼーションを検出することにより、ハイブリダイゼーションデータを作 製する工程;ならびに 該標的核酸が、該遺伝子ファミリーメンバーの1つに由来する該参照配列を有 するか否かを決定するためにハイブリダイゼーションデータをプロセスする工程 、を包含する、方法。 2.請求項1に記載の方法であって、前記標的が、前記遺伝子ファミリーメンバ ーをコードする遺伝子にハイブリダイズする核酸プローブにハイブリダイズする か否かを決定することにより、前記標的核酸分子を選択する工程をさらに包含す る、方法。 3.請求項1に記載の方法であって、前記プロセシング工程がプログラム可能な デジタルコンピューターによって行われる、方法。 4.請求項1に記載の方法であって、前記ポリヌクレオチドアレイが、前記遺伝 子ファミリーメンバーの各々について、前記遺伝子の高度な保存領域を規定する プローブセットおよび該遺伝子の高度な可変領域を規定するプローブセットをさ らに含む、方法。 5.請求項1に記載の方法であって、前記ポリヌクレオチドアレイが、前記遺伝 子ファミリーメンバーの各々について、前記遺伝子の少なくとも2つの高度な保 存領域を規定するプローブセットおよび該遺伝子の少なくとも2つの高度な可変 領域を規定するプローブセットをさらに含む、方法。 6.請求項1に記載の方法であって、前記領域がアミノ酸配列をコードし、そし て前記アレイが該アミノ酸配列をコードする縮重セットのヌクレオチド配列を規 定するプローブセットをさらに含む、方法。 7.請求項1に記載の方法であって、前記標的が遺伝子ファミリーメンバーの前 記領域の配列を有さない場合、前記標的核酸分子の前記ヌクレオチド配列を決定 する工程をさらに包含する、方法。 8.試料中の核酸が遺伝子ファミリーのメンバーであるか否かを決定する方法で あって、以下の工程: 少なくとも第1および第2のアッセイを包含する階層制のアッセイ技術を選択 する工程であって、選択される該第1のアッセイは第1の配列サインの存在、非 存在、または変異体の決定を提供し、そして選択される該第2のアッセイは第2 の配列サインの存在、非存在、または変異体の決定を提供し、ここで、該アッセ イの少なくとも1つが高密度核酸アレイを使用する工程; 該第1のアッセイを用いて該核酸試料を分析する工程;および 該核酸が該第1および第2のアッセイの結果に基づく該遺伝子ファミリーメン バーであるか否かを決定する工程、 を包含する、方法。 9.請求項8に記載の方法であって、前記第1の配列サインが遺伝子ファミリー の高度な保存領域である、方法。 10.請求項8に記載の方法であって、前記第2の配列サインが遺伝子ファミリ ーの非保存領域である、方法。 11.請求項8に記載の方法であって、前記核酸試料の全長配列を決定する工程 をさらに包含する、方法。 12.請求項8に記載の方法であって、前記遺伝子ファミリーがTGF-βファミリ ーである、方法。 13.請求項8に記載の方法であって、前記第1または第2の配列サインが10と 100との間のヌクレオチド長である、方法。 14.請求項13に記載の方法であって、前記第1または第2の配列サインが18 と30との間のヌクレオチド長である、方法。 15.分析のためのクローンを選択するための方法であって、以下の工程: 支持体に結合した多様なクローンを有する該支持体を提供する工程; 該支持体を、低い、中程度な、または高いストリンジェンシーの条件下で1つ 以上のポリヌクレオチドに曝し、該クローンと該ポリヌクレオチドとの間の少な くとも幾つかのハイブリダイゼーションを可能にする、工程; 該ポリヌクレオチドとハイブリダイズする該クローンを同定する工程;および 分析のための該同定工程において同定されない、少なくとも1つの該クローン を選択する工程、 を包含する、方法。 16.請求項15に記載の方法であって、前記支持体が高密度核酸アレイである 、方法。 17.分析のための試料を狭める方法であって、以下の工程: 核酸を含む試料を提供する工程; 高密度核酸アレイを用いて該試料が配列サインを含むか否かを分析する工程; および 該配列サインが存在しない場合のみ該核酸試料をさらに分析する工程、 を包含する、方法。 18.TGF-β遺伝子ファミリー由来の配列サインを含む、高密度核酸アレイ。
