Claims (1)
- 【特許請求の範囲】 1.試薬組成物であって、該組成物は、核酸増幅緩衝液、および内部参照分子を 含み、該内部参照分予は、 第1の発蛍光団、 第2の発蛍光団、および 核酸増幅条件下で増幅用ポリヌクレオチドに、配列特異的な様式でハイブリ ダイズしない、バックボーンコネクター を含み、ここで該バックボーンコネクターは、該第1の発蛍光団および該第2の 発蛍光団と、該第1の発蛍光団から該第2の発蛍光団へのエネルギー移動を可能 にするように結合している、試薬組成物。 2.前記バックボーンコネクターがポリヌクレオチドである、請求項1に記載の 組成物。 3.前記ポリヌクレオチドが、長さ2〜25個のヌクレオチドである、請求項2に 記載の組成物。 4.前記ポリヌクレオチドが(dT)8である、請求項3に記載の組成物。 5.前記第1の発蛍光団が、フルオレセイン、6-カルボキシフルオレセイン、2' ,4',5',7'-テトラクロロ-4,7-ジクロロフルオレセイン、2',7'-ジメトキシ-4',5 '-6-カルボキシローダミン(JOE)、N',N',N',N'-テトラメチル-6-カルボキシロー ダミン(TAMRA)、および6-カルボキシ-X-ローダミン(ROX)からなる群から選択さ れる、請求項1に記載の組成物。 6.前記第1の発蛍光団が6-カルボキシフルオレセインである、請求項5に記載 の組成物。 7.前記第2の発蛍光団が、フルオレセイン、6-カルボキシフルオレセイン、2' ,4',5',7'-テトラクロロ-4,7-ジクロロフルオレセイン、2',7'-ジメトキシ-4',5 '-6-カルボキシローダミン(JOE)、N',N',N',N'-テトラメチル-6-カルボキシロー ダミン(TAMRA)、および6-カルボキシ-X-ローダミン(ROX)からなる群から選択さ れる、請求項5に記載の組成物。 8.前記第2の発蛍光団が6-カルボキシ-X-ローダミンである、請求項6に記載 の組成物。 9.核酸増幅反応における増幅用ポリヌクレオチドの増幅生成物の量を測定する ための改良された方法であって、該方法が 内部増幅参照分子を該増幅反応に加える工程を含み、該内部参照分子は 第1の発蛍光団; 第2の発蛍光団;および 核酸増幅条件下で該増幅用ポリヌクレオチドにハイブリダイズしないバック ボーンコネクターを含み、 ここでバックボーンコネクターは、該第1の発蛍光団および該第2の発蛍光団と 、該第1の発蛍光団から該第2の発蛍光団へのエネルギー移動を可能にするよう に結合している、改良された方法。 10.前記核酸増幅反応がリアルタイムで測定される、請求項9に記載の方法。 11.前記バックボーンコネクターがポリヌクレオチドである、請求項9に記載 の組成物。 12.前記ポリヌクレオチドが、長さ2〜25個のヌクレオチドである、請求項1 0に記載の組成物。 13.前記ポリヌクレオチドが(dT)8である、請求項12に記載の組成物。 14.前記第1の発蛍光団が、フルオレセイン、6-カルボキシフルオレセイン、 2',4',5',7'-テトラクロロ-4,7-ジクロロフルオレセイン、2',7'-ジメトキシ-4' ,5'-6-カルボキシローダミン(JOE)、N',N',N',N'-テトラメチル-6-カルボキシロ ーダミン(TAMRA)、および6-カルボキシ-X-ローダミン(ROX)からなる群から選択 される、請求項9に記載の組成物。 15.前記第1の発蛍光団が6-カルボキシフルオレセインである、請求項14に 記載の組成物。 16.前記第2の発蛍光団が、フルオレセイン、6-カルボキシフルオレセイン、 2',4',5',7'-テトラクロロ-4,7-ジクロロフルオレセイン、2',7'-ジメトキシ-4' ,5'-6-カルボキシローダミン(JOE)、N',N',N',N'-テトラメチル-6-カルボキシロ ーダミン(TAMRA)、および6-カルボキシ-X-ローダミン(ROX)からなる群から選択 される、請求項14に記載の組成物。 17.前記第2の発蛍光団が6-カルボキシ-X-ローダミンである、請求項15に 記載の組成物。 18.核酸増幅反応を実施するためのキットで、該キットは、内部参照分子、な らびに、フルオレッサー-クエンチャープローブおよび熱安定性DNAポリメラーゼ からなる群から選択される1つ以上の試薬を含む、キット。 19.前記試薬がフルオレッサー-クエンチャープローブである、請求項18に 記載のキット。 20.前記試薬が熱安定性DNAポリメラーゼである、請求項18に記載のキット 。 21.前記キットがフルオレッサー-クエンチャープローブをさらに含む、請求 項20に記載のキット。
