Claims (1)
- 【特許請求の範囲】 1. (a)塩及び/又はポリペプチドを包含するオリゴ糖溶液を、カーボンを 含む固体担体と反応させて、オリゴ糖を実質的に固体担体に結合させ; (b)担体を洗浄して塩及び/又はポリペプチドを除去し; そして (c)結合されたオリゴ糖を担体から溶離する諸工程を含む、塩及び/又はポ リペプチドからオリゴ糖を分離する方法。 2. 固体担体がグラファイト化カーボンである、請求項1に記載の方法。 3. 担体がクロマトグラフカラム又はカートリッジに充填されている、請求項 1又は2に記載の方法。 4. 洗浄工程(b)が水をもってし、そして溶離工程(c)が有機調節剤を包 含する、請求項1〜3のいずれか一項に記載の方法。 5. 有機調節剤がアセトニトリルである、請求項4に記載の方法。 6. 有機調節剤がメタノール、エタノール、プロパノール及びブタノールから なる群から選ばれる、請求項4に記載の方法。 7. 有機調節剤の濃度が1〜99%(v/v)である、請求項5又は6に記載 の方法。 8. 結合したオリゴ糖は溶離工程(c)中に有機調節剤の濃度を変動させるこ とによって取り出される、請求項1〜7のいずれか一項に記載の方法。 9. 担体に結合された各種オリゴ糖は酸性又は塩基性調節剤と組み合わされた 有機調節剤の各種濃度によって溶離される、請求項1〜8のいずれか一項に記載 の方法。 10.酸性調節剤が希酸である、請求項9に記載の方法。 11.希酸がトリフルオロ酢酸である、請求項10に記載の方法。 12.トリフルオロ酢酸の濃度が0.01〜1%(v/v)である、請求項11 に記載の方法。 13.酸性又は塩基性調節剤の濃度が0.01〜5%(v/v)である、請求項 9に記載の方法。 14.塩が塩化ナトリウム及び塩化カリウム、苛性アルカリ、陽イオン緩衝剤、 陰イオン緩衝剤、両性イオン緩衝剤、及び界面活性剤である、請求項1〜13の いずれか一項に記載の方法。 15.苛性アルカリが水酸化ナトリウムであり、陽イオン緩衝剤がトリスであり 、陰イオン緩衝剤が燐酸塩及び酢酸塩であり、両性イオン緩衝剤がHEPES及 びNOPSであり、そして界面活性剤がドデシル硫酸ナトリウム(SDS)であ る、請求項14に記載の方法。 16.オリゴ糖が塩から分離される、請求項1〜15のいずれか一項に記載の方 法。 17.オリゴ糖がポリプペプチドから分離される、請求項1〜13のいずれか一 項に記載の方法。
