Claims (17)
1. Polipeptid koji obuhvaća ili se sastoji od aminokiselinske sekvence izabrane iz grupe koju čine:
(a) bilo koja od SEQ ID NO: 1, 2, 3 i 5 iz α3 lanca kolagena tipa VI; i
(b) aminokiselinska sekvenca s najmanje 80% sličnosti sa sekvencom prema bilo kojoj od SEQ ID NO: 1, 2, 3 i 5;
pri čemu je polipeptid dužine između 20 i 200 aminokiselina; i
pri čemu je polipeptid sposoban usmrtiti ili oslabiti rast mikroorganizama.
2. Polipeptid prema patentnom zahtjevu 1, gdje:
(i) su mikroorganizmi odabrani iz grupe koju čine bakterije, mikoplazme, kvasci, glјive i virusi; i/ili
(ii) polipeptid ima sposobnost vezati se za membranu mikroorganizma; i/ili
(iii) polipeptid ima sposobnost izazvati narušavanje membrane mikroorganizama; i/ili
(iv) polipeptid ima sposobnost potaknuti zatvaranje rane; i/ili
(v) polipeptid ima sposobnost ispoljiti antimikrobni efekt koji je veći ili jednak onom kod LL-37.
3. Polipeptid prema patentnom zahtjevu 1 ili 2, pri čemu:
(i) mikroorganizmi su gram-pozitivne ili gram-negativne bakterije, po potrebi gdje se mikroorganizmi biraju iz grupe koju čine: Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli, streptokok grupe A (npr. Streptococcus pyogenes), streptokok grupe B (npr. Streptococcus agalactiae), streptokok grupe C (npr. Streptococcus dysgalactiae), streptokok grupe D (npr. Enterococcus faecalis), streptokok grupe F (npr. Streptococcus anginosus), streptokok grupe G (npr. Streptococcus dysgalactiae equisimilis), alfa-hemolitički streptokok (npr. Streptococcus viridans, Streptococcus pneumoniae), Streptococcus bovis, Streptococcus mitis, Streptococcus anginosus, Streptococcus sanguinis, Streptococcus suis, Streptococcus mutans, Moraxella catarrhalis, „non-typeable“ Haemophilus influenzae (NTHi), Haemophilus influenzae b (Hib), Actinomyces naeslundii, Fusobacterium nucleatum, Prevotella intermedia, Klebsiella pneumoniae, Enterococcus cloacae, Enterococcus faecalis, Staphylococcus epidermidis, višestruko rezistentan Pseudomonas aeruginosa (MRPA), višestruko rezistentan Staphylococcus aureus (MRSA), višestruko rezistentna Escherichia coli (MREC), višestruko rezistentan Staphylococcus epidermidis (MRSE) i višestruko rezistentna Klebsiella pneumoniae (MRKP); i/ili
(ii) mikroorganizmi su bakterije koje su rezistentne na jedan ili više konvencionalnih antibiotika, po potrebi
mikroorganizam se bira iz grupe koju čine: rezistentan na više lijekova Staphylococcus aureus (MRSA), rezistentan na više lijekova Pseudomonas aeruginosa (MRPA), višestruko rezistentna Escherichia coli (MREC), višestruko rezistentan Staphylococcus epidermidis (MRSE) i višestruko rezistentna Klebsiella pneumoniae (MRKP).
4. Polipeptid prema bilo kojem od prethodnih patentnih zahtjeva, gdje: (i) polipeptid je suštinski netoksičan za ćelije sisavaca; i/ili
(ii) polipeptid je sposoban ispoljavati anti-endotoksični efekt; i/ili
(iii) polipeptid je izveden iz domene tipa A von Wilebrandovovog faktora; i/ili
(iv) polipeptid je izveden iz α3 lanca kolagena tipa VI; i/ili
(v) polipeptid je izveden iz N2, N3 ili C1 domena α3 lanca kolagena tipa VI; i/ili
(vi) polipeptid ima neto pozitivni naboj, po potrebi se naboj polipeptida kreće između +2 i +9; i/ili
(vii) polipeptid ima najmanje 30% hidrofobnih ostataka.
5. Polipeptid prema bilo kojem od prethodnih patentnih zahtjeva, pri čemu:
(i) polipeptid je dužine između 20 i 150 aminokiselina, na primjer između 20 i 100, 20 i 50, 20 i 40 ili 25 i 35 aminokiselina u dužini; i/ili
(ii) polipeptid sadrži jednu ili više aminokiselina koje su modificirane ili izvedene; jedna ili više aminokiselina su po potrebi modificirane ili izvedene PEGilacijom, amidacijom, esterifikacijom, acilacijom, acetilacijom i/ili alkilacijom; i/ili
(iii) polipeptid je rekombinantni polipeptid.
6. Polipeptid prema bilo kojem od prethodnih patentnih zahtjeva, gdje polipeptid obuhvaća ili se sastoji od aminokiselinske sekvence s najmanje 90% identičnosti prema bilo kojoj od SEQ ID NO: 1, 2, 3 ili 5.
7. Molekula nukleinske kiseline koji kodira polipeptid prema bilo kojem od prethodnih patentnih zahtjeva, ili vektor koji obuhvaća molekulu nukleinske kiseline, gdje je po potrebi vektor ekspresijski vektor.
8. Stanica domaćin koja obuhvaća molekulu nukleinske kiseline ili vektor prema patentnom zahtjevu 7.
9. Postupak za dobivanje polipeptida prema bilo kojem od patentnih zahtjeva od 1 do 6 koji obuhvaća:
(i) uzgoj populacije stanica domaćina u kulturi prema patentnom zahtjevu 8 pod uvjetima u kojima je spomenuti polipeptid eksprimiran, i izoliranje polipeptida iz njih; ili
(ii) sinteza polipeptida u tekućoj ili čvrstoj fazi.
10. Farmaceutski sastav koja sadrži polipeptid prema bilo kojem od patentnih zahtjeva od 1 do 6 zajedno s farmaceutski prihvatlјivim ekscipijensom, razrjeđivačem, nosačem, puferom ili pomoćnim sredstvom, gdje je po potrebi farmaceutski sastav pogodan za primjenu odabranim putem i to načinom odabranim iz grupe koja se sastoji od oralne primjene, parenteralne primjene i lokalne primjene.
11. Farmaceutski sastav prema patentnom zahtjevu 10, gdje je sastav u obliku polimernog gela, gdje po potrebi polimer sadrži kolagen.
12. Medicinski uređaj, implantat, proizvod za njegu rana ili materijal za upotrebu u istom, koji je obložen, impregniran, pomiješan ili na drugi način povezan s farmaceutskim sastavom prema patentnom zahtjevu 10 ili 11, gdje je po potrebi:
(i) uređaj, implantat, proizvod za njegu rana ili materijal za upotrebu u by-pass kirurgiji, ekstrakorporalnoj cirkulaciji, njezi rana i/ili dijalizi; i/ili
(ii) farmaceutski sastav je obložen, obojen, poprskan ili na drugi način nanesen na šav, protezu, implantat, zavoj za ranu, kateter, leću, kožni graft, zamjenu za kožu, fibrinsko ljepilo ili zavoj; i/ili
(iii) medicinski uređaj, implantat, proizvod za njegu rana ili materijal za upotrebu sadrži ili se sastoji od polimera, metala, metalnog oksida i/ili keramike.
13. Komplet koji sadrži farmaceutski sastav prema patentnom zahtjevu 10 ili 11 ili medicinski uređaj, implantat, proizvod za njegu rana ili materijal za upotrebu u istom, prema patentnom zahtjevu 12.
14. Polipeptid kao što je definirano u bilo kojem od patentnih zahtjeva od 1 do 6, ili molekula nukleinske kiseline kao što je definirano u patentnom zahtjevu 7, ili farmaceutski sastav kao što je definirano u patentnom zahtjevu 10 ili 11 za upotrebu u medicini.
15. Polipeptid prema bilo kojem od patentnih zahtjeva od 1 do 6, ili molekulu nukleinske kiseline kao što je definiran u patentnom zahtjevu 7, ili farmaceutski sastav prema patentnom zahtjevu 10 ili 11 za upotrebu u kurativnom i/ili profilaktičkom tretmanu mikrobnih infekcija, gdje po potrebi:
(i) mikrobna infekcija je sistemska infekcija; i/ili
(ii) mikrobna infekcija je rezistentna na jedan ili više konvencionalnih antibiotika; i/ili
(iii) mikrobnu infekciju izaziva mikroorganizam izabran iz grupe koju čine: Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli, streptokok grupe A (npr. Streptococcus pyogenes), streptokok grupe B (npr. Streptococcus agalactiae), streptokok grupe C (npr. Streptococcus dysgalactiae), streptokok grupe D (npr. Entero-coccus faecalis), streptokok grupe F (npr. Streptococcus anginosus), streptokok grupe G (npr. Streptococcus dysgalactiae equisimilis), alfa-hemolitički streptokok (npr. Streptococcus viridans, Streptococcus pneumoniae), Streptococcus bovis, Streptococcus mitis, Streptococcus anginosus, Streptococcus sanguinis, Streptococcus suis, Streptococcus mutans, Moraxella catarrhalis, „non-typeable“ Haemophilus influenzae (NTHi), Haemophilus influenzae b (Hib), Actinomyces naeslundii, Fusobacterium nucleatum, Prevotella intermedia, Klebsiella pneumoniae, Enterococcus cloacae, Enterococcus faecalis, Staphylococcus epidermidis, višestruko rezistentan Pseudomonas aeruginosa (MRPA) i višestruko rezistentan Staphylococcus aureus (MRSA), višestruko rezistentna Escherichia coli (MREC), višestruko rezistentan Staphylococcus epidermidis (MRSE) i višestruko rezistentna Klebsiella pneumoniae (MRKP); i/ili
(iv) mikrobnu infekciju izaziva mikroorganizam izabran iz grupe koju čine: rezistentan na više lijekova Staphylococcus aureus (MRSA) i višestruko rezistentan Pseudomonas aeruginosa (MRPA); i/ili
(v) polipeptid, molekula nukleinske kiseline ili farmaceutski sastav je za upotrebu u kombinaciji s jednim ili više dodatnih antimikrobnih agenasa, pri čemu se po potrebi jedno ili više dodatnih antimikrobnih agenasa bira iz grupe koju čine: antimikrobni polipeptidi i antibiotici.
16. Polipeptid kao što je definiran u bilo kojem od patentnih zahtjeva od 1 do 6, ili molekula nukleinske kiseline kao što je definirana u patentnom zahtjevu 7, ili farmaceutski sastav prema patentnom zahtjevu 10 ili 11 za upotrebu u njezi rana.
17. Postupak za in vitro ubijanje mikroorganizama koji obuhvaća dovođenje u kontakt mikroorganizama s polipeptidom kao što je opisano u bilo kojem od patentnih zahtjeva od 1 do 6, ili s molekulom nukleinske kiseline kao što je definirano u patentnom zahtjevu 7, ili s farmaceutskim sastavom kao što je definirano u patentnom zahtjevu 10 ili 11.
