Claims (15)
1. Postupak za povećanje sadržaja β-glukozidaze i/ili β-ksilozidaze enzimskog koktela iz celulolitičkog mikroorganizma koji proizvodi celulaze i/ili hemi-celulaze, naznačen time što sadrži:
- korak za proizvodnju enzima s dobivenim medijem koji sadrži enzime i mošt mikroorganizma, navedeni mošt se odvaja, ili ne odvaja, iz tekućine koja sadrži navedene enzime,
- korak hlađenja navedenog mošta na temperaturu koja iznosi između 4 i 20°C, što je temperatura ispod temperature koraka proizvodnje enzima, tijekom vremena koje iznosi između 12 i 90 sati tako da:
- je koncentracija tekućine β-glukozidaze ili β-ksilozidaze koja potječe iz koraka hlađenja veća od koncentracije tekućine koja potječe iz koraka proizvodnje enzima,
i/ili
- omjer volumen krutine / ukupni volumen je manji od navedenog omjera za korak proizvodnje enzima,
- i enzimski koktel se dobije na kraju koraka hlađenja.
2. Postupak prema patentnom zahtjevu 1 naznačen time što trajanje koraka hlađenja iznosi između 24 i 72 sata.
3. Postupak prema bilo kojem od prethodnih patentnih zahtjeva naznačen time što se, na kraju koraka proizvodnje enzima, mošt odvaja od tekućine.
4. Postupak prema prethodnom patentnom zahtjevu naznačen time što se navedeni odvojeni mošt razrjeđuje vodom.
5. Postupak prema patentnom zahtjevu 1 ili 2 naznačen time što se mošt, na kraju koraka proizvodnje enzima, ne odvaja od tekućine.
6. Postupak prema jednom od prethodnih zahtjeva naznačen time što se, nakon koraka hlađenja, enzimski koktel koji potječe iz mošta odvaja i dobije se preostali mošt.
7. Postupak prema jednom od patentnih zahtjeva 3 ili 4 i 6 naznačen time što se odvajanje provodi pomoću barem jednog od centrifugiranja ili filtracije / prešanja ili mikrofiltracije, kojima proizvoljno prethodi taloženje.
8. Postupak prema patentnom zahtjevu 6 naznačen time što se enzimi odijeljene tekućine mogu koncentrirati, na primjer ultrafiltracijom.
9. Postupak prema jednom od prethodnih zahtjeva naznačen time što se korak hlađenja provodi pri pH koji se nalazi između 3,5 i 5,5.
10. Postupak prema jednom od prethodnih zahtjeva naznačen time što se korak hlađenja provodi u atmosferi osiromašenoj kisikom, poželjno u anaerobnoj atmosferi.
11. Postupak prema prethodnom patentnom zahtjevu naznačen time što se ubrizgava neutralni plin, na primjer ugljikov dioksid ili dušik.
12. Postupak prema jednom od prethodnih zahtjeva naznačen time što mikroorganizam pripada rodu Trichoderma, naročito Trichoderma reesei, a poželjno je soj CL847.
13. Postupak prema jednom od prethodnih zahtjeva naznačen time što se navedeni preostali mošt podvrgava koraku hlađenja i enzimski koktel, odvojen ili ne odvojen od preostalog mošta, dobiva se na kraju koraka hlađenja i poželjno navedeni enzimski koktel se odvaja od preostalog mošta iz navedenog koraka hlađenja.
14. Postupak prema jednom od prethodnih zahtjeva naznačen time što se miješaju enzimski kokteli koji potječu iz mošta.
15. Postupak prema jednom od prethodnih zahtjeva naznačen time što je najmanje jedan enzimski koktel pomiješan s odvojenom tekućinom koja sadrži enzime i koja potječe iz koraka proizvodnje enzima.
