Claims (15)
1. Dijagnostički koristan nosač koji sadrži sredstvo za specifično hvatanje protutijela na epitop C-terminalnog kraja Ara h 7 izotipa 7.0201, pri čemu C-terminalni kraj sadrži SEQ ID NO 6 i pri čemu epitop sadrži SEQ ID NO 8, u uzorku od subjekta,
pri čemu je nosač odabran iz skupine koja sadrži kuglice, test traku, mikrotitarsku ploču, microarray, čvrsti polimer dobiven iz celuloze, blot iz skupine koja sadrži line blot i dot blot, staklenu površinu, klizač i biočip, te je najpoželjnije mikrotitarska ploča ili line blot,
pri čemu je sredstvo za specifično hvatanje protutijela polipeptid koji sadrži ili obuhvaća antigen na koji se veže protutijelo koje se treba uhvatiti ili detektirati ili njegova varijanta koja ima aktivnost vezanja protutijela.
2. Dijagnostički koristan nosač prema patentnom zahtjevu 1, naznačen time što nosač nadalje sadrži sredstvo za specifično hvatanje protutijela na jedan ili više daljnjih antigena iz skupine koja sadrži Ara h 2, Ara h 6, Ara h 1, Ara h 3, Ara h 9, Ara h 8 i Ara h 5, poželjnije iz skupine koja sadrži Ara h 2, Ara h 6, Ara h 1, Ara h 3, Ara h 9, poželjnije protutijelo na Ara h 2 i/ili protutijelo na Ara h 6, najpoželjnije protutijelo na Ara h 2.
3. Komplet naznačen time što sadrži dijagnostički koristan nosač prema bilo kojem od patentnih zahtjeva 1 do 2, kao i sredstvo za specifično otkrivanje uhvaćenog protutijela.
4. Komplet prema patentnom zahtjevu 3,
naznačen time što sredstvo za specifično hvatanje protutijela na epitop C-terminalnog kraja Ara h 7 izotipa 7.0201 pri čemu se C-terminalni kraj sastoji od SEQ ID NO 6 i pri čemu epitop sadrži SEQ ID NO 8, te je sredstvo za specifično hvatanje protutijela na jedan ili više daljnjih antigena na odvojenim nosačima ili
pri čemu sredstvo za specifično hvatanje protutijela na epitop C-terminalnog kraja Ara h 7 izotipa 7.0201 gdje C-terminalni kraj sadrži SEQ ID NO 6 i pri čemu epitop sadrži SEQ ID NO 8, te je sredstvo za specifično hvatanje protutijela na jedan ili više daljnjih antigena vezano na jednom, poželjno kovalentno vezano na jednom nosaču.
5. Postupak za dijagnozu alergije na kikiriki naznačen time što obuhvaća korak otkrivanja u uzorku od subjekta prisutnosti protutijela na epitopu C-terminalnog kraja Ara h 7 izotipa 7.0201, pri čemu C-terminalni kraj sadrži SEQ ID NO 6 i pri čemu epitop sadrži SEQ ID NO 8.
6. Postupak prema patentnom zahtjevu 5, naznačen time što nadalje obuhvaća korak otkrivanja u uzorku subjekta prisutnosti protutijela na jednom ili više daljnjih antigena, poželjno odabranih iz skupine koja sadrži Ara h 2, Ara h 6, Ara h 1, Ara h 3, Ara h 9, Ara h 8 i Ara h 5, poželjnije iz skupine koja sadrži Ara h 2, Ara h 6, Ara h 1, Ara h 3 i Ara h 9, najpoželjnije Ara h 2 i/ili Ara h 6.
7. Postupak prema bilo kojem od patentnih zahtjeva 5 do 6, naznačen time što se istovremeno određuje prisutnost protutijela na epitopu C-terminalnog kraja Ara h 7 izotipa 7.0201 koji sadrži SEQ ID NO 8 i prisutnost protutijela na jednom ili više daljnjih antigena, poželjno Ara h 2 i/ili Ara h 6.
8. Postupak prema bilo kojem od patentnih zahtjeva 5 do 7, naznačen time što se prisutnost protutijela na epitopu C-terminalnog kraja Ara h 7 izotipa 7.0201 koji sadrži SEQ ID NO 8 i prisutnost protutijela na jednom ili više daljnjih antigena određuju u prostorno odvojenim reakcijama vezivanja.
9. Postupak prema bilo kojem od patentnih zahtjeva 5 do 8, naznačen time što se prisutnost protutijela na epitopu C-terminalnog kraja Ara h 7 izotipa 7.0201 koji sadrži SEQ ID NO 8 i prisutnost protutijela na jednom ili više daljnjih antigena određuju reakcijom u jednoj posudi.
10. Nosač, komplet ili postupak prema bilo kojem od patentnih zahtjeva 1 do 9, naznačen time što je protutijelo na izotipu 7.0201 protutijelo monospecifično prema SEQ ID NO 6, poželjnije prema sekvenci iz skupine koja sadrži SEQ ID NO 7, SEQ ID NO 8, SEQ ID NO 9 i SEQ ID NO 10, najpoželjnije SEQ ID NO 8.
11. Uporaba polipeptida koji sadrži SEQ ID NO 8, poželjnije sekvence iz skupine koja sadrži SEQ ID NO 6, SEQ ID NO 7, SEQ ID NO 8, SEQ ID NO 9 i SEQ ID NO 10, poželjnije SEQ ID NO 6, najpoželjnije Ara h 7 izotip 7.0201 ili njegova varijanta koja se veže na protutijelo na Ara h 7 izotip 7.0201 od subjekta alergičnog na kikiriki, pri čemu je polipeptid rekombinantni, imobilizirani pročišćeni ili fuzijski protein, poželjno pročišćeni i rekombinantni i imobilizirani, te nosača prema bilo kojem od patentnih zahtjeva 1 do 2 ili kompleta prema bilo kojem od patentnih zahtjeva 3 do 4 naznačena time što je za povećanje osjetljivosti dijagnoze alergije na kikiriki.
12. Uporaba polipeptida koji sadrži SEQ ID NO 8, poželjnije sekvence iz skupine koja sadrži SEQ ID NO 6, SEQ ID NO 7, SEQ ID NO 8, SEQ ID NO 9 i SEQ ID NO 10, poželjnije SEQ ID NO 6, najpoželjnije Ara h 7 izotip 7.0201 ili njegova varijanta koja se veže na protutijelo na Ara h 7 izotip 7.0201 od subjekta alergičnog na kikiriki, pri čemu je polipeptid rekombinantni, imobilizirani pročišćeni ili fuzijski protein, poželjno pročišćeni i rekombinantni i imobilizirani, naznačena time što je za proizvodnju kompleta za dijagnozu alergije na kikiriki.
13. Uporaba prema bilo kojem od patentnih zahtjeva 11 do 12, naznačena time što je polipeptid koji sadrži SEQ ID NO 8, poželjnije sekvencu iz skupine koja sadrži SEQ ID NO 6, SEQ ID NO 7, SEQ ID NO 8, SEQ ID NO 9 i SEQ ID NO 10, poželjnije SEQ ID NO 6, najpoželjnije Ara h 7 izotip 7.0201 ili njegova varijanta koja se veže na protutijelo na Ara h 7 izotip 7.0201 od subjekta alergičnog na kikiriki u pripravku koji sadrži navedeni polipeptid i Ara h 2 i/ili Ara h 6 ili njegovu varijantu koja se veže na protutijelo na Ara h 2 ili Ara h 6 od subjekta alergičnog na kikiriki, redom.
14. Protutijelo, poželjno protutijelo razreda IgE, IgG1 i IgG4, koje se specifično veže za SEQ ID NO 6, poželjnije na sekvencu iz skupine koja sadrži SEQ ID NO 7, SEQ ID NO 8, SEQ ID NO 9 i SEQ ID NO 10, najpoželjnije na SEQ ID NO 8, pri čemu je poželjno protutijelo izolirano.
15. Uporaba protutijela prema patentnom zahtjevu 14 naznačena time što je za dijagnozu alergije na kikiriki ili za proizvodnju kompleta za dijagnozu alergije na kikiriki.
