Claims (14)
1. Rekombinantni mikroorganizam koji proizvodi hondroitin, definiran kao nerazgranati glikozaminoglikan, kojega čine izmjenjujući ostaci d-glukuronske kiseline (GlcUA) i N-acetil-d-galaktozamina (GalNAc), vezani vezama β1-3 (GlcUA → GalNAc) i β1-4 (GalNAc → GlcUA), naznačen time što je navedeni mikroorganizam izveden iz mikroorganizma koji pripada skupini s antigenom K, gdje je u navedenom mikroorganizmu izvorno prisutan gen kfoE, koji kodira enzim odgovoran za dodavanje fruktoznih ostataka na nerazgranatu okosnica hondroitina, inaktiviran potpunom ili djelomičnom delecijom ili supstitucijom navedenog gena ili disrupcijom insercijom dodatnog nukleotidnog slijeda, te gdje navedeni gen kfoE kodira protein koji se bira iz skupine koju čine:
(A) protein koji ima aminokiselinski slijed SEQ ID NO:2;
(B) protein koji ima aminokiselinski slijed SEQ ID NO:2 modificiran delecijom, supstitucijom ili insercijom jedne ili više aminokiselina, te ima aktivnost fruktozil-transferaze; i
(C) protein koji ima aminokiselinski slijed homologan najmanje 50% aminokiselinskom slijedu u SEQ ID NO:2 i ima aktivnost fruktozil-transferaze.
2. Rekombinantni mikroorganizam u skladu s patentnim zahtjevom 1, naznačen time što inaktiviran gen kfoE je a DNA koji se bira iz skupine koju čine:
(a) DNA koja ima nukleotidni slijed SEQ ID NO:1;
(b) DNA koja hibridizira s DNA koja ima nukleotidni slijed komplementaran onom u SEQ ID NO:1 i kodira protein s aktivnošću fruktozil-transferaze; i
(c) DNA koja ima nukleotidni slijed homologan najmanje 50% nukleotidu u SEQ ID NO:1 i kodira protein s aktivnošću fruktozil-transferaze.
3. Rekombinantni mikroorganizam u skladu s bilo kojim od patentnih zahtjeva 1 i 2, naznačen time što je gen kfoE inaktiviran njegovom potpunom ili djelomičnom supstitucijom kasetom za otpornost na kanamicin i njenim naknadni uklanjanjem, što rezultira potpunom ili djelomičnom delecijom gena kfoE.
4. Rekombinantni mikroorganizam u skladu s bilo kojim od patentnih zahtjeva 1 do 3, naznačen time što je izveden iz bakterije koja pripada rodu koji se bira između Escherichia, Haemophilus, Campylobacter, Gloeocapsa i Vibrio.
5. Rekombinantni mikroorganizam u skladu s patentnim zahtjevom 4, naznačen time što je izveden iz bakterije koja pripada vrsti Escherichia coli.
6. Rekombinantni mikroorganizam u skladu s patentnim zahtjevom 5, naznačen time što je izveden iz serotipa vrste Escherichia coli koji pripada skupini 2 antigena K.
7. Rekombinantni mikroorganizam u skladu s patentnim zahtjevom 6, naznačen time što je izveden iz soja Escherichia coli 05:K4:H4U1-41.
8. Rekombinantni mikroorganizam u skladu s bilo kojim od patentnih zahtjeva 1 do 7, naznačen time što je Escherichia coli DSM23578 ili Escherichia coli DSM23644.
9. Rekombinantni mikroorganizam u skladu s patentnim zahtjevom 8, naznačen time što je Escherichia coli DSM23644.
10. Postupak biotehnološke proizvodnje hondroitina, naznačen time što se upotrebljava rekombinantni mikroorganizam odabran između sojeva Escherichia coli DSM23578 i Escherichia coli DSM23644.
11. Postupak u skladu s patentnim zahtjevom 10, naznačen time što je rekombinantni mikroorganizam Escherichia coli DSM23644.
12. Postupak biotehnološke proizvodnje hondroitina, naznačen time što se sastoji u sljedećim koracima:
a. uzgoja u pogodnom mediju rekombinantnog mikroorganizma u skladu s bilo kojim od patentnih zahtjeva 1 do 9;
b. prikupljanja i pročišćavanja hondroitina prisutnog u mikrobiološkoj kulturi.
13. Postupak biotehnološke proizvodnje hondroitina u skladu s patentnim zahtjevom 12, naznačen time što je rekombinantni mikroorganizam Escherichia coli DSM23578 ili Escherichia coli DSM23644.
14. Postupak u skladu s patentnim zahtjevom 13, naznačen time što je rekombinantni mikroorganizam Escherichia coli DSM23644.
