Claims (14)
1. Komplet za izotermičko pojačavanje ciljane molekule dvolančane nukleinske kiseline sadrži:
(a) uzvodna početnica i nizvodna početnica, svaka sadrži molekulu jednolančane DNK gdje ima manje od 30 nukleotida, čiji je najmanje jedan dio komplementaran sekvenci ciljane molekule;
(b) oligonukleotid koji uključuje molekulu jednolančane DNK gdje ima manje od 30 mukleotida, pri čemu je najmanje jedan dio tog olikognukleotida komplementaran dijelu ciljne sekvence koja intervenira kod uzvodnih i nizvodnih početnica i pri čemu taj oligonukleotid ima 3' terminus koji nije moguće produžiti;
(c) rekombinaza; pri čemu uzvodna i nizvodna početnica ne pojačavaju ciljnu molekulu neovisno od stanja oligonukleotida, kada se oligonukleotid dovodi u kontakt s rekombinazom, on napada ciljanu molekulu dvolančane nukleinske kiseline, pri tome daje komplementarnu regiju jednolančanu ciljanu molekulu i susjedne regije čime se omogućava početnicama pristup za vezivanje komplementarne jednolančane DNK na ciljanu molekulu.
2. Komplet iz patentnog zahtjeva 1 pri čemu i uzvodna i nizvodna početnica uključuje molekulu jednolančane DNK gdje ima manje od 25 nukleotida.
3. Komplet iz bilo kojeg patentnog zahtjeva 1 pri čemu i uzvodna i nizvodna početnica uključuje molekulu jednolančane DNK gdje ima 15 do 23 nukleotida.
4. Postupak u skladu s bilo kojim od prethodnog patentnog zahtjeva pri čemu rekombinaza uključuje T4 rekombinazu UvsX.
5. Komplet iz patentnog zahtjeva 4 pri čemu su proteini koji prate rekombinazu obuhvaćeni, kao što je gp32 jednolančani vezni protein i/ili UvsY agens učitavanja rekombinaze.
6. Komplet iz bilo kojeg prethodnog patentnog zahtjeva pri čemu oligonukleotid olakšava odvajanje ciljnog dupleks šablona ili proizvoda produžavanja početnice na ciljnu nukleinsku kiselinu.
7. Komplet iz patentnog zahtjeva 6 još sadrži jedan ili više dodatnih oligonukleotida radi olakšanog odvajanje ciljnog dupleksa putem tog intervenirajućeg oligonukleotida.
8. Komplet iz patentnog zahtjeva 7 pri čemu se dodatni oligonukleotid veže na lanac koji je ispustio oligonukleotid i grana se pomjera u proksimalnu dupleks nukleinsku kiselinu.
9. Komplet iz bilo kog prethodnog patentnog zahtjeva pri čemu taj oligonukleotid još obuhvaća nizvodni element na svom 3' terminusu koji je komplementaran ciljnoj sekvenci i koji nije supstrat polimeraze.
10. Komplet iz patentnog zahtjeva 9 pri čemu nizvodna početnica uključuje sekvencu koja je komplementarna sekvenci nizvodnog elementa.
11. Komplet iz bilo kojeg prethodnog patentnog zahtjeva koji još uključuje polimerazu koja izmješta lanac.
12. Komplet iz bilo kojeg prethodnog patentnog zahtjeva pri čemu uzvodna početnica uključuje sekvencu koja se preklapa s oligonukleotidom.
13. Komplet iz bilo kojeg prethodnog patentnog zahtjeva još uključuje sustav detekcije za nadgledanje pojačavanja ciljnane molekule merenjem signala koji je moguće detektirati.
14. Komplet iz patentnog zahtjeva 13 pri čemu je sustav detekcije:
(a) interhelacijska boja, preporučljivo SYBR zelena, i/ili
(b) sustav sonde koji uključuje najmanje jedan fluorofor i opcijski još uključuje sredstvo za gašenje boje.
