Abstract

Penicillins of the general formula <FORM:0989882/C2/1> and non-toxic salts thereof, where R1, R2 and R3 are the same or different and each is a hydrogen atom or a nitro, chloro, bromo, hydroxy, alkanoyloxy or alkoxy group and R4 is an alkanoyloxy group containing up to 6 carbon atoms are prepared by reacting 6-aminopenicillanic acid or a neutral salt thereof with an appropriate acylating agent. Examples describe the production of a -acetoxybenzylpenicillin in DL, D and L forms, and of a -acetoxyp - chlorobenzyl -, a - acetoxy - p - aminobenzyl-, a -propionoxybenzyl-, a -dichloroacetoxybenzyl- and a -trimethylacetoxybenzyl-penicillin. a -Hydroxybenzyl penicillin may be obtained by enzymatic hydrolysis of a -acetoxybenzyl-penicillin. Propionyl mandelyl chloride and dichloroacetyl mandelyl chloride are prepared by treating mandelic acid with respectively propionyl chloride and dichloroacetyl chloride, followed by treatment of the products with thionyl chloride. Acetyl - p - chloromandelyl chloride may be prepared by treating p-chloromandelic acid with acetyl chloride and heating the product with thionyl chloride. Specification 880,042 is referred to. Reference has been directed by the Comptroller to Specification 925,281.
