Abstract

The invention comprises 9a -fluoro-21-substituted - 11b - hydroxy - 16a - methyl - pregna-1, 4 - dien - 3, 20 - diones having the structural formula: <FORM:0970484/C1/1> and a process for the preparation thereof by dehydrating a 21-acyl-16a -methyl-17-desoxyprednisolone to yield the corresponding 21-acyloxy-16a -methyl-pregna-1, 4, 9(11)-triene-3, 20-dione, hydrobrominating and formylating this compound and reacting the resulting 9a -bromo compound with a weak alkali to yield 9b , 11b -epoxy-21-hydroxy-16a -methyl-pregna-1, 4-diene-3, 20-dione, acylating this compound to a 21-acyloxy-9b , 11b -epoxy-16a -methyl-pregna-1, 4-diene-3, 20-dione and hydrofluorinating this compound to produce the corresponding 21-acyloxy - 9a - fluoro - 11b - hydroxy-16a -methyl-pregna-1, 4-diene-3, 20-dione. Dehydration may be effected with methane or p-toluene sulphonyl chloride, and bromination and formylation by the action of N-bromo-succinimide and perchloric acid in the presence of dimethylformamide. The 21-acyloxy substituent may be saponified. Specification 970,482 is referred to.
