Abstract
The invention comprises compounds of the formula: <FORM:0952591/C2/1> (wherein R is hydrogen or a lower alkyl group and, when R is hydrogen, A is -COOR1 or -CONR2R3, R1 being an alkyl group of 3 to 8 carbon atoms and R2 and R3 together with the nitrogen atom forming a 5- or 6-membered saturated heterocyclic ring optionally containing a further hetero atom, or, when R is lower alkyl, A is -COOH, -COOR4 or -COO(CH2)nNR2R3, R4 being an alkyl group of 1 to 8 carbon atoms and n being 2, 3 or 4, the word "lower" referring to groups containing at most 4 carbon atoms) and acid-addition and quaternary ammonium salts of the amines; their preparation (1) when R is hydrogen and A is -COOR1, by esterifying 4, 4-bis(41-hydroxyphenyl)-pentanoic acid with the alcohol R1OH in presence of an esterification catalyst; or (2) when R is hydrogen and A is -CONR2R3, by reacting an alkyl ester of 4, 4-bis (41-hydroxyphenyl)-pentanoic acid with the amine NHR2R3; or (3) when R is alkyl and A is -COOH, by etherifying 4, 4-bis (41-hydroxyphenyl)-pentanoic acid with a dialkyl sulphate and separating the desired product from the reaction mixture by alkaline extraction; or (4) when R is alkyl and A is -COOR4, R and R4 being the same, by the method of (3), the required ether-esters being separated from the main product i.e. ether-acids, by alkaline extraction of the latter; or (5) when R is alkyl and A is -COOR4, R and R4 being different, by direct esterification of the products of (3) with an excess of the alcohol R4OH in presence of an acid catalyst; or (6) when R is alkyl and A is -COO (CH2)nNR2R3, by reacting an alkali metal salt of the products of (3) with the appropriate alkylamino halide. The invention also comprises the preparation of 4, 4-bis (41-hydroxyphenyl)-pentamide by reacting a corresponding alkyl ester with aqueous ammonia at room temperature. Detailed examples are provided. The above derivatives of 4, 4-bis (41-hydroxyphenyl)-pentanoic acid, which are stated to be of value as therapeutics, may be made up into pharmaceutical compositions with suitable carriers.
