Abstract
The invention comprises 21-(211-diethylaminoethoxy - ethyl 3-amino -4- n-butoxybenzoate monohydrochloride, and the preparation thereof either by reacting 3-nitro -4- n-butoxy-benzoic acid chloride with 2-(21-diethylaminoethoxy)-ethanol, forming the hydrochloride of the resulting 21-(211-diethylaminoethoxy) - ethyl 3-nitro -4-n-butoxy-benzoate and reducing this, or by reacting methly or ethyl 3-nitro -4-n-butoxybenzoate with the above mentioned alcohol in the presence of a metal alcoholate in solution in an inert solvent to obtain the nitro ester and then forming the hydrochloride and reducing this as before. The above processes may be combined with the preparation of the starting materials, the acid chloride being prepared from 3-nitro -4-n-butoxy-benzoic acid, itself in turn prepared by saponification of methyl, ethyl or butyl 3-nitro -4-n-butoxy-benzoate, themselves prepared by nitration of the corresponding alkyl p-(n-butoxy)-benzoates which in turn are prepared by reacting the corresponding alkyl p-hydroxybenzoates with an n-butyl halide in the presence of a neutralizing agent or the alkali metal salts of these phenols with an n-butyl halide in butanol. Examples are given. The amino-alkoxy ester of the invention, which is stated to be a local anaesthetic, may be made up into pharmaceutical compositions with a suitable carrier. These may take the form of an injection solution in ampoules.
