Abstract

A method of making a collagen sponge, having interconnected pores, comprises freezing an aqueous dispersion of collagen fibres, removing the water by immersing the frozen mass in a water-miscible, dehydrating solvent, and drying the resulting collagen mass. The dehydrating bath contains sufficient quantity of a base, such as ammonium hydroxide, to neutralize the acid present in the collagen dispersion. In one example, cattle tendon is cleaned and sliced and then treated with an enzyme solution to dissolve elastin. It is then swollen by the addition of cyano-acetic acid and forced through a series of screens. After being frozen in steel moulds the blocks are removed and immersed in a series of baths of isopropanol containing a small proportion of ammonium hydroxide, to dehydrate the mass, which is finally air dried. Acetic acid, cyanoacetic acid and lactic acid are referred to as suitable for use in producing the collagen dispersion and either acetone or isopropanol may be used for dehydration.
