506,095. Vaccines and the like. CHEMICAL WORKS. April 26, 1938, No. 12467. Convention date, May 8, 1937. [Class 81 (i)] Material for inoculation purposes is prepared by freeing cultures of bacteria &c. from water by exposure to a high vacuum at temperatures below 0‹ C. and killing the bacteria &c. in the dry state by treatment with anhydrous cell poisons or radiations of short wave length. The cultures are frozen rapidly by immersion in a mixture of carbon dioxide and ether, or liquid air, and dried in a vacuum of less than 0À001 mm. of mercury. The dehydrated organisms are then killed by treatment with anhydrous cell poisons such as hydrocyanic acid, hydrogen sulphide, carbon disulphide or formaldehyde, or by ultra violet light, the process being carried out preferably in a solvent, for example benzene, xylene, halogenated hydrocarbons or ether. The product after being freed from solvent may be taken up in physiological salt solution or in a non-aqueous oil or fat, e.g. paraffin oil, olive oil, wool fat. Example 1. Staphylococci after freezing and drying are killed by a solution of hydrocyanic acid in benzene. The dry product is suspended in a mixture of anhydrous wool fat and olive oil. Example 2. Streptococci, after separation from a culture, freezing and drying, are killed by means of a solution of formaldehyde in benzene. The product is dispersed in physiological salt. solution containing a trace of pyridine. Example 3. Dehydrated tubercle bacilli are killed by extraction with carbon disulphide. Example 4. A culture of tubercle bacilli obtained by culture in a collodion sac inserted in the abdonimal cavity of a guinea pig is treated according to the method of example 2. Example 5. The brain of a fowl infected with fowl plague virus is rubbed to a paste and treated according to examples 1-3.
