Abstract

Streptomycin is produced in enhanced yields by fermenting a nutrient medium at pH5 to 8 with a streptomycin-producing micro-organism, e.g. Streptomyces griseus in the presence of an alkali metal salt of p-(2-hydroxy-1-naphthylazo) benzene sulphonic acid, an alkali metal salt of diphenyldiazo-bis-a -naphthylaminesulphonic acid or an alkali metal salt of p-dimethyl aminophenylazobenzene sulphonic acid. The medium contains (a) carbohydrate such as sucrose, dextrose or starch (b) nitrogen source such as ammonium salts, corn steep liquor, casein, soybean meal and peptones (c) mineral salts such as potassium dihydrogen phosphate, zinc sulphate, magensium sulphate and calcium carbonate. The pH is preferably 6 to 7,5, the temperature of cultivation 24 to 30 DEG C. and the duration 1 to 5 days. Cultivation may be surface or submerged, which is preferred. The dyestuff is added before or after inoculation with the Streptomyces griseus, in which case it is added within 24 hours of initiation of the fermentation. It is used in quantities of 0,20 to 2,5 g. per litre of the medium. Alkali metal dye salts such as potassium and lithium may be used but the sodium salt is preferred.
