Abstract

Tetracycline (omegamycin) is produced by cultivating Streptomyces viridifaciens ATCC-11989, or a mutant thereof, in an aqueous nitrogenous nutrient medium containing carbohydrate solution under submerged aerobic conditions and recovering the antibiotic therefrom. The initial pH of the medium is 6.0-6.2, the temperature of cultivation 24 DEG to 30 DEG C., and preferably 26 DEG to 28 DEG C., and duration of cultivation 2 to 3 days. The medium contains a source of (a) carbon, e.g. starch, sucrose, glucose, maltose, dextrose, glycerol, galactose, scylose, arabinose, rhamnose, fructose, lactose, insulin and dextrins; (b) nitrogen, e.g. amino acids, casein, fish meal, soybean meal, meat extracts, liver cake, urea, nitrates, ammonium compounds, distillers' solubles, corn steep liquor, wheat steep liquor, whey, corn or wheat gluten, peptone, offals, brewers' yeast, cottonseed meal, lactalbumin and tryptone; (c) growth substances; and (d) minerals, e.g. sodium chloride, potassium phosphate, magnesium sulphate, sodium nitrate and calcium carbonate. Tetracycline is recovered from the clarified broth by (a) solvent extraction with a water-immiscible solvent such as butanol or methyl isobutyl ketone; or (b) adsorption on e.g. silica. The extract from (a) is washed, concentrated by distillation and the antibiotic precipitated by the addition of petroleum ether. The antibiotic in the form of base or acid salt depending upon pH at which extraction is made, may be further purified by treating with ammonium hydroxide and adsorption on silica. The antibiotic is eluted from the silica by means of dilute acid. Salts with acids are formed by adding an organic or inorganic acid to a suspension of the antibiotic in water. Specified are the hydrochloride, sulphate, phosphate, citrate, tartrate and gluconate. The alkali metal salts are prepared by treating an aqueous suspension of the antibiotic with an alkali metal hydroxide. Rb values are given for the following solvent systems: (a) water; (b) aqueous sodium citrate; (c) butanol saturated with water; (d) dry methyl isobutyl ketone; (e) aqueous methanolpiperidine of pH 9.5; (f) aqueous methanolglacial acetic acid; (g) water saturated butanol, piperidine and p-toluene sulphonic acid; (h) saturated butanol, piperidine, p-toluene sulphonic acid and lauric acid; and (i) isoamyl alcohol, carbon tetrachloride and aqueous sodium citrate. Reference has been directed by the Comptroller to Specification 747,779.
