Abstract
A new antibiotic, amphomycin, is produced by cultivating Streptomyces canus, which is described, or a mutant thereof, in an aqueous, nutrient containing carbohydrate solution under submerged aerobic conditions at e.g. 24 DEG to 30 DEG C. for 2 to 7 days and pH 7.0 to 7.2. Specified carbohydrates are starch, sugars such as glucose, sucrose and lactose, glycerol, insulin and dextrin. The medium also contains (a) nitrogen source, e.g. casein and amino acids, fish meal, soybean meal, meat extracts, corn-steeping liquor, wheat steeping liquor, whey, brewers yeast, cottonseed meal, lactalbumin, distiller's solubles, liver cake, urea, nitrates, ammonium compounds and others; (b) mineral salts, e.g. sodium chloride, potassium phosphate, magnesium sulphate, sodium nitrate, and (c) buffers. The antibiotic is recovered by adjusting the filtered or unfiltered broth to approximately pH 2.0 by, e.g. hydrochloric acid and extracting with butanol, especially n-butanol, or pentanol. The sodium salt is recovered by extracting the organic solvent extract with water containing sodium hydroxide to pH 7.3 to 7.4. Further purification involves one or more of the following steps: (a) treatment of solutions especially organic solvent solutions with activated carbon; (b) formation of Reinecke's salt; (c) precipitation by means of a non-solvent, e.g. ethyl acetate; (d) precipitation at the isoelectric point namely pH 3.3 to 3.6. Amphomycin is effective against B. mycoides, B. aureus, M. tetragenus, Staph. aureus and B. subtilis; it is amphoteric forming salts with acids and metals; it is soluble in water except over the range pH 3.0 to 4.0; soluble in methanol in both acid and salt form, soluble in alcohols in acid form only; is extractable from water of pH 2.0 by butanol or pentanol but not by methyl isobutyl ketone, benzene, ether, ethyl acetate or amyl acetate; forms ammonium and Reinecke's salt; absorbs ultra-violet light only in the 210-230 m region; exhibits negative response to ninhydrin, Sakaguchi, Molisch and Ehrlich-Pauly tests; is stable for at least 10 days in aqueous solutions of pH 2 to 10; it contains carbon, hydrogen, oxygen and nitrogen.
