Abstract
A new antibiotic anisomycin (also known by the Trade name "Flagecidin") is produced by cultivating Streptomyces griseolus ATCC 11796, or Streptomyces roseochromogenus isolate No. 14576-4, or mutants thereof, in an aqueous nutrient medium under submerged aerobic conditions. A temperature of 24 DEG to 30 DEG C. and a period of 1 to 4 days is suitable. The medium contains (a) a source of carbohydrate, <PICT:0768364/IV(b)/1> <PICT:0768364/IV(b)/2> e.g. sugars, starch, glycerol and corn meal; (b) a source of nitrogen, e.g. casein, soybean meal, peanut meal, wheat glutein, cottonseed meal, lactalbumin, enzymatic digest of casein and tryptone; (c) a source of growth substances, e.g. distillers' solubles, yeast extract and molasses fermentation residues; (d) mineral salts such as sodium chloride, potassium phosphate, sodium nitrate, magnesium sulphate; and (e) trace metals such as copper, zinc and iron. A buffer, e.g. calcium carbonate and antifoaming agent may be present. Anisomycin is isolated from the liquid broth by (A) solvent extraction, or (B) adsorption. Solvent extraction is at pH6 to 10 by means of waterimmiscible solvents such as benzene, toluene, ethyl acetate, butyl acetate, methyl isobutyl ketone butanol and chloroform. Purification is obtained by treating the extract with water of below pH2.5, re-extracting the solution with the organic solvent at pH6 to 10 and precipitating by the addition of a non-solvent such as cyclohexane, hexane and carbon tetrachloride. It may be recrystallized from butanol, ethyl acetate, benzene or ethylene dichloride. Adsorption is an (a) charcoal, (b) ion-exchange resins, or (c) alumina. Anisomycin is a white basic substance effective against pathogenic protozoa and fungi; soluble in water, methyl alcohol, ethyl alcohol, dioxane and chloroform; insoluble in hexane, cyclohexane and carbontetrachloride; forms salts with acids such as hydrochloric, sulphuric and phosphoric; base has melting point of 140-141 DEG C. and [a ]D25= -30 DEG (in methanol) [a ]D25 = - 45 DEG \sB3 DEG ; exhibits three peaks in the ultra-violet at 224, 277 and 283.5mm ; dissolved in chloroform shows peaks in the infra-red at 3545, 3450, 3320, 2890, 2800, 1725, 1610, 1582, 1515, 1470, 1447, 1380, 1320, 1302, 1242, 1178, 1036 and 962; contains 63.5 per cent carbon, 7.2 per cent hydrogen, 5.2 per cent nitrogen and 24.1 per cent oxygen.
