Abstract
A process for preparing adrenocorticotrophin (ACTH) from a mammalian blood substance such as whole blood, plasma, serum, defibrinated blood and especially beef plasma or serum comprises treating it with an ACTH-activating agent such as pyridoxine preferably by incubating at a temperature from 15 DEG to 150 DEG C., the preferred range being 90 DEG to 100 DEG C., at a pH of 3.5 to 5.5, preferably 4.7 or 4.8, the pH being obtained by agents such as hydrochloric acid, sodium hydroxide, acetic acid-sodium acetate buffer. An organic reducing agent containing at least one sulphydryl group such as cysteine, beta-mercapto-ethanol, thioglycollic acid may be added to stabilize the ACTH. The ACTH activity may be generated in the blood substance by the treatment with the agent and the ACTH then extracted in the usual ways or a pro-ACTH substance may be extracted which is then treated with the agent.
