Abstract

A crystalline insulin preparation having a prolonged therapeutic effect comprises insulin, a zinc salt, an alkaline protein or protein split product, and a phenol or phenol derivative mixed together in aqueous medium in approximately isophanous conditions (i.e. such that after the removal of the insulin complex from preparations of the above type, the addition of insulin to one portion of the residual liquor and of alkaline protein to the other, produces the same degree of opalescence in both portions) at a pH of 6-8, preferably in the presence of a buffer such as sodium phosphate or acetate. Crystallization is improved in the presence of inorganic salts such as sodium chloride, which may be added to produce an isotonic preparation. Alternatively, an excess of such salt may be used and isotonicity produced by the addition of e.g. glucose. Specified proteins are salmiridine, clupeine, scombrine, sturine, thynnine, cyprinine, cyclopterine, a split product of salmiridine and thymus histone. Suitable phenols are o-, m-, p-cresol; m-, p-, chlorophenol; m-, p-nitrophenol and resorcin.
