Abstract

The invention comprises retrocortin esters of the formula <FORM:0826360/IV (b)/1> (wherein R is a carboxyacyl radical), and alkali and alkaline earth metal salts thereof, together with a process for their preparation by reacting retrocortin with an acylating agent derived from a polybasic carboxylic acid, e.g. an acid anhydride or halogenide in the presence of a tertiary amine and conveniently in an organic solvent, e.g. pyridine, benzene or chloroform and, if desired, treating the resulting ester with an aqueous solution (preferably containing a lower aliphatic alcohol) of a basic alkali or alkaline earth metal compound, e.g. a carbonate, bicarbonate, or hydroxide, to form a salt. Specified polybasic acids are succinic, glutaric, maleic, adipic and phthalic, and the reaction is preferably effected in pyridine, benzene or chloroform as solvent. In examples: (1) retrocortin is stirred at room temperature in pyridine containing succinic anhydride, the mixture added dropwise to excess dilute HC1 when retrocortin 21-hemi-succinate precipitates; subsequent treatment with sodium carbonate in aqueous ethanol gives the corresponding sodium salt (2), as in (1) using maleic anhydride to yield the 21-hemi-maleate, and (3), as in (1) using phthalic anhydride to give the 21-hemiphthalate.
