Abstract

A process for the manufacture of purified ACTH preparations is characterized in that ACTH is adsorbed on unmodified carboxy methyl cellulose in an aqueous sytem at a pH below 8 after which the ACTH is eluted at a pH which differs from that of adsorption, in which use is made of CMC which is insoluble at the pH values at which the adsorption and elution occur. The adsorption may take place by stirring ACTH solution with CMC and eluting after filtration or centrifugation or may be carried out in a column filled with CMC and then eluting from the column. The crude ACTH solution is preferably acidified with an organic acid such as acetic, propionic or lactic acid and adsorption preferably conducted at a pH from 2.5 to 4.5. Elution is conducted by acids such as hydrochloric, hydrobromic, sulphuric or phosphoric acid, preferably at a pH below 2, using for preference 0.1 N hydrochloric acid.
