Abstract

Thyrotropin is selectively precipitated from an aqueous solution of thyrotropin and non-thyroid stimulating proteins from pituitary material by the addition of a mono-hydroxy alkane containing less than 4 carbon atoms until the concentration of the said mono-hydroxy alkane is from 5 to 35 per cent by volume. Ethanol is the preferred precipitating agent. The fractionation is carried out at low temperatures preferably between -5 DEG C. and +5 DEG C. The process is particularly adapted for separating thyrotropin from gonadotropin. The thyrotropin precipitate is purified by a process which involves forming a solution at pH 4, removing precipitates formed on the addition of acetone, until finally the thyrotropin is precipitated at an 80 per cent acetone concentration. A detailed procedure of extraction and purification is described in the examples.
