Abstract
Xanthine derivatives of the general formula <FORM:0759981/IV(a)/1> (wherein R1 and R2 are alkyl groups each containing from 1 to 5 carbon atoms and R3 is an acetonyl or propargyl group) are prepared by reacting the appropriate 1,3-dialkyl-xanthine unsubstituted in the 7-position in the presence of a tertiary base (preferably triethylamine) with a compound R3-halogen or, when R3 is an acetonyl group, a ketal thereof. When R3 is the propargyl group this may be subsequently transformed into the acetonyl group by the addition of 1 molecule of water to the triple bond. Examples illustrate the use of theophylline and, respectively, chloroacetone and propargyl bromide in the process of the invention, which is performed by refluxing the reactants with triethylamine in ethanol. 7-Acetonyltheophylline is also prepared from 7-propargyltheophylline by refluxing the latter with sulphuric acid and mercury oxide.
