Abstract

266,341. Lilly & Co., E., (Assignees of Clowes, G. H. A., Walden, G. B., and Scott, J. P.). Feb. 16, 1926, [Convention date]. Void [Published under Sect. 91 of the Acts]. Antitoxines. - Anti-pneumococcic serum of anti-toxic character obtained by known processes is purified by fractional precipitation, preferably fractional salting.out by means of sodium chloride. According to an example the serum is diluted with water, and salt is added to give a concentration of 18 to 20 gms. per 100 cc. A precipitate is formed which contains the antitoxin together with more easily precipitated impurities ; this is redisssolved in water at about pn 8.0 and again precipitated with salt at a concentration of 15 to 20 gms. per 100 cc. The second precipitate is redissolved and treated with salt at a concentration of 7.5 to 15.0 gms. per 100 c.c. to remove the readily precipitated impurities and the solution remaining is treated with a more concentrated salt solution to cause the formation of a final precipitate containing the anti-toxin. The final precipitate is separated and extracted with weakly alkaline water to give a solution, which may be used as such. or may be further purified by repetition of the fractional salting-out process. The precipitate obtained at the salt concentration of 7.5 to 15 gms. per 100 c.c. may be re-dissolved and further treated to recover any anti-toxin contained therein.
