1468494 Reference standard for enzyme determinations. WARNER-LAMBERT CO 15 Sept 1975 [4 Oct 1974] 37747/75 Heading G1B A coloured reference standard for use in diagnostic determinations involving enzymatic reactions in which 1-(p-iodophenyl)-5-(p-nitrophenyl)-3- phenylformazan is produced comprises an aqueous solution of: A. from 0À001 to 0-020 wt. per cent of 1-(piodophenyl)-5-(p-nitrophenyl)-3-phenylformazan; B. from 0À01 to 0À10 wt. per cent of serum albumin; C. from 2 to 10 wt. per cent of N,N'-dimethylformamide or dimethyl sulphoxide; and D. from 2 to 10 wt. per cent of isopropanol; (the percentages being based on the weight of the total solution) and said aqueous colour standard solution having an absorbence maximum at 500 manometers; An inert bulking agent e.g. dextran may be present in an amount of 5-20 wt. per cent and the reference standard then lyophilized to an form which is stable for at least six months at 4 and 24‹C. The reference may be used in the determination of such enzyme activities as serum bactate dehydrogenase, creative phosphokinase and glucose- 6-phosphate dehydrogenase.
