Abstract
1459705 Biuret analysis COMMONWEALTH SCIENTIFIC & INDUSTRIAL RESEARCH. ORGANISATION 1 April 1975 [2 April 1974] 13365/75 Heading G1B A single-solution reagent for the colorimetric determination of protein comprises an aqueous biuret reagent (i.e. a reagent comprising potassium sodium tartrate, potassium hydroxide and a source of cupric ions which on contact with protein will give a violet coloration) and an aliphatic alcohol which is miscible with water. The alcohol is preferably isopropanol. Protein in a solution sample may be determined by heating said sample with the single-solution reagent, allowing colour to develop, separatingundissolved matter from the sample and determining the protein concentration by colorimetric measure- ment. A suitable apparatus for carrying out the determination comprises a heated and stirred reaction vessel, means for dispensing desired volumes of reagent and protein containing material to said reaction vessel, means for dispensing a predetermined volume of sample/reagent mixture from said reaction vessel to means adapted to clarify said mixture and dispense a clarified sample therefrom, wherein the passage of the mixture through the apparatus is controlled automatically. Preferably the apparatus includes means for dispensing reagent to said reaction vessel in an amount determined proportionately to the amount of proteinaceous sample. A suitable form of dispensing means is shown in Fig. 2 and comprises a beam 11 supported on a fulcrum 12, a sample holder 13 and reagent weighing vessel 15 being attached thereto. In operation, valve 19 opens, thus admitting reagent into vessel 15, the beam being consequently depressed until flag 17 interrupts light entering sensor 18, when valve 19 accordingly shuts and valve 20 opens, allowing reagent to syphon out of vessel 15 until flag 17 clears the light ray. The beam lock is engaged and the sample holder is then filled with sample. The lock is then disengaged and the beam falls until flag 17 interrupts the light ray, at which point valve 21- opens and reagent syphons into the reaction vessel until, as the beam rises, the flag clears the light ray and the valve 21 closes. Thus the amount of reagent dispensed to the reaction vessel will be in the same fixed proportion relative to the sample amount.
