Abstract

1,219,624. Determination of creatinine. E. MERCK A.G. 10 June, 1968 [20 July, 1967], No. 27507/68. Heading B1X. A colour reagent for the determination of creatinine by measurement of the light absorption of creatinine picrate comprises lithium picrate and lithium hydroxide. The reagent may be made by mixing an 8% to 12% aqueous solution of lithium hydroxide with an aqueous solution of picric acid, preferably saturated (i.e. about 1.2%), optionally itself containing lithium hydroxide, in volume ratio 1:3 to 1:6. The solution may be lyophilised in single - test amounts. Alternatively the reagent may be formed by mixing solid lithium hydroxide and solid picric acid, and preferably also a water-soluble inert salt as diluent. In the determination of creatinine, the reagent is added to the sample, which may be initially deproteinized with trichloroacetic acid or perchloric acid, and the creatinine picrate formed is determined spectrophotometrically.
