Abstract

A composition for the determination of a -hydroxybutyrate dehydrogenase, e.g. in body liquids of animals and man, comprises nicotinamide adenine dinucleotide, 2:6-dichloroindophenol, phenazine methosulphate, and a -hydroxybutyrate and a pH 6.8-8.5 phosphate buffer. In the presence of a -hydroxybutyrate dehydrogenase, the a -hydroxy butyrate is oxidised to a -ketobutyrate and the dinucleotide is reduced: the dyestuff system (phenazine methosulphate 2:6-dichloroindophenol) is then in turn reduced to give the leuco form of the second component. The materials may be separately packed in the form of the following aqueous solutions, concentrations of some of the components being specified:-(1) phosphate buffer, dinucleotide, 2:6-dichloroindophenol, a -hydroxybutyrate, (2) phosphate buffer, dinucleotide, 2:6-dichloroindophenol and (3) phenazine methosulphate. Solutions (1) and (3) provide the reagent solution, solutions (2) and (3) provide a blank. Alternatively the composition may be impregnated in a suitable carrier e.g. paper.
