Abstract
A composition for the determination of isocitrate dehydrogenase, e.g. in body fluids of animals and man, comprises sodium isocitrate, nicotinamide adenine dinucleotide phosphate, a source of Mg\sW\sW ions, 2:6-dichloroindophenol, phenazine, methosulphate and a triethanolamine buffer of pH 6.8-8.5. In the presence of the dinucleotide and Mg\sW\sW, isocitrate dehydrogenase catalyses the conversion of isocitrate to a -ketoglutarate and CO2: the dye-stuff system (2:6-dichloroindophenol and phenazine methosulphate) is then reduced by the resultant reduced form of the dinucleotide. The materials may be put up as a test pack in the form of the following aqueous solutions: (1) triethanolamine buffer, nicotinamide adenine dinucleotide phosphate, dichloroindophenol, MgCl2, trisodium isocitrate, (2) as (1) but omitting the citrate and (3) phenazine methosulphate. Solution (2) is used with (3) as a control. Alternatively the composition may be impregnated in a suitable carrier e.g. filter paper strips, and the product lyophilised.
